Abstract

The application provides a method for rapidly permeating cells and application thereof, and relates to the technical field of biology.The method for rapidly permeating cells provided by the application selects tert-butyl alcohol instead of dimethylbenzene as a permeating agent, utilizes the characteristic that the tert-butyl alcohol can form needle-shaped crystals in a frozen state, mixes the cells with the tert-butyl alcohol, controls the cooling speed and time to realize the permeation effect of physical punching on the cells, reduces the risk of operators, avoids the shortcomings of dimethylbenzene as a permeating agent, shortens the cell permeation time, and improves the efficiency.In addition, low-concentration tert-butyl alcohol also has the effects of reducing protein oxidation and breaking, and protecting antibody binding sites, thereby avoiding the problem of loss of antibody recognition sites caused by the use of dimethylbenzene.
