Description

Technical Field

[0001] This invention relates to cosmetic compositions comprising oligopeptides, tetrapeptides, and hexapeptides, said cosmetic compositions for providing enhanced anti-aging benefits. Background Technology

[0002] The skin is the body's primary barrier, protecting internal organs from external stimuli. Skin undergoes both internal and external aging. Skin aging is a complex process, and due to aging, changes in the skin differ from those in other organs. Characteristics of internal or temporal skin aging include dryness, visible free lines and wrinkles, uneven skin pigmentation, loss of elasticity, and sagging. External factors, including exposure to sunlight, pollutants, and cigarette smoke, can accelerate the skin aging process, particularly on the face.

[0003] One of the main causes of skin aging is the degradation of collagen and elastin, which are major components of the extracellular matrix that provides structural support and mechanical strength to the skin. Lumican is a small, leucine-rich proteoglycan involved in the regulation of collagen fibrillation and the organization of the extracellular matrix. Matrix metalloproteinases are proteases capable of dissolving peptide bonds and thus degrading collagen. Upregulating the expression of lumican genes, promoting collagen production, or reducing matrix metalloproteinases typically leads to enhanced anti-aging benefits.

[0004] Therefore, the inventors have recognized the need for further development of solutions to combat aging by upregulating the expression of photoglucan genes, promoting collagen production, and / or reducing matrix metalloproteinases. Surprisingly, it has been found that improved anti-aging benefits can be provided by including oligopeptides, tetrapeptides, and hexapeptides in the cosmetic compositions of the present invention. Summary of the Invention

[0005] In a first aspect, the present invention relates to a cosmetic composition comprising (a) an oligopeptide selected from oligopeptide-1, palmitoyl oligopeptide-1, acetyl oligopeptide-1, myristoyl oligopeptide-1, or combinations thereof; (b) a tetrapeptide selected from tetrapeptide-9, acetyl tetrapeptide-9, palmitoyl tetrapeptide-9, myristoyl tetrapeptide-9, or combinations thereof; and (c) a hexapeptide selected from hexapeptide-11, acetyl hexapeptide-11, palmitoyl hexapeptide-11, myristoyl hexapeptide-11, or combinations thereof.

[0006] In a second aspect, the present invention relates to a method for providing benefits selected from enhancing collagen production in the skin, improving skin elasticity, reducing the appearance of wrinkles, reducing sagging, anti-aging, skin rejuvenation, and upregulating the expression of photoglycan genes, or combinations thereof, the method comprising the step of applying the composition of the invention to a desired surface.

[0007] In a third aspect, the present invention relates to the use of the cosmetic compositions of the present invention for providing benefits selected from enhancing collagen production in the skin, improving skin elasticity, reducing the appearance of wrinkles, reducing sagging, anti-aging, skin restoration, and upregulating the expression of photoglucan genes.

[0008] All other aspects of the invention will become more readily apparent upon consideration of the following detailed description and embodiments. Detailed Implementation

[0045] Example Example 1 This embodiment demonstrates that the peptide combination according to the present invention upregulates the expression of the photoprotein glycan gene.

[0046] The gene expression of optical proteoglycans in different samples was evaluated according to the following procedure. L929 fibroblasts in good growth condition during the logarithmic growth phase were used, with a concentration of 1×10⁻⁶ cells / cells. 4Cells were seeded at a density of 10 cells / well in 48-well plates and incubated overnight at 37°C with 5% CO2. Cells were treated for 24 hours according to Table 1. After treatment, total RNA was extracted from each sample using the RNAprep PurePlant Plus Kit (DP441, Tiangen) according to the manufacturer's instructions.

[0047] Extracted RNA was reverse transcribed to generate template cDNA using the PrimeScript kit. Target genes were amplified using the appropriate primers on an ABI Vii 7 real-time PCR system (Applied Biosystems, Thermo Fisher Scientific, Carlsbad, CA, USA) with Hieff™ qPCRSYBR Green Master Mix. The relative quantification of gene expression was calculated using the comparative Ct method. All samples were first normalized to the housekeeping gene GAPDH, and then normalized again to their corresponding controls to obtain ΔΔCt values. The fold change relative to this control (2^(-ΔΔCt)) was calculated and compared between treatments. All tests were performed at least three times, and Table 1 shows the fold change in gene expression of the optical proteoglycan.

[0048] Table 1

[0049] The level of active ingredient is expressed as a weight percentage based on the amount of culture medium.

[0050] a: WKPep ® Soligopeptide-PLUS, purchased from Winkey Technology in China.

[0051] b: WKPep ® D-tide, purchased from Winkey Technology in China.

[0052] c: WKPep ® Rpeptide, purchased from Winkey Technology in China.

[0053] As demonstrated in Table 1, the fold change of 1 was significantly higher than the product of the fold changes of samples A, B, and C. Therefore, it was unexpected that the combination of oligopeptide-1, acetyl tetrapeptide-9, and hexapeptide-11 significantly upregulated the expression of photoglucan genes, thus providing anti-aging-related benefits.

[0054] Example 2 This embodiment demonstrates the reduction of matrix metalloproteinase 1 (MMP1) through the peptide combination of the present invention.

[0055] The inhibition of MMP1 expression was tested using the following procedure. Human fibroblasts in the exponential growth phase (Kunming Cell Bank, Chinese Academy of Sciences) were taken and digested with 0.25% (w / w) trypsin solution to detach the adherent cells. Cells cultured in PBS were then exposed to UVB (80 mJ / cm²). 2 Irradiate for 15 minutes, then incubate for 48 hours in medium containing or without the active ingredient, according to Table 2. After incubation, MMP1 levels were obtained using the MMP-1 ELISA kit (IEBO, catalog number: E0097h) according to the manufacturer's instructions. The fold reduction was calculated by (Level of MMP1 for control / Level of MMP1 for control) . Each test was repeated three times, and the results are shown in Table 2.

[0056] Table 2

[0057] The level of active ingredient is expressed as a weight percentage based on the amount of culture medium.

[0058] As shown in Table 2, when the three peptides are combined, the reduction fold of MMP1 (Sample 2) is higher than the product of the reduction folds of the three individual peptides (G, H, and I), or the product of the reduction folds of any two peptide combinations and the third individual peptide (J and I, or K and H, or L and G). It was unexpectedly found that the peptide combinations of the present invention provide synergistically better inhibition of MMP1, thus providing anti-aging benefits.
