Description

Technical Field

[0001] This invention relates to novel heterocyclic compounds as inhibitors of Casitas B-cell lymphoma-b (Cbl-b), which can be used to prepare medicaments for treating diseases associated with Cbl-b. These medicaments are particularly useful for the prevention and treatment of cancer. Background Technology

[0002] In recent years, novel therapies aimed at removing suppressive signals from immune cells to enhance the productive immune system have attracted attention. In this regard, Casitas B-cell lymphoma-b (Cbl-b) is a RING-type E3 ubiquitin ligase expressed in various immune cell lineages, where it negatively regulates immune cell activity. Effector T cells are key players in regulating the immune system against a variety of diseases and conditions, including cancer, and compounds that activate effector T cells, such as inhibitors of Cbl-b, have been shown to enhance the secretion and proliferation of T-cell-derived cytokines and exhibit anti-cancer activity. Given the important role of Cbl-b in regulating multiple signal transduction mechanisms in both innate and adaptive immunity, inhibition of Cbl-b offers opportunities for treating various diseases, including cancer immunotherapy.

[0003] This invention relates to the development of low molecular weight compounds required for the effective inhibition of the E3 ubiquitin ligase Cbl-b as described above. Summary of the Invention

[0228] [Experimental Example: Cbl-b Inhibitory Activity]

[0229] The ability to inhibit Cbl-b activity was measured by monitoring the degree of Cbl-b ubiquitination in the presence of candidate compounds.

[0230] In 1% DMSO (final concentration), the concentrations of the example compounds were serially diluted 3-fold from the highest concentration of 1 µM to 0.00005 µM to prepare a total of 10 concentrations. Each concentration of the example compound was pre-incubated with 0.5 nM Cbl-b (BPSBioscience, 80415) in U2 detection buffer (BPS Bioscience, 78856) for 30 min and then measured at room temperature in a 10 µL reaction volume in a 384-well plate (Corning, 3824). After a 30-minute pre-incubation with the compounds of the examples, the reaction was initiated by adding 5 µL of substrate mixture (consisting of 20 nM UBE1 (BPSBioscience, 80301), 50 nM UbcH5b (BPS Bioscience, 80314), 1.5 nM TYRO3 (BPSBioscience, 40293), and 0.25 × Ubi-Mix (BPS Bioscience, 82185)) and 5 µL of 15.5 µM ATP (ThermoFisher, R0441) to each well of the plate. The enzyme reaction was allowed to proceed for 2 hours, and the change in enzyme activity over time was measured every 3 minutes using a SpectraMax i3x plate reader (Molecular Devices) at 615 / 665 nm. After all reactions were completed, the IC50 was calculated from the obtained data values using the following four-parameter logistic curve equation. 50 value.

[0232] In the above equation, "Top" represents the maximum value of the y-axis, and "Bottom" represents the minimum value of the y-axis. Furthermore, "Hillslope" represents the slope of the tangent line at the inflection point of the dose-response curve.
