Abstract
Description
Technical Field
[0001] This application claims priority based on Korean Patent Application No. 10-2022-0035446 filed on March 22, 2022 and Korean Patent Application No. 10-2022-0082719 filed on July 5, 2022, and all contents disclosed in the specifications and drawings of those applications are incorporated herein by reference.
[0002] The present disclosure relates to a composition for improving scalp conditions. Background Art
[0003] In modern society, most people use a wide variety of products to clean their scalp and hair and make them clean. However, the main ingredients of these products include surfactants, bubble enhancers, emulsifiers, thickeners and preservatives, which may induce scalp irritation and itching.
[0004] In addition, many people use hair dyes or hair perms to change their hairstyle for beauty purposes. For some sensitive scalps, physical and chemical stimulation caused by hair dyes or hair perms may damage the scalp.
[0005] In addition, changes in temperature or humidity in daily life, exposure to sunlight or pollutants such as dust, etc. may also irritate the scalp. In addition, stress or nutritional deficiency may also worsen the scalp condition.
[0006] The scalp is composed of the epidermis, dermis, and subcutaneous tissue. Normally, the stratum corneum of the epidermis of the scalp is replaced by a new stratum corneum at a cycle of about one month. For a healthy scalp, when the stratum corneum is replaced, small-sized keratin falls off and is replaced by new keratinocytes. However, when the replacement deviates from the normal state and causes an excess of keratinocytes, there are cases where lumps gather to a degree visible to the naked eye and fall off, which is called dandruff. Dandruff is a problem of the scalp, but it does not cause much problem to human health. Although there are differences in degree, it is a phenomenon that occurs on the scalp of almost all people.
[0007] The causes of dandruff known so far include increased sebum secretion of the scalp, increased proliferation and activity of microorganisms, abnormal keratinization due to abnormal scalp metabolism, and scalp irritation caused by fatty acids produced by lipase.
[0008] Furthermore, if there is a lot of dandruff, it may cause physical stimulation such as scratching or rubbing the scalp due to itching, which may cause wounds or inflammation. The pruritus that occurs at this time is also induced by histamine and arachidonic acid metabolites, which causes a complex inflammatory reaction and stimulates nerve cells, thereby inducing itching.
[0009] In addition, regarding the symptoms of dandruff, the scalp's sebum secretion and sweat, as well as the decomposition of keratin caused by microorganisms, induce body odor on the scalp. As these metabolites oxidize, the scalp may emit an unpleasant odor. Because the odor of the scalp is related to the distribution of microorganisms, the scalp can create an environment (hair, humidity, etc.) where microorganisms can survive more than other parts of the skin.
[0010] The most commonly used active ingredient in shampoos currently on the market for preventing dandruff is zinc pyrithione. Zinc pyrithione has antimicrobial effects, but has the characteristics of high specific gravity, water insolubility, and difficulty in maintaining a dispersed state in a dosage form. In addition, zinc pyrithione is irritating, so in Korea, research and development of products that can replace it with other ingredients are being gradually carried out.
[0011] Attempts have been made to suppress the generation of dandruff or improve scalp pruritus by applying or taking existing natural ingredients such as plant extracts to the scalp, but the disadvantage is that they are not very effective in improving dandruff, excessive keratin loss, keratin hyperplasia, pruritus, etc. Therefore, there is a need to develop a composition for improving scalp conditions that is effective in improving dandruff, excessive keratin loss, keratin hyperplasia, pruritus, etc. Summary of the invention
[0012] Technical issues

[0037] Below, in order to help understand the present invention, examples and the like are described in detail. However, the embodiments of the present invention can be deformed into various forms, and the scope of the present invention should not be interpreted as being limited to the following embodiments. The embodiments of the present invention are provided to more completely describe the present invention to those of ordinary skill in the art to which the present invention belongs.
[0038] Experimental Example 1: Confirmation of the effect of suppressing inflammation
[0039] (1) Evaluation of inflammatory response inhibition rate
[0040] In order to confirm the anti-inflammatory activity of gluconolactone and climbazole, the ability to inhibit the generation of nitric oxide (NO), one of the substances that induce inflammation, was analyzed. The RAW 264.7 cells used in the experiment are murine macrophage cell lines, which are used as the main model for anti-inflammatory activity cell experiments and are purchased from ATCC (American Type Tissue Collection). The above RAW 264.7 cells were cultured in RPMI1640 (Gibco) medium containing 10% FBS (fetal bovine serum, Gibco) and 1% penicillin / streptomycin (Gibco).
[0041] In order to confirm the anti-inflammatory activity of gluconolactone and climbazole, the above RAW 264.7 cells were cultured at 2.0× 10 4 The cells were injected and attached to a 96-well plate and cultured for 24 hours. Then, 1 μg / mL of lipopolysaccharide (LPS), gluconolactone, and climbazole were simultaneously treated for 24 hours, and the NO production inhibition ability was analyzed by Griess reaction.
[0042] The Griess reaction is a method for quantitatively analyzing the amount of NO generated from cells treated with a sample and released into a culture medium. A Griess reaction solution is prepared by mixing a 1% sulfonamide solution and a 0.1% naphthalenediamine dihydrochloride solution dissolved in a 5% phosphoric acid solution in a ratio of 1:1.
[0043] For the cell culture fluid treated with the sample for 24 hours, 50 μL was taken from each well and dispensed into a 96-well plate. After that, the prepared Griess reaction solution was dispensed into each well at 50 μL in the same amount as the cell culture fluid, and the color change caused by NO generation and inhibition was measured by absorbance at 540 nm using a multi-plate reader. The experimental results are expressed as the average of 3 replicates for each gluconolactone and climbazole treatment combination.
[0044] In order to confirm whether gluconolactone and climbazole induce cytotoxicity, CCK-8 assay was performed using 50 μL of cell culture medium. CCK-8 assay is one of the methods for measuring cell viability. A measurement reaction solution of 1 / 10 of the cell culture medium is added, reacted at 37°C for 1 hour, and then the absorbance at 450nm is measured using a multiplate reader. No cytotoxicity was observed between the measured concentrations.
[0045] The LPS-induced NO inhibitory activity was expressed as a percentage relative to a control group (A) not treated with LPS and a sample and a control group (B) to which only LPS was added without adding a sample. The percentage of the inflammation inhibitory activity in each sample (C) was calculated using the following formula.
[0046] Inflammation suppression activity (%) = {(BC) / (BA)}×100
[0047] A is the absorbance at 540 nm of the control group treated with neither LPS nor the sample, B is the absorbance at 540 nm of the control group treated with only LPS, and C is the absorbance at 540 nm of the experimental group treated with both LPS and the sample.
[0048] The values calculated for the inflammatory response inhibitory activities of gluconolactone and climbazole of the present disclosure are shown in Table 1 below.
[0049] [Table 1]
[0050]
[0051] From the experimental results described in Table 1 above, it was confirmed that gluconolactone or climbazole inhibited the inflammatory response, respectively. In particular, it was confirmed that the inflammatory response inhibition rate was significantly increased when gluconolactone and climbazole were co-treated.
[0052] (2) Study on the synergistic effect of gluconolactone and climbazole in inhibiting inflammation
[0053] When using gluconolactone and climbazole together, Colby formula is used to calculate the expected expected value (inflammatory response inhibition rate). The calculation process and its result are as follows. "Synergistic effect" refers to ["Calculation of the synergistic and antagonistic responses of herbicide combinations" Weeds, (1967), 15, 20-22 pages] defined.
[0054] Colby formula: expected value = A + B - (A × B / 100)
[0055] A: The efficacy of ingredient A observed at the same concentration as used in the mixture
[0056] B: The efficacy of ingredient B observed at the same concentration as used in the mixture
[0057] In order to calculate the expected inflammatory response inhibition rate (expected value) when gluconolactone and climbazole are used together, the inflammatory response inhibition rate of the experimental group treated with only LPS and gluconolactone was substituted into A of the above Colby formula, and the inflammatory response inhibition rate of the experimental group treated with only LPS and climbazole was substituted into B, thereby calculating the expected value according to the Colby formula, and the values are then shown in Table 2 below.
[0058] In addition, the degree of synergistic effect was calculated and shown together in the following Table 2. The numerical values in Table 2 were rounded off to the second decimal place and indicated only to the first decimal place.
[0059] [Table 2]
[0060]
[0061] From the results shown in Table 2 above, it can be confirmed that the actual inflammatory response inhibition rate of gluconolactone and climbazole together increased by about 14.4% in the combination of using 25 μM of each substance, compared with the expected value expected when gluconolactone and climbazole were used together. That is, it can be seen that the combination of gluconolactone and climbazole showed a significant synergistic effect in inhibiting inflammatory response.
[0062] Experimental Example 2: Evaluation of improvement in scalp condition
[0063] In order to confirm the efficacy of the scalp condition improving composition prepared according to the present disclosure, Example 1 prepared a scalp condition improving composition (shampoo) containing gluconolactone and climbazole, and Comparative Example 1 prepared a composition not containing gluconolactone and climbazole (see Table 8).
[0064] The experimental group consisted of 12 adults aged 26 to 45 with scalp pruritus, and the experiment was conducted in two groups of 6. The evaluation product was a wash-off shampoo type, and after using 3 ml of the product once a day for 2 weeks, the degree of improvement in the scalp condition was evaluated using the following items.
[0065] (1) Evaluation of transepidermal water loss (TEWL) of the scalp
[0066] Transepidermal water loss indicates the extent of water evaporation from the skin and tends to increase when the skin's barrier function is impaired.
[0067] In order to measure the transepidermal water loss of the scalp of the test subject in the state of more than 8 hours after washing the hair, the scalp was left to stand for 20 minutes under constant temperature and humidity conditions (23°C, relative humidity 40%) to pass the application time. After 20 minutes of adaptation, the transepidermal water loss was measured at the four intersections of the top of the head, the head, the front of the head, the back of the head and the midline of the head using a transepidermal water loss meter (Tewameternano probe, MPA6, C+K Electronic Co., Germany).
[0068] After that, the scalp condition improving composition (Comparative Example 1, Example 1) was used for 2 weeks, and the transepidermal water loss of the same part was measured after 1 week and 2 weeks of using the composition. Among the subjects who used Comparative Example 1, one person stopped the test midway.
[0069] The measurement results are shown in Tables 3 and 4 below as the average values of 5 people in Group 1 of Comparative Example and 6 people in Group 1 of Example. Figure 1 middle.
[0070] [Table 3]
[0071]
[0072] From Table 3 and Figure 1 It can be confirmed that when Example 1 (which is a scalp condition improving composition containing gluconolactone and climbazole) is used, the transepidermal water loss is significantly reduced compared to when Comparative Example 1 (which is a control group not containing gluconolactone and climbazole) is used.
[0073] (2) Evaluation of improvement of scalp hypertrophy and keratinization
[0074] In order to evaluate the keratin-thickening improving effect of the scalp condition improving composition of the present disclosure, keratin was collected from the scalp of the above-mentioned test subjects and the thickness of the keratin was analyzed.
[0075] The scalp hypertrophic keratin was analyzed using Visioscan VC98 (C+K Electronic Co., Germany). For keratin collection and measurement, a special tape, namely Black D-Squame tape, was prepared in the shape of 0.5 cm × 2.5 cm and attached to the scalp surface of the subject at the four intersections of the top of the head, the head, the front of the head, the back of the head and the midline of the head for 10 seconds. The tape was then removed and attached to Visioscan (CCD camera) and photographed to measure the amount of keratin collected. It is difficult to exclude the interference of hair from the desquamation index and the total amount of keratin collected after software analysis, so the ratio of the highest hypertrophic keratin (G4) to the total area of keratin collected and analyzed (G0+G1+G2+G3+G4) was defined as the "hypertrophic keratin ratio" and analyzed.
[0076] Hypertrophic keratin ratio = G4 / (G0+G1+G2+G3+G4)
[0077] The above parameters G0, G1, G2, G3, G4, etc. refer to those defined in “Relationship between lip skin biophysical and biochemical characteristics with corneocyte unevenness ratio as a new parameter to assess the severity of lip scaling” International Journal of Cosmetic Science, (2021), Vol. 43, No. 3, pp. 275-282.
[0078] The above-mentioned hypertrophic keratin ratio is expressed as an improvement rate relative to the measured value before product use (0 week), and the calculation formula of the improvement rate is as follows.
[0079] Improvement rate of hypertrophic keratin = (ab) / a×100
[0080] a: Ratio of thickened keratin before using the product
[0081] b: Ratio of keratin thickening after product use
[0082] The measurement results are shown in Table 4 below as the average values of the 5 people in the group of Comparative Example 1 and the 6 people in the group of Example 1. The representative images of the group of Example 1 before and after the use of the product are shown in Figure 2 middle.
[0083] [Table 4]
[0084]
[0085] From Table 4 and Figure 2 It was confirmed that when Example 1 (which is a scalp condition improving composition containing gluconolactone and climbazole) was used, the thickened keratin was improved compared with when Comparative Example 1 (which is a control group not containing gluconolactone and climbazole) was used.
[0086] (3) Analysis of interleukin-8 in the collected keratin
[0087] In order to confirm the efficacy of the scalp condition improving composition of the present disclosure in improving the secretion of inflammatory factors, the interleukin-8 content in keratin was analyzed using keratin collected from the scalp of the above-mentioned test subjects.
[0088] Interleukin-8 (IL-8) is a representative inflammatory factor that is known to increase in keratin samples of lesions such as dandruff or seborrhoeic dermatitis in literature such as "Dandruff / seborrhoeic dermatitis is characterizedby an inflammatory genomic signature and possible immune dysfunction: transcriptional analysis of the condition and treatment effects of zincpyrithione" British Journal of Dermatology, (2012), Vol. 166, No. S2, pp. 33-40.
[0089] For keratin collection, a special tape, Black D-Squame tape, was prepared in a size of 0.5 cm × 2.5 cm and attached to the scalp surface of the subject at the four intersections of the top, head, front head, back head and midline of the head for 10 seconds, then the tape was removed and the removed keratin was dissolved using 1X PBS containing 0.05% Tween 20 + 0.5 mM EDTA. The keratin was sonicated for 60 minutes together with the buffer solution, and the dissolved keratin solution was quantitatively analyzed using a human IL-8 ELISA kit (Human IL-8 ELISA kit) (R&D system).
[0090] In addition, the total protein amount in the dissolved keratin solution was confirmed by BCA protein assay (ThermoFisher Scientific), and the amount of IL-8 in the keratin was normalized with the total protein amount and analyzed.
[0091] The analysis results of IL-8 are shown in Table 5 below as average values of the five subjects in the group of Comparative Example 1 and the six subjects in the group of Example 1.
[0092] [Table 5]
[0093]
[0094] From Table 5 above, it can be confirmed that when Example 1 (a scalp condition improving composition containing gluconolactone and climbazole) was used, IL-8 secretion was improved compared with when Comparative Example 1 (a control group not containing gluconolactone and climbazole) was used.
[0095] (4) Sensory evaluation
[0096] In order to confirm the efficacy of the scalp condition improving composition prepared according to the present disclosure, the following questionnaire survey was conducted on the subjects participating in the above evaluation, and a sensory evaluation was performed. The evaluation products were the water-rinsable shampoo types of Comparative Example 1 and Example 1. After using each product once a day within 1 week, a questionnaire survey was conducted to confirm the subjective improvement of the scalp condition, and the evaluation scale was 0 to 5 points. The self-diagnosis criteria for each item are shown in Table 6 below.
[0097] [Table 6]
[0098]
[0099] The results of the sensory evaluation are shown in Tables 7 and 8 below as the average values of the five samples in Comparative Example 1 and the six samples in Example 1. Figure 3 middle.
[0100] [Table 7]
[0101] Satisfaction Improve scalp itching Improve exfoliation Improve scalp odor Thrill Comparative Example 1 2.90±1.02 2.80±1.30 2.80±0.84 2.20±0.84 1.60±0.96 Example 1 3.67±1.08 3.42±1.56 2.92±1.20 3.08±1.02 0.83±0.26
[0102] From Table 7 and Figure 3 It can be confirmed that when Example 1 (which is a scalp condition improving composition containing gluconolactone and climbazole) is used, scalp pruritus, keratin loss, and scalp odor are significantly improved, and scalp irritation is also reduced compared to when Comparative Example 1 (which is a control group not containing gluconolactone and climbazole) is used.
[0103] It can be seen from the above experimental results that the scalp condition improving composition disclosed in the present invention has significantly excellent effects of improving transepidermal water loss, improving hypertrophic keratin, improving IL-8 secretion, improving scalp pruritus, improving shedding keratin, and improving scalp odor through the combination of gluconolactone and climbazole.
