The invention discloses a resistance detection device with automatic calibration function and a detection method, which relate to the technical field of resistance detection, and comprise a workbench, wherein a base and a fixed mold are arranged above the workbench, a hydraulic cylinder is arranged below the base, and a telescopic cylinder is arranged above the base to fix a circuit board, two brackets are arranged at the upper end of the workbench, two fixing rods are arranged at the upper end of the bracket, the two fixing rods are respectively connected with a mobile detection unit through a crossbeam, a detection probe is arranged below the mobile detection unit to detect the resistance, a visual positioning part and a laser ranging induction part are arranged on the fixed mold, a laser ranging sensor measures the distance of the laser ranging induction part, so as to judge whether the fixed mold is horizontal, and leveling is performed through the hydraulic cylinder, a CCD camera takes a picture of the visual positioning part, the device identifies the current position, and controls a motor to adjust the position of the mobile detection unit to a predetermined origin, and performs calibration, so as to avoid the accuracy reduction caused by error accumulation after long-term operation.
