Abstract

The invention belongs to the technical field of antibodies, and particularly relates to an antibody for specifically treating milk goat lamb aphtha and application of the antibody. The method comprises the following steps: collecting a virus solution at a diseased part of a diseased sheep, and carrying out subculture amplification on goat skin fibroblasts to obtain an ORFV virus strain; immunizing milk goats with the attenuated vaccine to prepare high-titer immune serum; the method comprises the following steps: separating ORFV specific B cells from PBMC (peripheral blood mononuclear cells) by adopting a flow sorting technology; antibody genes are amplified through nested PCR, an expression vector is constructed, and 293T cells are transfected to express the monoclonal antibody. The obtained monoclonal antibody 2 has high affinity and specificity, the sequence of a heavy chain CDR1-3 region of the monoclonal antibody 2 is SEQ ID NO: 1-3, and the sequence of a light chain CDR1-3 region of the monoclonal antibody 2 is SEQ ID NO: 4-6. The antibody can be used for diagnosing, treating and preventing the orf, has the characteristics of high efficiency, specificity and stability, and provides a new biological preparation for preventing and controlling the orf.
