Abstract

This invention provides the application of human umbilical cord stem cell (hUCMSCs) external vesicles in the preparation of anti-peritoneal fibrosis drugs, belonging to the field of biomedical technology. In this invention, human umbilical cord mesenchymal stem cells (hUCMSCs) are pre-induced for 24–72 h under conditions of 0.5%–3% oxygen (v / v) and 4.0%–4.5% glucose (w / w), followed by purification via differential centrifugation, ultracentrifugation, size exclusion chromatography, and ultrafiltration to obtain external vesicles. This invention simultaneously optimizes the yield and function of external vesicles. High-glucose pre-induction allows hUCMSCs to adapt to and respond to a high-glucose microenvironment during vesicle secretion. By constructing a high-glucose environment similar to that of peritoneal dialysis patients in vitro, hUCMSCs are allowed to "sense" the pathological state in advance, secreting external vesicles rich in higher concentrations of anti-inflammatory and anti-fibrotic active molecules.
