The application provides application of aromatic retinoic acid in improving embryo induction rate of barley and rice in vitro microspore, and belongs to the technical field of plant cell engineering. In the application, the proportion of swollen and dense microspores is significantly increased after the barley free microspores are pretreated by 0.15-0.25 muM aromatic retinoic acid, which indicates that the pretreatment of aromatic retinoic acid has a great promoting effect on the induction and start of embryo development of the barley free microspores. The yield of callus obtained by adding 0.2-0.4 muM aromatic retinoic acid in the induction medium of the rice free microspore culture is also significantly higher than that of the control, which indicates that the addition of aromatic retinoic acid in the rice free microspore culture process can improve the embryo induction efficiency. The method for improving the embryo induction rate of the barley / rice in vitro microspore is low in cost, simple and easy to operate, safe and non-toxic, and lays a foundation for the breeding of barley or rice.
