A utr element hh and its construction method and application

Abstract

The application provides a UTR element Hh and a construction method and application thereof, and relates to the technical field of mRNA. Ribosome load prediction and secondary structure optimization are respectively performed on the 5'UTR of a gene with the NCBI number AF339412.1 and the 5'UTR of a gene with the NCBI number MK475748.1 through a PaddleHelix platform, an optimized sequence obtained avoids inhibitory hairpin structures, and the expression amount of luciferase is obviously improved compared with the natural UTR. On the basis of the optimized sequence, HBB and hbb are directionally assembled through a molecular cloning technology to form a HBB-hbb chimera (Hh), the DNA sequence of the Hh is shown as SEQ NO. 1, the RNA sequence is shown as SEQ NO. 2, and the protein expression amount is further improved.
