A recombinant human type I collagen, its preparation method and application

Abstract

This invention discloses a recombinant human type I collagen, its preparation method, and its applications. The amino acid sequence of the recombinant human type I collagen is shown in SEQ ID NO.1. This invention effectively solves the problems of insolubility and purification difficulties of the expression product (recombinant collagen) by rationally designing the sequence of natural human type I collagen, retaining the core functional domains, removing non-essential and insoluble telopeptide regions, and introducing hydrophilic short peptides that facilitate soluble expression. This invention also optimizes the codons for mammalian CHO cells, achieving efficient secretory expression of the synthesized nucleic acid sequence in CHO cells in suspension culture, significantly reducing production costs. The recombinant human type I collagen of this invention fully retains key cell recognition sequences (such as GER), enabling it to promote cell adhesion, spread, and proliferation.
