Application of C5aR1 inhibitors in the preparation of drugs for the treatment of complement-related inflammatory diseases

Abstract

This invention discloses the application of C5aR1 inhibitors in the preparation of therapeutic drugs for complement-related inflammatory diseases, belonging to the field of biomedical technology. The C5aR1 inhibitor can inhibit C5aR1 gene expression, reduce C5aR1 protein content, or downregulate C5aR1 protein expression, and is selected from one or more of nucleic acid molecules, small molecule compounds, antibodies, peptides, proteins, recombinant expression vectors, and recombinant viral vectors. The nucleic acid molecules include miR-132-3p inhibitors having the sequence shown in SEQ ID NO:1 or its modified products; the recombinant expression vectors include expression vectors carrying the ZFP36L1 encoding nucleic acid sequence shown in SEQ ID NO:2 or its modified products. This invention can effectively block hypoxia-induced abnormally high expression of C5aR1 on the cell surface, and can be formulated into various pharmaceutical dosage forms after adding pharmaceutically acceptable excipients. This invention provides a novel direction for drug development and clinical intervention strategies for complement-related inflammatory diseases mediated by excessive activation of the C5aR1 pathway under hypoxic microenvironment.
