Orf virus four-gene deletion strain and vaccine preparation and application thereof

Abstract

The invention provides an orf virus four-gene deletion strain and preparation and application of a vaccine thereof, and belongs to the technical field of epidemic prevention. According to the invention, an orf three-gene deleted strain rGS14-TrypMut is used as a parent strain, and a gene VIL-10 is knocked out, so that an orf virus four-gene deleted strain rGS14-QuadMut is obtained. The rGS14-TrypMut disclosed by the invention has an overall growth speed similar to that of a parent strain. According to the invention, the stable virus titer of the rGS14-QuadMut in a primary sheep testis cell line is stabilized at 105.5 to 106.5 TCID50 / 0.1 ml; and after multiple generations of passage, the stability of the rGS14-QuadMut is good. The rGS14-QuadMut disclosed by the invention has the advantages of genetic stability, weak toxicity, high safety, strong immunogenicity and the like, and the immunosuppression effect can be obviously reduced, so that the safety of a vaccine is improved.
