Abstract
This invention discloses a genetic transformation method for Leymus chinensis using Agrobacterium, belonging to the fields of plant biotechnology and molecular breeding technology. This method uses callus induced from mature embryos of Leymus chinensis as the recipient material. By optimizing callus screening, Agrobacterium infection, co-culture, resistance screening, differentiation and regeneration, and rooting culture conditions, a highly efficient genetic transformation system suitable for different varieties and genotypes of Leymus chinensis was established. Using this method, regenerated plants can be obtained from different varieties and genotypes of Leymus chinensis, with an average regeneration efficiency of 40.69% and an average genetic transformation efficiency of 11.03%. The regeneration efficiency of superior callus lines can reach 100%, and the transformation efficiency can reach 72%, and positive transgenic plants can be obtained.
