Abstract

The application provides a detection method of porcine coronavirus, which comprises the following steps: extracting RNA of a sample to be detected; reversely transcribing the obtained RNA of the sample to be detected into cDNA; adding a pre-designed detection kit to perform PCR amplification reaction, to obtain a PCR amplification product, wherein the detection kit comprises a first DPO primer pair for detecting porcine transmissible gastroenteritis virus, a second DPO primer pair for detecting porcine coronavirus type D, a PCR premix, and nuclease-free water; identifying the PCR amplification product by using gel electrophoresis, and setting positive and negative standard samples as controls; and judging whether the porcine coronavirus exists in the sample to be detected by the gel electrophoresis result. The application can quickly and accurately distinguish and detect two main coronaviruses causing pig diarrhea, i.e., porcine transmissible gastroenteritis virus and porcine coronavirus type D, in the same reaction.
