Description

Technical Field

[0001] This application relates to the field of instruments for biological cell engineering, specifically an automated collection device for adipose matrix cells. Background Technology

[0002] Human adipose tissue is a connective tissue primarily composed of clustered adipocytes supported by loose connective tissue. Like bone marrow and other tissues, adipose tissue is isolated from the human mesenchymal tissue and is rich in an easily separable matrix. A 2002 study defined this adipose-derived regenerative functional cell population as "processed liposuction" (PLA) and suggested that adipose tissue could serve as a source of regenerative functional cells with long-term therapeutic potential in numerous fields. Similar to mesenchymal stem cells, PLA also possesses the ability to differentiate into osteoblasts, chondrocytes, myogenes, or adipocytes in vitro.

[0003] Current adipose tissue matrix cell extraction procedures require manual intervention in multiple steps. Each manual intervention necessitates opening the extraction system, which introduces the risk of external contaminants entering the system and contaminating the adipose tissue or matrix cells. Therefore, manual intervention requires operators with extensive experience and skill to ensure efficiency and prevent contamination of the extracted matrix cells, placing high demands on the operator's expertise. Summary of the Invention

[0004] To reduce the possibility of contamination in the extraction system and to improve the automation and extraction efficiency of the adipose matrix cell extraction system, this application provides an automated adipose matrix cell collection device.

[0005] An automated adipose-derived matrix cell collection device includes: a piping unit; a cleaning unit; a waste liquid unit; a collection unit including a collection container; a containment unit connected to the cleaning unit and the waste liquid unit via the piping unit, forming a fluid path from the cleaning unit to the containment unit and a fluid path from the containment unit to the waste liquid unit; and a centrifugation unit including a rotatable centrifugation chamber, the internal cavity of which includes a residence region one and a residence region two with different centrifugation radii, the residence region one being located radially outside the residence region two within the centrifugation chamber, for collecting... The container is connected to the first residence area via a piping unit to form a fluid path from the first residence area to the collection container. The waste liquid unit is connected to the second residence area via a piping unit to form a fluid path from the second residence area to the waste liquid unit. A peristaltic pump unit is provided in the piping unit to drive the fluid to flow in the fluid path. A clamp valve body unit is provided in the piping unit to control the opening and closing of each fluid path. The piping unit connecting the centrifuge chamber and the collection container is provided with a sterile disconnector that can disconnect and seal the piping to disconnect the collection container from the piping unit.

[0006] Autologous adipose tissue is added to the containment unit. An automated system adds adipose-cleansing solution from the cleaning unit to the containment unit containing the adipose tissue. The containment unit then agitates and allows the mixture to settle, resulting in separation of fat and waste liquid. A waste liquid unit collects the waste liquid from the containment unit. Next, enzyme solution is added manually or automatically to the adipose tissue in the containment unit. The enzymatically hydrolyzed adipose tissue is then transferred to a centrifuge chamber for centrifugation, separating the fluid containing adipose matrix cells from the waste liquid. The waste liquid unit automatically collects the waste liquid generated during centrifugation, leaving the centrifuge chamber containing the adipose matrix cells. The cleaning solution from the cleaning unit is then automatically added to the centrifuge chamber, which continues to rotate, cleaning the fluid containing the adipose matrix cells. The cleaned waste liquid from the centrifuge chamber is automatically transported to the waste liquid unit for collection. Finally, the cleaned fluid containing the adipose matrix cells from the centrifuge chamber is automatically transported to a collection container in the collection unit. A sterile disconnect device disconnects the collection container from the tubing unit, and the collection container preserves the adipose matrix cells.

[0007] In one embodiment of this application, along the straight line of the rotation axis of the centrifuge chamber, from the first end of the centrifuge chamber to the second end, the radial dimension of the inner cavity of the centrifuge chamber gradually increases, and the fluid path from the centrifuge chamber to the waste liquid unit starts at the second end and connects with the second residence area.

[0008] As the centrifuge chamber rotates, the fluid containing adipose matrix cells adheres to the inner wall of the chamber. Over time, the fluid begins to stratify radially within the chamber. The densest adipose matrix cells are thrown towards the area with the largest radial dimension, entering the first residence area with a larger centrifugation radius. The waste liquid, containing less dense substances such as free lipid droplets, water, and enzymes, is suspended in the second residence area, which is closer to the centrifuge chamber's rotation axis and has a smaller centrifugation radius. Since the fluid path from the centrifuge chamber to the waste liquid unit starts at the second end and connects to the second residence area, the device automatically pumps the waste liquid from the second residence area into the waste liquid unit. This achieves simultaneous centrifugation and extraction of waste liquid from the centrifuge chamber, thereby increasing the concentration of adipose matrix cells within the centrifuge chamber and facilitating their enrichment.

[0009] Furthermore, the rotation axis of the centrifuge chamber is vertically oriented, with the second end located above the first end. The starting point of the fluid path from the centrifuge unit to the collection container is connected to the inner cavity of the centrifuge chamber at the first end.

[0010] While centrifuging, the waste liquid generated during centrifugation is pumped to the waste liquid unit. After the centrifuge chamber stops rotating, the device automatically pumps the fat washing solution from the washing unit into the centrifuge chamber. The centrifuge chamber enters a short cycle of rotation and stopping, allowing the fluid containing adipose matrix cells to be fully mixed with the fat washing solution, thus washing the fluid containing adipose matrix cells. Subsequently, the centrifuge chamber maintains high-speed rotation for N minutes, causing the fluid enriched with adipose matrix cells and the waste liquid generated after washing to separate into two layers: adipose matrix cells in residence area one and waste liquid in residence area two. During centrifugation, the device automatically pumps the washed waste liquid to the waste liquid unit, achieving separation of adipose matrix cells and waste liquid. Then, the centrifuge chamber stops rotating, and the fluid containing adipose matrix cells flows downward to the first end of the centrifuge chamber under the action of gravity. The device automatically pumps the fluid at the first end into the collection container, thereby collecting the adipose matrix cells.

[0011] In one embodiment of this application, the rotation axis of the centrifuge chamber is vertically oriented. The inner cavity of the centrifuge chamber includes a first cavity and a first space located radially outside the first cavity. The chamber wall separates the first cavity and the first space. Along the direction from the lower end to the upper end of the centrifuge chamber, the radial dimension of the first cavity gradually increases. The inner wall of the first space, which is furthest from the rotation axis of the centrifuge chamber in the radial direction, gradually moves away from the rotation axis of the centrifuge chamber. The upper end of the first cavity is connected to the upper end of the first space. At the connection between the first cavity and the first space, a first residence area is located in the first space, and a second residence area is located in the first cavity. The end point of the fluid path from the receiving unit and the cleaning unit to the centrifuge unit is located in the first cavity, and the starting point of the fluid path from the centrifuge unit to the waste liquid unit is located at the lower end of the first cavity. The starting point of the fluid path from the centrifuge unit to the collection container is located at the lower end of the first space.

[0012] The above structure is another implementation of the centrifuge chamber. The centrifuge chamber has a first cavity and a first space. When the fluid containing adipose matrix cells in the containing unit enters the centrifuge chamber and the centrifuge chamber starts to rotate, the fluid begins to rise in the centrifuge chamber under the action of centrifugal force. When it reaches the connection between the first space and the first cavity, the adipose matrix cells with the highest density in the fluid will be thrown towards the part with the largest radial dimension of the centrifuge chamber. They will then enter the first residence area with a larger centrifugal radius through the connection, that is, enter the upper end of the first space. The waste liquid remains in the first cavity, located in the second residence area with a smaller centrifugal radius, thereby achieving the separation of adipose matrix cells and waste liquid. Subsequently, the centrifuge chamber stops rotating, and the fluid returns to its original state under the influence of gravity. Under the action of the device, the fluid enriched with adipose matrix cells falls in the first space, and the waste liquid falls in the first cavity. The waste liquid unit collects the waste liquid. Then, the device automatically injects the fat cleaning solution from the cleaning unit into the first cavity. Subsequently, the centrifuge chamber rotates, and the fluid enriched with adipose matrix cells in the first space comes into contact with the cleaning solution in the first space at the connection point to clean the adipose lipid cells. Then, the centrifuge chamber stops rotating, and the device automatically pumps the fluid enriched with adipose matrix cells at the bottom of the first space to the collection container. Then, a sterile disconnector is used to disconnect the connection between the collection container and the pipeline unit, so that the collection container can transfer and preserve the adipose matrix cells.

[0013] In one embodiment of this application, the automated adipose matrix cell collection device further includes an enzyme solution unit, which is connected to a containment unit via a pipeline unit to form a fluid path from the enzyme solution unit to the containment unit.

[0014] This device can automatically pump the enzyme solution to the containment unit, thereby enzymatically digesting the adipose tissue within the containment unit. Compared to manually adding the enzyme solution to the containment unit, this device reduces the possibility of contamination of adipose matrix cells and improves collection efficiency by automatically adding the enzyme solution to the collection system.

[0015] In one embodiment of this application, the enzyme solution unit includes an enzyme bottle mounting structure, which includes an air exchange pipe capable of inserting an enzyme bottle and equipped with an air filter, and an enzyme bottle connecting pipe capable of inserting an enzyme bottle to allow fluid to enter and exit the enzyme bottle; the pipeline unit includes an enzyme bottle working pipe, and an enzyme bottle working branch pipe one and an enzyme bottle working branch pipe two connected to one end of the enzyme bottle working pipe, the other end of which is connected to the enzyme bottle connecting pipe; the enzyme bottle working branch pipe one is connected to a receiving unit to form a fluid path from the enzyme solution unit to the receiving unit; the enzyme bottle working branch pipe two is connected to a cleaning unit to form a fluid path from the cleaning unit to the enzyme solution unit; the peristaltic pump unit includes an enzyme bottle peristaltic pump, which is provided in the enzyme bottle working pipe; the clamp valve body unit includes an enzyme bottle clamp valve body, which is provided in both the enzyme bottle working branch pipe one and the enzyme bottle working branch pipe two.

The drive pipeline includes working end one and working end two; working end one is connected to the receiving unit through pipeline one and to the cleaning unit through pipeline two; working end two is connected to the centrifuge chamber through pipeline three and to the cleaning unit through pipeline four.

[0017] In one embodiment of this application, the waste liquid unit includes a waste liquid container; the pipeline unit includes a waste liquid main pipe, a waste liquid branch pipe one, and a waste liquid branch pipe two; one end of the waste liquid main pipe is connected to the waste liquid container, and the other end is connected to the containing unit through the waste liquid branch pipe one, and to the centrifuge chamber through the waste liquid branch pipe two; the peristaltic pump unit includes a waste liquid peristaltic pump, and the waste liquid main pipe is equipped with a waste liquid peristaltic pump; the clamp valve body unit includes a waste liquid clamp valve body, and the waste liquid branch pipe one and the waste liquid branch pipe two are respectively equipped with waste liquid clamp valve bodies.

[0018] The waste liquid unit is connected to the containment unit and the collection unit through separate pipelines, which can avoid waste liquid contamination of the fluid path from the containment unit to the centrifugation unit, from the centrifugation unit to the collection unit, and from the cleaning unit to the containment unit and the collection unit, and is conducive to improving the purity of the adipose matrix cells collected by this device.

[0019] In one embodiment of this application, the peristaltic pump unit includes a collecting peristaltic pump, the clamp valve body unit includes a collecting clamp valve body, the pipeline unit includes a collecting pipeline, the collecting container is connected to the centrifuge chamber through the collecting pipeline, and the collecting pipeline is equipped with a collecting peristaltic pump and a collecting clamp valve body.

[0020] The centrifuge chamber is connected to the collection container through a separate pipeline, which can prevent residual fluid from other pipelines in the pipeline unit from entering the adipose matrix cells that have already been centrifuged and cleaned in the centrifuge chamber and affecting them.

[0021] In one embodiment of this application, the automated adipose matrix cell collection device further includes a liposuction unit connected to the receiving unit.

[0022] The liposuction unit allows fat tissue extracted from the body to be directly injected into the receiving unit, eliminating the need for manual transfer of fat tissue to the receiving unit. This reduces the possibility of contamination of fat tissue during manual transfer and improves the efficiency of fat tissue collection by automatically transferring it to the receiving unit. [0025] To provide a clearer understanding of the technical features, objectives, and effects of this application, specific embodiments of this application are now described with reference to the accompanying drawings. In the drawings, the same reference numerals indicate components with the same or similar structures but the same function.

[0026] In this document, “illustrative” means “serving as an example, illustration or description”, and any illustration or implementation described herein as “illustrative” should not be construed as a more preferred or advantageous technical solution.

[0027] To keep the drawings concise, only the parts relevant to this application are shown schematically in each drawing, and they do not represent the actual structure of the product. In addition, to make the drawings concise and easy to understand, in some drawings, only one of the components with the same structure or function is shown schematically, or only one of them is labeled.

[0028] The "fixed connection" between the two components in this application is achieved through methods such as integral manufacturing, threaded connection, snap welding, and bonding, so that the relative position between the two components does not change or rotate relative to each other during use.

[0029] In this application, "pivotal connection" refers to a component that can rotate relative to another component about a pivot axis. One of the two components has a pivot hole, and the other has a pivot shaft that can be inserted into the pivot hole and is coaxial with the pivot hole. The two components are pivotally connected through the pivot hole and pivot shaft structure, where the axis of the pivot hole is the pivot axis. Alternatively, both components have pivot holes, and the two components are pivotally connected by other pivot members with pivot shafts. Or, both components have pivot shafts, and the two components are pivotally connected by other pivot members with pivot holes. There are several ways to achieve axial and radial limiting between two components without limiting rotation. For example, both the pivot hole and the pivot shaft have circular cross-sections. The pivot hole has two shoulders, and the shape of the pivot hole matches that of the pivot shaft. When the pivot shaft is inserted into the pivot hole, the pivot hole radially limits the pivot shaft, and the two pivot hole shoulders axially limit the two shoulders. Thus, the pivot shaft can only rotate around its own axis within the pivot hole. Of course, other methods can also be used to achieve axial and radial limiting when the two components are pivotally connected.

[0030] The controller in this application is a device that can receive input signals, perform logic processing or calculations, and output control signals to drive other devices or systems to work. The controller can be a microcontroller (MCU), a PLC, or an FPGA, and a control program adapted to this application can be set in the controller.

[0031] In this application, the clamp valve body refers to a valve body in which the clamp can reciprocate to squeeze and close the pipeline, or move away from the pipeline so that the pipeline can return to its natural state under its own elasticity, thus opening the pipeline. The abutment part that cooperates with the clamp to squeeze and close the pipeline can be set on the valve body, or it can be a separate design from the clamp, with the abutment part set on other parts of the device.

[0032] Please see Figures 1 to 14 Learn more about this application.

[0033] To automate the entire process of collecting adipose stromal cells from adipose tissue, this application provides an automated adipose stromal cell collection device.

[0034] See Figure 1 The automated adipose matrix cell collection device includes a pipeline unit, a container unit, a cleaning unit, a centrifugation unit, a waste liquid unit, a collection unit, a controller, a peristaltic pump unit, a clamp valve unit, and a 600-degree sterile disconnector.

[0035] The piping unit consists of multiple pipes.

[0036] The containing unit includes a container for containing adipose tissue, such as a processing tank with an inner cavity, which can hold untreated adipose tissue.

[0037] The cleaning unit includes a container holding a fat cleaning solution, which includes any one or more of the following: physiological saline, Ringer's solution, lactated Ringer's solution, acetate Ringer's solution, compound sodium acetate Ringer's solution, and sodium bicarbonate Ringer's solution.

[0038] The centrifugation unit includes a centrifuge chamber 500 that can rotate around a rotation axis. The rotating centrifuge chamber 500 can centrifuge the fluid containing adipose matrix cells, thereby separating the adipose matrix cells from the waste liquid containing low-density substances such as free lipid droplets, water, and enzymes.

[0039] Waste liquid unit, which includes a container capable of holding waste liquid.

[0040] A peristaltic pump unit located in the piping unit that drives fluid to flow in the fluid path.

[0041] A clamp valve unit located in a pipeline unit to control the opening and closing of each fluid path.

[0042] The collection unit includes a collection container, which can be a heat-sealed export cell collection bag (e.g., a sterile bag directly connected to a TPE tube) from an existing product. The tubing unit connects the centrifuge chamber 500 to the collection container and is equipped with a sterile disconnector 600 capable of disconnecting and sealing the tubing to disconnect the collection bag from the tubing unit. After the device automatically pumps the enriched adipose-derived matrix cells from the centrifuge chamber 500 into the sterile bag, the controller activates the heat-sealed sterile disconnector 600, heating the metal indenter to melt the TPE tube wall. Upon cooling, this melts the wall and forms a one-piece sealing plug, disconnecting the cell collection bag from the tubing and sealing the cell collection bag. Of course, the collection container and the sterile disconnector 600 coupled therewith can also be other implementations. For example, the collection container is a cell collection bag with an aluminum ring pre-installed on the tubing. The blade of the sterile disconnector 600 can squeeze the aluminum ring and disconnect the tubing, thereby sealing the cell collection bag and separating the cell collection bag from the tubing. Of course, the collection container and the sterile disconnector 600 can also be other implementations, which will not be elaborated here.

[0043] The containment unit is connected to the cleaning unit and the waste liquid unit via a piping unit, forming a fluid path from the cleaning unit to the containment unit and a fluid path from the containment unit to the waste liquid unit. The centrifuge unit includes a rotatable centrifuge chamber 500. The internal cavity configuration of the centrifuge chamber 500 includes a first residence area 501 and a second residence area 502 with different centrifugal radii. The first residence area 501 is located radially outside the second residence area 502 of the centrifuge chamber 500. The collection container is connected to the first residence area 501 via a piping unit to form a fluid path from the outer residence area 501 to the collection container. The waste liquid unit is connected to the second residence area 502 via a piping unit to form a fluid path from the second residence area 502 to the waste liquid unit.

[0044] Furthermore, in one embodiment of this application, the automated adipose tissue cell collection device further includes an enzyme solution unit, which is connected to a receiving unit via a piping unit, forming a fluid path from the enzyme solution unit to the receiving unit. This device can automatically add enzyme solution to the receiving unit, eliminating the need for manual addition of enzyme solution to adipose tissue.

[0045] The automated workflow of this device is briefly described below. Step 1: Add the adipose tissue into the processing tank of the containing unit (or other types of containers); Step 2: See... Figure 2 The fat-cleansing solution in the cleaning unit is pumped into the processing tank. The adipose tissue and the fat-cleansing solution are mixed, agitated (the structure of the processing tank can be referenced in CN218115441U), and then allowed to settle and separate into layers. The lower layer consists of blood, anesthetic swelling fluid, and LR (lactated Ringer's solution), while the upper layer is the cleansed adipose tissue. Step 3: See... Figure 3 Through the fluid path from the receiving unit to the waste liquid unit, this device automatically pumps the waste liquid from the treatment tank to the container in the waste liquid unit, while the cleaned fatty tissue remains in the treatment tank: Step 4: See Figure 5 The enzyme solution in the enzyme solution unit is pumped into the processing tank through a fluid path from the enzyme solution unit to the containing unit. The enzymes in the enzyme solution enzymatically hydrolyze the adipose tissue, releasing the adipose matrix cells tightly wrapped in the collagen fiber network intact and gently, forming a cell suspension; Step 5: See Figure 6 The fluid in the treatment tank is pumped into the centrifuge chamber 500 of the centrifuge unit; Step 6: The centrifuge chamber 500 rotates and centrifugation begins. The densest adipose matrix cells are thrown towards the larger centrifugation radius of the centrifuge chamber 500 and enter the preset residence area 1 501. The less dense waste liquid is thrown towards the smaller centrifugation radius of the centrifuge chamber 500 and is located in the preset residence area 2 502. The centrifuged waste liquid is pumped into the container of the waste liquid unit through the fluid path from the centrifuge unit to the waste liquid unit. See [link to relevant documentation]. Figure 7 The centrifuged adipose matrix cells are retained in the centrifuge chamber at 500°C; Step 7: See Figure 8 The fat washing solution is pumped into the centrifuge chamber 500 via a fluid path from the washing unit to the centrifuge unit. The centrifuge chamber 500 rotates, washing the adipose matrix cells within. Step eight: The centrifuge chamber 500 centrifuges, separating the adipose matrix cells from the waste liquid. The waste liquid in the centrifuge chamber 500 is pumped to the waste liquid unit. Step nine: The centrifuge chamber 500 stops rotating, and the fluid containing the adipose matrix cells in the centrifuge chamber 500 is pumped to the collection container in the collection unit. See [link to relevant documentation]. Figure 9 Step 10: The controller controls the aseptic disconnector 600 to operate, causing the collection container to separate from the piping unit and seal it.

Figure 4 , Figure 5 The controller opens the enzyme bottle clamp valve 305 on the enzyme bottle working branch pipe 2 111, and closes the other clamp valves in the clamp valve unit. The controller also controls the enzyme bottle peristaltic pump 201 to operate. The fat cleaning solution in the cleaning unit enters the enzyme bottle connected to the enzyme bottle connecting pipe 112 through the enzyme bottle working branch pipe 2 111, the enzyme bottle working pipe 109, and the enzyme bottle connecting pipe 112, mixing with the enzyme powder inside the enzyme bottle. At the same time, the gas inside the enzyme bottle enters the outside through the ventilation pipe 113 to balance the gas pressure inside and outside the enzyme bottle. See [link to relevant documentation]. Figure 10 Subsequently, the controller opens the enzyme bottle clamp valve 305 on the enzyme bottle working branch pipe 110, closes the other clamp valves in the clamp valve unit, and controls the enzyme bottle peristaltic pump 201 to work. The enzyme solution in the enzyme bottle enters the processing tank through the enzyme bottle working pipe 109 and the enzyme bottle working branch pipe 110 to enzymatically hydrolyze the adipose tissue. At the same time, outside air enters the enzyme bottle after being filtered by the air filter, balancing the air pressure inside and outside the enzyme bottle. See [link to relevant documentation]. Figure 10 When performing step five, please refer to... Figure 6The controller opens working clamp valve body 1 (301) and working clamp valve body 3 (303), while closing the other clamp valve bodies in the clamp valve body unit. The working peristaltic pump 200 operates, and the fluid containing enzymes and adipose tissue in the treatment tank enters the centrifuge chamber 500 through pipe 1 (101), drive pipe 100, and pipe 3 (103). During step six, the controller opens waste liquid clamp valve body 306 on waste liquid branch pipe 2 (107), closes the other clamp valve bodies in the clamp valve body unit, and controls the waste liquid peristaltic pump 202 to operate. The waste liquid in the centrifuge chamber 500 is pumped to a container in the waste liquid unit for storage. During step seven, see [link to relevant documentation]. Figure 8 The controller opens working clamp valve body 2 (302) and working clamp valve body 3 (303), closes the other clamp valve bodies in the clamp valve body unit, and controls the working peristaltic pump 200 to operate, pumping the grease cleaning solution in the cleaning unit into the centrifuge chamber 500. During step eight, the controller opens waste liquid clamp valve body 306 on waste liquid branch pipe 2 (107), closes the other clamp valve bodies in the clamp valve body unit, and controls the waste liquid peristaltic pump 202 to operate, pumping the waste liquid in the centrifuge chamber 500 into a container in the waste liquid unit for storage. See [link to relevant documentation]. Figure 7 When performing step nine, the controller opens the collection clamp valve 300 on the collection line 108, closes the remaining clamp valves in the clamp valve unit, and activates the collection peristaltic pump 203. This pumps the fluid containing adipose matrix cells within the centrifuge chamber 500 to the collection container in the collection unit. (See [link to relevant documentation]). Figure 9 .

[0052] Of course, those skilled in the art to which this application pertains will understand that the enzyme in the enzyme bottle in this application is not limited to being in the form of enzyme powder, but can also be in the form of enzyme solution. If the enzyme in the enzyme bottle is in the form of enzyme solution, then it is not necessary to use the fat cleaning solution in the cleaning unit to reconstitute the enzyme powder, so the enzyme bottle working branch pipe 2 111 and the enzyme bottle clamp valve body 305 provided on the enzyme bottle working branch pipe 2 111 can be eliminated.

[0053] To meet the automation requirements of the collection device, this application has made corresponding designs for the centrifuge chamber 500, thereby realizing the automatic discharge of waste liquid and the collection of adipose matrix cells.

[0054] See Figure 11 , Figure 12 , Figure 14The pipeline unit includes a centrifuge chamber 500 connecting structure, which includes a stationary part 400 and a rotating part 401. The interior of the stationary part 400 is hollow. The outer surface of the stationary part 400 is provided with three through holes that communicate with the interior. The three through holes are respectively connected to the third pipe 103, the second waste liquid branch pipe 107, and the collection pipe 108. The rotating part 401 is inserted into the stationary part 400 and is coaxially arranged with the stationary part 400. The rotating part 401 is pivotally connected to the stationary part 400 and the rotating part 401 is rotaryly sealed with the stationary part 400. The rotating part 401 can rotate in the hollow area inside the stationary part 400. The outer wall of the rotating part 401 is provided with three annular grooves along its axial direction. The three annular grooves are correspondingly arranged and connected with the three through holes provided in the stationary part 400. Sealing rings are provided at both ends of the rotating part 401 and between the three annular grooves, so that the annular grooves are not connected to each other and are not connected to the outside. For better explanation of this application, the three annular grooves are named the first annular groove 402, the second annular groove 403, and the third annular groove 404. Since the annular grooves are arranged circumferentially along the rotating part 401, the three annular grooves remain connected to the three through holes provided on the stationary part 400 when the rotating part 401 rotates. The rotating part 401 is also provided with an inlet channel 405, a waste discharge channel 406, and a collection channel 407. The inlet channel 405 is connected to the third pipe 103 through the first annular groove 402, the waste discharge channel 406 is connected to the second waste liquid branch pipe 107 through the second annular groove 403, and the collection channel 407 is connected to the collection pipe 108 through the third annular groove 404.

[0055] See Figure 11 , Figure 12 In one embodiment of this application, along the straight line of the rotation axis of the centrifuge chamber 500, from the first end to the second end of the centrifuge chamber 500, the radial dimension of the inner cavity of the centrifuge chamber 500 gradually increases. (See also...) Figure 11The first end is located below the second end, and the rotation axis of the centrifuge chamber 500 is vertically oriented. The second end is located above the first end. The centrifuge chamber 500 is fixedly connected to the rotating part 401, and the rotation axis of the centrifuge chamber 500 is coaxial with that of the rotating part 401. The rotating part 401 and the centrifuge chamber 500 can rotate synchronously. The waste discharge channel 406 extends from the second end to the second retention area 502, so that the starting point of the fluid path from the centrifuge chamber 500 to the waste liquid unit is connected to the second retention area 502 at the second end. The collection channel 407 communicates with the inner cavity of the centrifuge chamber 500 at the first end, and the starting point of the fluid path from the centrifuge unit to the collection container is connected to the inner cavity of the centrifuge chamber 500 at the first end. The connection between the inlet channel 405 and the centrifuge chamber 500 can be located at either the first or second end, such that the end point of the fluid path from the containing unit and the cleaning unit to the centrifugation unit is connected to the centrifuge chamber 500. Fluid containing adipose matrix cells or adipose washing solution can enter the centrifuge chamber 500 through the pipe 103, the first annular groove 402, and the inlet channel 405. Preferably, the inlet channel 405 is connected to the inner cavity of the centrifuge chamber 500 at the first end. This ensures that when fluid containing adipose matrix cells or adipose washing solution enters the centrifuge chamber 500 through the inlet channel 405, the inlet channel 405 is closer to the bottom of the centrifuge chamber 500, thereby reducing the impact force of the fluid on the bottom of the centrifuge chamber 500 and protecting the adipose matrix cells inside the centrifuge chamber 500.

[0056] The fluid in the containing unit or the fat cleaning solution in the cleaning unit can enter the centrifuge chamber 500 through pipe 103, the first annular groove 402, and the inlet channel 405 to execute step five or step seven. The rotating part 401 rotates synchronously with the centrifuge chamber 500, so the waste discharge channel 406 will rotate synchronously with the centrifuge chamber 500 and remain stationary relative to it. Therefore, during the rotation of the centrifuge chamber 500, due to the change in the radial dimension of the inner cavity of the centrifuge chamber 500 in the direction from the first end to the second end, the fluid inside the centrifuge chamber 500 moves along the inner wall of the centrifuge chamber 500 towards the second end under the action of centrifugal force, ultimately reaching the state shown in the attached figure, where the fat matrix cells are located in the residence area 501, and the waste liquid... When the waste liquid is in the second residence area 502, the controller controls the waste liquid clamp valve body 306 on the waste liquid branch pipe 2 107 to open, and the other clamp valve bodies in the clamp valve body unit to close. The controller controls the waste liquid peristaltic pump 202 to work. The waste liquid in the second residence area 502 is pumped to the container in the waste liquid unit for storage through the waste discharge channel 406, the second annular groove 403, and the waste liquid branch pipe 2 107. This achieves the simultaneous centrifugation and discharge of waste liquid in the centrifuge chamber 500, thus realizing the waste liquid discharge work in step six or step eight. When step eight is completed and step nine begins, the centrifuge chamber 500 stops rotating. The adipose matrix cells located in the residence area 501 flow towards the first end under the action of gravity. Upon reaching the first end, the controller controls the collection clamp valve 300 on the collection pipeline 108 to open, while the remaining clamp valves in the clamp valve unit close. The collection peristaltic pump 203 operates, and the fluid containing adipose matrix cells in the centrifuge chamber 500 enters the container of the collection unit through the collection channel 407, the third annular groove 404, and the collection pipeline 108. Then, step ten is performed to complete the collection of adipose matrix cells.

[0057] Preferably, the centrifuge chamber 500 is conical in shape. Of course, the centrifuge chamber 500 can also be other shapes, as long as it meets the condition that the radial dimension of the inner cavity of the centrifuge chamber 500 gradually increases from the first end to the second end.

[0058] Preferably, a partition 505 is provided between the first residence area 501 and the second residence area 502. The partition 505 is located at the connection between the waste discharge channel 406 and the second residence area 502. When centrifuging and discharging waste liquid simultaneously, the partition 505 can reduce the number of adipose matrix cells enriched in the first residence area 501 that are drawn into the waste discharge channel 406, thereby reducing the loss of adipose lipid cells. Those skilled in the art to which this application pertains will understand that the first end can also be located above the second end, which can also achieve the simultaneous centrifugation and discharge of waste liquid from the centrifuge chamber 500 via the waste discharge channel 406, so that the fluid containing adipose matrix cells remains in the centrifuge chamber 500, which will not be elaborated further here.

[0059] See Figure 13 , Figure 14 This application also provides another design for the centrifuge chamber 500. In one embodiment of this application, the rotation axis of the centrifuge chamber 500 is vertically arranged. The inner cavity of the centrifuge chamber 500 includes a first cavity 503 and a first space 504 radially disposed outside the first cavity 503. The chamber wall of the centrifuge chamber 500 separates the first cavity 503 and the first space 504. Along the direction from the lower end to the upper end of the centrifuge chamber 500, the radial dimension of the first cavity 503 gradually increases. The first space 504 is located in the radial direction of the centrifuge chamber 500, and the inner wall of the space 504, which is furthest from the rotation axis of the centrifuge chamber 500, gradually moves away from the rotation axis of the centrifuge chamber 500. The upper end of the first cavity 503 is connected to the upper end of the first space 504. At the connection between the first cavity 503 and the first space 504, the first residence area 501 is located in the first space 504 and at the upper end of the first space 504, and the second residence area 502 is located in the first cavity 503 and at the upper end of the first cavity 503. The inlet channel 405 is connected to the first cavity 503, and the end point of the fluid path from the receiving unit, the cleaning unit to the centrifugal unit is connected to the first cavity 503; the waste discharge channel 406 extends to the lower end of the first cavity 503, and the starting point of the fluid path from the centrifugal unit to the waste liquid unit is located at the lower end of the first cavity 503; the collection channel 407 extends to the lower end of the first space 504, and the starting point of the fluid path from the centrifugal unit to the collection container is located at the lower end of the first space 504.

[0060] The fluid in the containing unit or the grease-cleaning solution in the cleaning unit can enter the first cavity 503 through pipe 103, the first annular groove 402, and the inlet channel 405 to execute step five or step seven. During step six, because the radial dimension of the first cavity 503 gradually increases in the direction from the lower end to the upper end of the centrifuge chamber 500, when the centrifuge chamber 500 rotates, the fluid containing lipoma cells in the first cavity 503 gradually moves upward under the action of centrifugal force, reaching the upper end of the first cavity 503 and the connection point between the first cavity 503 and the first space 504. Due to the density difference between the lipoma cells and the waste liquid, the lipoma cells enter the first space 504 through the connection point between the first cavity 503 and the first space 504, reaching the first residence area 501, while the waste liquid is located in the second residence area 501 within the first cavity 503. At point 2, after the fluid containing adipose matrix cells and the waste liquid have been stratified radially in the centrifuge chamber 500, the centrifuge chamber 500 stops rotating. Since there is a partition wall between the first cavity 503 and the first space 504, the fluid containing adipose matrix cells falls into the first space 504 under the action of gravity after centrifugation, and the waste liquid falls into the first cavity 503 under the action of gravity. The two will not remix. Finally, the waste liquid flows to the lower end of the first cavity 503. The waste liquid in the first cavity 503 enters the container in the waste liquid unit through the waste discharge channel 406, the second annular groove 403, and the waste liquid branch pipe 107, thereby performing step six above. When performing step seven, the fat cleaning solution in the cleaning unit enters the centrifuge chamber 500 through pipe 3 103, the first annular groove 402, and the inlet channel 405. Subsequently, the centrifuge chamber 500 rotates. Due to the shape of the first cavity 503, the fat cleaning solution moves along the inner wall of the first cavity 503 to the upper end of the first cavity 503. Fat cleaning solution is distributed at the connection between the first cavity 503 and the first space 504. Since the direction from the lower end to the upper end of the centrifuge chamber 500 is along the direction from the lower end to the upper end, the inner wall of the first space 504 that is furthest from the rotation axis of the centrifuge chamber 500 in the radial direction gradually moves away from the rotation axis of the centrifuge chamber 500. Therefore, when the centrifuge chamber 500 rotates, the fluid containing adipose matrix cells in the first space 504 will gradually rise along this inner wall and eventually reach and fill the upper end of the first space 504, contacting the fat cleaning solution located at the upper end of the first cavity 503. The fat cleaning solution can clean the fluid containing adipose matrix cells, thereby performing step seven. When performing step eight, the working principle is similar to that of step six. After the centrifuge chamber 500 stops rotating, the waste liquid flows to the lower end of the first cavity 503 under the action of gravity. Then, under the pumping action of the waste liquid peristaltic pump 202, the waste liquid located at the lower end of the first cavity 503 enters the container in the waste liquid unit through the waste discharge channel 406, the second annular groove 403, and the waste liquid branch pipe 107.When performing step nine, the fluid containing adipose matrix cells located at the lower end of the first space 504 enters the container of the collection unit through the collection channel 407, the third annular groove 404, and the collection pipe 108 extending to the lower end of the first space 504. Then, step ten is performed to complete the collection of adipose matrix cells.

[0061] Adding fat washing solution to the centrifuge chamber 500 to wash the fluid containing adipose matrix cells after centrifugation allows the centrifuge chamber 500 to rotate multiple times and stop circulating in a short period of time, thereby improving the mixing effect between the fluid containing adipose matrix cells and the fat washing solution.

[0062] Those skilled in the art to which this application pertains will understand that the arrangement of the piping units in this application is not limited to the manner described above, but can also be in other ways. For example, the cleaning unit is connected to the processing tank in the containing unit and the centrifuge chamber 500 in the centrifuge unit via a pipe, and each of the two pipes is equipped with a clamping valve and a peristaltic pump; the waste liquid unit is connected to the processing tank in the containing unit and the centrifuge chamber 500 in the centrifuge unit via a pipe, and each of the two pipes is equipped with a clamping valve and a peristaltic pump; the enzyme bottle connecting pipe 112 of the enzyme bottle mounting structure of the enzyme liquid unit is connected to the processing tank in the containing unit via a pipe, and the pipe is equipped with a clamping valve and a peristaltic pump; the centrifuge chamber 500 in the centrifuge unit is connected to the collection container in the collection unit via a pipe, and the pipe is equipped with a clamping valve and a peristaltic pump; the processing tank in the containing unit is connected to the centrifuge chamber 500 in the centrifuge unit via a pipe, and the pipe is equipped with a clamping valve and a peristaltic pump.

[0063] Each clamp valve and each peristaltic pump is electrically connected to the controller, which can control the opening and closing of the clamp valve and the rotation of the peristaltic pump to automatically execute the step of collecting adipose matrix cells.

[0064] There are several ways to drive the centrifuge chamber 500 to rotate, for example, for Figure 12 or Figure 14 The centrifuge chamber 500 is positioned such that its lower end is fixedly connected to the motor shaft by bolts, and the stationary part 400 is fixedly connected to the frame of the main body of the device by bolts, thereby enabling the centrifuge chamber 500 to rotate around the rotation axis. The motor is electrically connected to the controller, and the controller can control the rotation of the motor.

[0065] Furthermore, in one embodiment of this application, the automated adipose tissue stromal cell collection device further includes a liposuction unit connected to a receiving unit. The liposuction unit can be a liposuction machine in the prior art, which includes a liposuction needle, a negative pressure unit, and a collection tank for collecting mixed fat fluid (containing fat, swelling fluid, blood, and tissue fragments). The liposuction needle, the negative pressure unit, and the upper end of the collection tank are connected. When the negative pressure pump in the negative pressure unit is working, the air pressure inside the collection tank decreases, and the mixed fat fluid in the human body is drawn into the collection tank. The bottom of the collection tank is also connected to the processing tank in the receiving unit through a connecting pipe. The connecting pipe is equipped with a peristaltic pump and a clamping valve. When the mixed fat fluid is drawn in by negative pressure, the connecting pipe is closed by the clamping valve. After the mixed fat fluid is collected, the clamping valve on the connecting pipe is opened, and the peristaltic pump is working, so that the mixed fat fluid in the collection tank of the receiving unit enters the processing tank. Subsequently, the clamping valve on the connecting pipe is closed, and the peristaltic pump stops working, thereby automatically completing the above-mentioned step one and reducing the possibility of adipose tissue contamination. The entire process, from the collection of adipose tissue to the production of fluid containing adipose stromal cells, is automated, which helps to improve the collection efficiency of adipose stromal cells and reduce the possibility of contamination of adipose stromal cells.

The technical solutions in each embodiment can also be appropriately combined to form other embodiments that can be understood by those skilled in the art.
