Double-layer DNA hydrogel as well as preparation method and application thereof

Abstract

The invention discloses a double-layer DNA hydrogel and a preparation method and application thereof, the hydrogel comprises an upper photo-thermal layer hydrogel and a lower extraction layer hydrogel, and the hydrogel is of a three-dimensional network structure; the upper photo-thermal layer hydrogel contains a photo-thermal material; the lower extraction layer hydrogel contains a capture probe H1-CAR, and the capture probe is connected with the gel through an amidation reaction between carboxyl and amino on the gel; the preparation method comprises the following steps: mixing a hydrogel ligand, a cross-linking agent and an initiator, then adding a capture probe, uniformly mixing, and adding an accelerant for cross-linking to form lower-layer gel; mixing a photo-thermal material with the hydrogel precursor solution, covering the lower-layer gel with the mixture, adding an accelerant, and drying to obtain the double-layer DNA hydrogel. According to the hydrogel material prepared by the invention, through the arrangement of a two-layer structure, high-efficiency photothermal conversion is realized, the selective enrichment efficiency of lower-layer DNA hydrogel on ARGs is remarkably improved, and the detection sensitivity is improved.
