Abstract

The invention provides a preparation method of pericyte-like phenotype mesenchymal stem cells and application of the pericyte-like phenotype mesenchymal stem cells in promoting angiogenesis, and relates to the technical field of cell biology. According to the preparation method, mesenchymal stem cells in the logarithmic phase are induced and cultured in a culture medium containing nitric oxide donors with effective concentration for 3 days, the final concentration of sodium nitroprusside is 200 mu M, and the pericyte-like phenotype mesenchymal stem cells can be obtained. The P-MSC is represented as a slender and flat pericyte-like form, pericyte markers PDGFR-beta, CYP1B1 and S1PR1 are obviously up-regulated, an endothelial / hematopoietic marker CD34 is not obviously expressed, and dry markers CD271, c-myc and p-eNOS are continuously expressed and have stable phenotypes. The P-MSC can significantly enhance the angiogenesis ability by promoting endothelial cell tubulation and up-regulating secretion of pro-angiogenic factors such as PDGF-BB, ANGPT1, VEGF alpha and the like, can be prepared into a pharmaceutical composition or a cell preparation, and is used for treating ischemic diseases such as diabetic wounds, peripheral artery diseases, myocardial infarction and cerebral arterial thrombosis.
