Intermediates for the preparation of glucagon and glp-1 dual agonists and processes for their preparation

Abstract

This invention provides novel intermediates and methods useful in the manufacture of mazdutide or its pharmaceutically acceptable salts. The method of this invention uses Fmoc-Gly-Gly-OH instead of sequentially coupling Fmoc-Gly-OH, and employs direct coupling of the peptide chain with an OSU-activated ester of the side chain synthesized via DIC / HOSu. This shortens the reaction cycle, reduces potential racemization and impurities, resulting in fewer steps while maintaining and improving quality and purity.
