Abstract

The invention relates to a culture method and application of periosteal organs, and belongs to the technical field of biomedicine. The invention provides a culture method of periosteum organs. The culture method comprises the following steps: (1) digesting periosteum tissues to obtain a cell resuspension; (2) centrifuging the cell resuspension, taking precipitate to obtain periosteum cells, and mixing the periosteum cells with a basement membrane matrix to obtain a periosteum cell resuspension; and (3) mixing the periosteal cell resuspension with a ROCK inhibitor, and adding the mixture into an organoid culture medium for culturing to obtain the periosteal organoid. The first periosteum organ capable of being amplified in vitro for a long time is constructed, aging of HSC in the in-vitro culture process can be effectively inhibited, the repairing effect of the periosteum organ on bone defects is verified by means of thighbone and skull defect models, and the potential application of the periosteum organ on bone tissue regeneration is revealed.
