Abstract

The invention discloses a leukocyte differential counting kit, comprising: a hemolytic agent comprising an organic acid salt buffer solution, a cationic surfactant, a zwitterionic surfactant, an organic alcohol and a preservative; the cell staining agent comprises a fluorescent dye. A small amount of organic alcohol is added into the hemolytic agent, so that the osmotic pressure of the hemolytic agent is controlled within the hypotonic range of 100-150mOsm / L, a pressure difference is formed inside and outside cells, leukocytes are easier to crush and fix, and meanwhile, the dosage of a surfactant can be reduced; besides, weak acidity formed by organic acid anions in the organic acid salt buffer solution is combined with a low osmotic pressure environment, so that the leukocytes can stably maintain the form while the leukocytes are broken, and the leukocyte cell membrane stabilizing effect of the zwitterionic surfactant is combined, so that the leukocyte cell membrane stabilizing effect is improved; by means of the method, lymphocytes, monocytes, neutrophils (basophilic granulocytes) and eosinophils in an aged sample and a fresh sample can be accurately classified and counted at the same time.
