Abstract

This invention discloses a near-infrared fluorescent probe for detecting fluoride ions, its preparation, and its application. The probe, named DCSX-OT, utilizes a near-infrared fluorescent dye, DCSX-OH, as the fluorescent signal reporter group, with dicyanomethylene-4H-chromene as the electron acceptor group and thiosubstituted thionone as the electron donor group. A fluoride ion-responsive probe is constructed by protecting the phenolic hydroxyl group with tert-butyldimethylsilyl and using a self-eliminating benzyl group as the linker arm. This probe exhibits excellent properties such as high selectivity, high sensitivity, a large Stokes shift (165 nm), and a long emission wavelength (810 nm) for fluoride ions. It enables the detection and imaging of fluoride ions in live cells and live mouse tissues, and can accurately determine the fluoride ion content in real water samples and toothpaste samples. This technology provides powerful tool support for the detection of fluoride ions in biological systems, environmental samples, and commodities, and has significant application value in physiological and pathological research, environmental monitoring, and food safety testing.
