The E2-IL28B fusion protein prepared by CHO-S cells was mixed with ISA 201 VG adjuvant to prepare a classical swine fever subunit vaccine. This solved the safety risks of CSF attenuated vaccines and the problem of insufficient immunization efficacy of E2 subunit vaccines, and achieved a stronger immune response and antibody production.

Abstract

本发明公开了一种猪瘟病毒E2‑IL28B融合蛋白及其作为抗原在制备猪瘟亚单位疫苗中的应用。猪瘟病毒E2‑IL28B融合蛋白的核苷酸序列如SEQ ID NO.1所示。本发明将E2蛋白与免疫增强剂IL‑28B蛋白融合,利用哺乳动物CHO‑S细胞表达E2‑IL28B融合蛋白,免疫小鼠后发现,E2‑IL28B融合蛋白的体液免疫和细胞免疫效果均优于E2蛋白,表明E2‑IL28B融合蛋白更适合作为CSF亚单位疫苗的有效抗原。本发明将为CSF新型疫苗的研发提供技术支撑。
