Abstract
Description
Technical Field
[0001] This application relates to the field of plant active ingredient extraction and separation technology, and more specifically, it relates to a process for simultaneous extraction of bamboo polysaccharides and bamboo flavonoids. Background Technology
[0002] Bamboo is a abundant renewable biomass resource in my country, and its processing byproducts contain a large amount of bioactive bamboo polysaccharides and bamboo flavonoids. Bamboo polysaccharides have various physiological functions such as immunomodulation and antioxidation, and are widely used in food additives and health foods. Bamboo flavonoids have anti-inflammatory, antibacterial, and lipid-lowering effects, and have high development value in the cosmetics and pharmaceutical intermediates industries. Currently, industrial production mostly uses a stepwise extraction process to obtain the two components separately, which has problems such as long process flow, low resource utilization rate, and high production cost. Therefore, developing an efficient simultaneous extraction process for bamboo polysaccharides and bamboo flavonoids has become a key focus of the industry.
[0003] Existing simultaneous extraction processes for bamboo polysaccharides and bamboo flavonoids mostly employ direct extraction using an ethanol-water mixed solvent, with some processes supplemented by physical methods such as ultrasound and microwaves to enhance mass transfer. However, due to the significant differences in the solubility characteristics of bamboo polysaccharides and bamboo flavonoids—bamboo polysaccharides are readily soluble in the aqueous phase, while bamboo flavonoids are more readily soluble in the organic phase—single or simply mixed solvent systems are unlikely to provide the optimal dissolution environment for both components simultaneously. This often results in a decrease in the extraction rate of one component while increasing the extraction rate of the other, making it difficult to effectively improve the extraction efficiency of the total active ingredients. Summary of the Invention
[0004] To address the problem of mutual restriction between the extraction rates of bamboo polysaccharides and bamboo flavonoids and the low overall extraction efficiency in the simultaneous extraction of bamboo polysaccharides and bamboo flavonoids in the prior art, this application provides a process for the simultaneous extraction of bamboo polysaccharides and bamboo flavonoids.
[0005] The simultaneous extraction process of bamboo polysaccharides and bamboo flavonoids provided in this application adopts the following technical solution: A process for simultaneous extraction of bamboo polysaccharides and bamboo flavonoids includes the following steps: S1. Steam Explosion and Endogenous Material Recycling: Bamboo raw materials are subjected to steam explosion treatment, and the condensate and extrusion liquid generated during the explosion process are fully recovered and mixed to form recycled liquid. S2. Preparation of thermosensitive homogeneous medium: At a first temperature below the system's turbidity point, add thermosensitive polymer, phase-forming salt, and inclusion solubilizing agent to the recycled liquid obtained in S1, stir to dissolve, and prepare a homogeneous extraction medium. S3. Low-temperature homogeneous synchronous extraction: The bamboo powder obtained after explosion in S1 is mixed with the homogeneous extraction medium prepared in S2 at a predetermined material-liquid ratio, and the mixture is stirred and extracted for a predetermined time at a second temperature below the turbidity point to obtain an extraction mixture. S4. Temperature-controlled phase separation and solid-liquid separation: The extraction mixture obtained in S3 is heated to a third temperature higher than the turbidity point and allowed to stand, so that the system is separated in situ into an upper polymer-rich phase enriched with flavonoids, a lower salt-rich phase enriched with polysaccharides, and a residue layer embedded and floated by polymer. Then, a one-time solid-liquid separation is performed to collect the upper liquid phase and the lower liquid phase respectively. S5. Product recovery and media recycling: The upper phase liquid collected in S4 is cooled below the turbidity point and restored to homogeneity. Membrane separation is performed to recover flavonoids and retain the polymer solution. The lower phase liquid collected in S4 is concentrated, precipitated with ethanol, and dried to obtain bamboo polysaccharide. The recovered polymer solution and salt solution are recycled for the next batch in step S2.
[0006] By employing the above-mentioned technical solution, steam explosion treatment can break down the dense lignin-cellulose complex structure of bamboo cell walls, fully exposing the active ingredients encapsulated within the cells. Simultaneously, the generated condensate and extrusion liquid contain endogenous organic acids, oligosaccharides, and amino acids. These endogenous substances can reduce the hydration degree of the temperature-sensitive polymer, synergistically enhancing the salting-out effect of subsequently added phase-forming salts to precisely control the cloud point of the temperature-sensitive polymer system without the need for additional acid / base or specialized turbidity-adjusting reagents. In the low-temperature homogeneous state, the system lacks phase interfaces, significantly reducing mass transfer resistance and providing a favorable dissolution environment for both hydrophilic bamboo polysaccharides and hydrophobic bamboo flavonoids. Upon heating, the system undergoes a thermally induced phase transition. Bamboo polysaccharides, due to their hydrophilic properties, remain in the salt-rich phase, while bamboo flavonoids, due to their hydrophobic properties and the mediating effect of inclusion solubilizers, accumulate in the polymer-rich phase, achieving integrated coupling of simultaneous extraction and in-situ separation.
[0007] Preferably, in step S1, the conditions for the steam explosion treatment are: saturated steam pressure of 1.5-2.5 MPa and pressure maintenance time of 60-300 seconds.
[0008] By adopting the above technical solution, this pressure and time range can fully disrupt the cell wall connection structure of bamboo, ensuring the effective release of active ingredients, while avoiding excessive explosion that could lead to the breakage of polysaccharide glycosidic bonds and the oxidative degradation of flavonoid nuclei. Under these conditions, the concentration of endogenous substances released is moderate, which can directly form a matching turbidity-regulating system with the subsequently added phase-forming salt, eliminating the need for concentration adjustment of the recycled solution.
[0009] Preferably, in step S2, the thermosensitive polymer is an ethylene oxide-propylene oxide random copolymer with an average molecular weight range of 2500-4000, and the amount of the thermosensitive polymer added is 8-18 wt% based on the total mass of the homogeneous extraction medium.
[0010] By adopting the above technical solution, the ethylene oxide-propylene oxide random copolymer exhibits reversible low critical dissolution temperature characteristics. Only a physical state transition occurs with temperature changes, without any chemical structural changes, allowing for multiple recycling. This molecular weight range ensures the polymer possesses good water solubility and rapid, reversible phase transition; too small a molecular weight results in insignificant phase transitions, while too large a molecular weight reduces water solubility. Matching the addition amount with the concentration of the phase-forming salt and endogenous substances enables the formation of a stable two-phase system with a clear phase interface and no obvious intermediate emulsion layer.
[0011] Preferably, in step S2, the phase-forming salt is sodium citrate or dipotassium hydrogen phosphate, and its addition amount is 6-14 wt% based on the total mass of the homogeneous extraction medium; the inclusion solubilizing agent is hydroxypropyl-β-cyclodextrin, and its addition amount is 1-4 wt% based on the total mass of the homogeneous extraction medium.
[0012] By employing the above technical solution, sodium citrate and dipotassium hydrogen phosphate, being organic weak acid salts, exhibit good compatibility with endogenous organic acids in the recycled solution. The salting-out process is mild and does not damage the molecular structure of bamboo polysaccharides. Simultaneously, they act as buffers, maintaining pH stability and preventing degradation of active ingredients under acidic or alkaline conditions. Hydroxypropyl-β-cyclodextrin possesses a hydrophobic cavity structure, enabling it to form stable inclusion complexes with hydrophobic flavonoid molecules, significantly improving the solubility of flavonoids in the aqueous phase. Furthermore, it mediates the transfer of flavonoid molecules to the polymer-rich phase, enhancing the separation selectivity of the two active ingredients.
[0013] Preferably, in step S2, the first temperature is 15-25℃; the turbidity point of the homogeneous extraction medium is located in the range of 30-38℃ due to the influence of endogenous substances in the recycled liquid.
[0014] By adopting the above technical solution, the first temperature is lower than the system's cloud point, ensuring complete polymer hydration and forming a homogeneous and transparent solution, providing an environment with low mass transfer resistance for the extraction process. The endogenous substance and the phase-forming salt synergistically regulate the cloud point to this range, ensuring a moderate temperature difference between extraction and phase separation. This guarantees homogeneous stability of the system during extraction while enabling rapid phase separation after heating, achieving near-room temperature phase separation. This avoids damage to the active ingredient structure caused by high-temperature operation and reduces process energy consumption.
[0015] Preferably, in step S3, the predetermined material-liquid ratio is a mass ratio of bamboo powder to homogeneous extraction medium of 1:20-40; the second temperature is 15-25℃; the stirring speed of the stirring extraction is 40-100 rpm; and the predetermined time is 1.5-3 hours.
[0016] By employing the above technical solution, the material-to-liquid ratio ensures that the extraction medium fully wets the bamboo powder particles, allowing the active ingredients to diffuse sufficiently from the cells into the medium. The extraction temperature is maintained below the cloud point to preserve the homogeneity of the system. Low-speed stirring prevents excessive breakage of the bamboo powder particles, avoiding the generation of too much fine residue that could affect subsequent phase separation and solid-liquid separation. Simultaneously, without causing degradation of the active ingredients, it enhances the mass transfer process between the solid and liquid phases, thereby improving extraction efficiency.
[0017] Preferably, in step S4, the third temperature is 35-50℃; the settling time is 30-60 minutes; and the one-time solid-liquid separation is completed using plate and frame filtration or a disc centrifuge.
[0018] By employing the above technical solution, the third temperature, higher than the system's cloud point, triggers a phase change and dehydration of the temperature-sensitive polymer, forming hydrophobic aggregates. The settling time ensures sufficient stratification of the two phases, while simultaneously allowing the polymer aggregates to fully adsorb and encapsulate the fine bamboo powder residue. Since the aggregate density is less than that of the aqueous phase, it floats on the upper layer of the system. Plate and frame filters and disc centrifuges are commonly used solid-liquid separation equipment in industrial applications, capable of separating the residue from the two liquid phases in a single step, eliminating the need for prior filtration and phase separation, thus simplifying the process.
[0019] Preferably, in step S5, the processing step of the upper phase liquid is as follows: the upper phase liquid is cooled to 15-20℃ to restore it to homogeneity, separated by an ultrafiltration membrane with a molecular weight cutoff of 3000-5000, the permeate is collected and dried to obtain bamboo flavonoid product, and the retained polymer solution is collected.
[0020] By employing the above technical solution, cooling to this temperature range allows the polymer to rehydrate, restoring the system to a homogeneous state. The polymer molecules are uniformly dispersed in the solution, preventing clogging of the ultrafiltration membrane and improving membrane separation efficiency and lifespan. The ultrafiltration membrane's molecular weight cutoff matches the molecular sizes of the polymer and flavonoids, efficiently retaining polymer molecules while allowing flavonoid molecules to permeate, achieving rapid separation. The retained polymer solution can be directly reused without additional treatment.
[0021] Preferably, in step S5, the treatment step of the lower phase liquid is as follows: vacuum concentrate to 1 / 5-1 / 8 of the original volume at 60-70℃, add 3-5 times the volume of anhydrous ethanol for alcohol precipitation for 2-4 hours, centrifuge to collect the precipitate, and obtain bamboo polysaccharide by low temperature vacuum drying.
[0022] By employing the above technical solutions, vacuum concentration can remove most of the water from the lower phase liquid at a lower temperature, avoiding the breakage of glycosidic bonds in polysaccharides caused by high temperatures. During alcohol precipitation, slow addition of anhydrous ethanol with gentle stirring prevents the formation of a gel-like precipitate and reduces impurity encapsulation. This alcohol precipitation ratio and time ensure sufficient polysaccharide precipitation while minimizing co-precipitation of impurities. Low-temperature vacuum drying removes residual water from the precipitate while preserving the natural structure of the polysaccharide, yielding bamboo polysaccharide products.
[0023] Preferably, in step S5, the medium circulation includes: distilling the supernatant obtained after centrifugation to collect the precipitate to recover ethanol, and then regenerating the residual salt and cyclodextrin aqueous solution by nanofiltration desalting, which together with the recovered polymer solution are used as the circulating medium, and are reused after being supplemented with new components in proportion in the next batch of step S2.
[0024] By adopting the above technical solution, ethanol used in the alcohol precipitation process can be recovered by distillation, realizing the recycling of ethanol. Nanofiltration desalination can remove small molecule impurities accumulated during the recycling process, avoiding the impact of impurities on subsequent extraction and phase separation effects. After the regenerated components are mixed with the recovered polymer solution, a small amount of new components can be added to make up for the small losses of each component during the recycling process, maintain the extraction and phase separation performance of the system, significantly reduce the consumption of chemical reagents, and reduce the amount of waste emissions from the process.
[0025] In summary, this application has the following beneficial effects: 1. This application adopts a combination of steam explosion endogenous material full reuse and temperature-sensitive homogeneous medium system. It utilizes the endogenous material in the reused liquid and the phase-forming salt to synergistically regulate the turbidity point without the need to add additional turbidity adjusting reagents. At the same time, it realizes low-temperature homogeneous low-resistivity extraction and in-situ selective phase separation by heating, which solves the problem of mutual restriction between the extraction rates of the two active ingredients and significantly improves the efficiency of simultaneous extraction.
[0026] 2. The method of this application achieves extraction, phase separation and solid-liquid separation in one step by designing to embed floating solid residues during the phase change of temperature-sensitive polymers. This reduces the loss of active ingredients caused by multi-step separation operations, simplifies the process flow, shortens the production cycle, and reduces energy and material consumption in the separation process.
[0027] 3. In this application, a closed-loop circulation design of the extraction medium is preferred, in which the recovered polymer solution and the regenerated salt and cyclodextrin aqueous solution are used together as the circulation medium. Only a small amount of new components need to be added to maintain the stability of the system, which greatly reduces the consumption of chemical reagents, reduces production costs, and reduces the emission of process waste.
[0028] 4. Since this application uses near-room temperature conditions below 50°C to complete the entire extraction and phase separation process, it avoids the damage to polysaccharide glycosidic bonds and flavonoid core structure caused by the high temperature and high pressure environment of traditional processes, effectively preserving the natural structure and bioactivity of active ingredients and improving the intrinsic quality of the final product.
[0029] 5. The method of this application, by setting a wide range of process parameters, is suitable for bamboo raw materials of different types and parts, and adopts standardized membrane separation, vacuum concentration and drying technology to ensure stable product performance between batches. The process operation is simple and controllable, and it has good potential for industrial scale-up production. Attached Figure Description
[0030] Figure 1 This is a flowchart of a simultaneous extraction process of bamboo polysaccharides and bamboo flavonoids provided in this application. Detailed Implementation
[0031] The present application will be further described in detail below with reference to embodiments and comparative examples. Unless otherwise specified, the experimental methods used below are conventional methods. Unless otherwise specified, the materials, reagents, methods and instruments used are all conventional materials, reagents, methods and instruments in the art, which can be obtained by those skilled in the art through commercial channels or prepared according to literature methods.
[0032] Technical Concept: Bamboo polysaccharides and bamboo flavonoids, as high-value-added active ingredients in bamboo, have broad application prospects in the food, pharmaceutical, and cosmetic fields. Simultaneous extraction of these two components is a key direction for improving the utilization rate of bamboo resources. Existing simultaneous extraction processes generally suffer from mutual constraints on the extraction rates of the two components. The fundamental reason lies in the significant differences in the solubility characteristics of bamboo polysaccharides and bamboo flavonoids; a single or simply mixed solvent system cannot simultaneously provide the optimal dissolution environment for both. Furthermore, existing processes often employ a sequential model of extraction followed by separation, and these multi-step operations not only prolong the production cycle but also lead to significant losses of active ingredients during transfer. Some aqueous two-phase extraction processes require the addition of acids, alkalis, or special turbidity-adjusting reagents, which not only increases production costs but may also introduce impurities that affect the safety of the final product.
[0033] This technical solution addresses the aforementioned core issues by first employing steam explosion pretreatment to break up the dense cell wall structure of bamboo. Simultaneously, it fully recovers the condensate and extrusion liquid generated during the explosion process, using them as the extraction medium substrate. The endogenous organic acids and oligosaccharides contained within these substances synergistically interact with the phase-forming salts to precisely control the cloud point of the temperature-sensitive polymer system, eliminating the need for additional turbidity-adjusting reagents. Secondly, a temperature-sensitive homogeneous extraction medium system is constructed, achieving interface-free, low-resistivity simultaneous extraction at temperatures below the cloud point. Heating triggers a thermally induced phase transition, causing bamboo polysaccharides and bamboo flavonoids to accumulate in the two phases based on their different solubility properties. Simultaneously, the temperature-sensitive polymer's ability to encapsulate and float fine residues during the phase transition enables extraction, phase separation, and solid-liquid separation to be completed in one step. Finally, a closed-loop circulation design of the entire component medium recovers and regenerates the polymers, salts, and ethanol used in the extraction process, significantly reducing chemical reagent consumption and lowering the environmental impact and production costs.

The specific steps for processing the upper phase liquid are as follows: the upper phase liquid is cooled to 20°C to restore it to a homogeneous state, and then separated using an ultrafiltration membrane with a molecular weight cutoff of 5000. The ultrafiltration pressure is 0.25 MPa, the ultrafiltration temperature is 25°C, the permeate is collected and spray-dried with an inlet air temperature of 170°C, an outlet air temperature of 85°C, and a drying time of 1.5 hours to obtain the bamboo flavonoid product. The retained polymer solution is collected, sealed, and stored to prevent degradation. The specific steps for processing the lower phase liquid are as follows: The liquid is vacuum concentrated to 1 / 5 of its original volume at 70℃ (vacuum degree: -0.09 MPa) for 1 hour; five times the volume of anhydrous ethanol is added for alcohol precipitation for 4 hours at 6℃; the precipitate is collected by centrifugation at 6000 rpm for 20 minutes; the precipitate is washed three times with anhydrous ethanol, each time using three times the mass of the precipitate, and then dried under low-temperature vacuum at 55℃ for 2 hours to obtain bamboo polysaccharides. The media recycling process includes: recovering ethanol from the supernatant obtained after centrifugation and collection of precipitate by vacuum distillation at a temperature of 85°C and a pressure of 0.04 MPa, with the recovered ethanol having a purity of ≥98%, which can be recycled for the alcohol precipitation step; regenerating the residual salt and cyclodextrin aqueous solution by nanofiltration desalting, with the nanofiltration membrane having a molecular weight cutoff of 200 Da and a nanofiltration pressure of 0.5 MPa; the regenerated aqueous solution and the recovered polymer solution are used together as the recycling medium, and are reused after being supplemented with new components at a ratio of 8 wt% in the next batch of step S2.
[0049] Comparative Example 1: The only difference between this comparative example and Example 1 is that the recycled liquid formed by mixing the condensate and the extrusion liquid in S1 is replaced with an equal volume of deionized water. An additional 0.5 wt% citric acid is added to the deionized water to simulate the turbidity adjustment effect of the endogenous organic acid in the recycled liquid. The other raw material ratios, process steps and all parameters are completely consistent with Example 1.
[0050] Comparative Example 2: The only difference between this comparative example and Example 1 is that the inclusion solubilizing agent hydroxypropyl-β-cyclodextrin is removed in S2. The remaining raw material ratios, process steps and all parameters are completely consistent with Example 1.
[0051] Comparative Example 3: The only difference between this comparative example and Example 1 is that the phase-forming salt in S2 is replaced with an equal mass of ammonium sulfate, and the amount added is still 10 wt%. The other raw material ratios, process steps and all parameters are completely consistent with Example 1.
[0052] Comparative Example 4: The only difference between this comparative example and Example 1 is that the second temperature for low-temperature homogeneous synchronous extraction in S3 is set to 40°C, which is higher than the system's turbidity point of 34°C. The remaining process steps, raw material ratios, and all parameters are completely consistent with Example 1.
[0053] Comparative Example 5: The only difference between this comparative example and Example 1 is that: no media circulation is performed in S5, and the retained polymer solution, the salt after nanofiltration regeneration, and the cyclodextrin aqueous solution are directly discarded. Freshly prepared extraction media are used in each S2 step. The remaining process steps, raw material ratios, and all parameters are completely consistent with Example 1.
[0054] Comparative Example 6: The only differences between this comparative example and Example 1 are as follows: the thermosensitive polymer, phase-forming salt, and inclusion solubilizer are removed in S2, and the extraction medium is replaced with a 70 vol% ethanol aqueous solution; in S3, the blasted bamboo powder and the 70 vol% ethanol aqueous solution are mixed at a mass ratio of 1:30 and refluxed at 80°C for 2.25 hours; the temperature-controlled phase separation step in S4 is removed, and the extract is filtered and the ethanol is recovered by vacuum distillation. The concentrate is then precipitated with alcohol and dried to obtain a mixed product of bamboo polysaccharides and bamboo flavonoids; media circulation is not performed in S5; the remaining process steps and parameters are the same as in Example 1.
[0055] Test Item 1: Extraction rate test of bamboo polysaccharides and bamboo flavonoids. This test was performed in accordance with GB / T40632-2021 "Determination Method of Polysaccharides in Bamboo Leaves" and SN / T4592-2016 "Determination of Total Flavonoids in Exported Food". Bamboo polysaccharide products and bamboo flavonoid products prepared in Examples 1-3 and Comparative Examples 1-6, as well as bamboo raw material powder processed in the corresponding process, were accurately weighed as follows: 5.00g bamboo raw material powder, 2.00g bamboo polysaccharide product, and 1.00g bamboo flavonoid product. The bamboo raw material powder was pulverized, defatted, and dried for later use. The bamboo polysaccharide product and bamboo flavonoid product were dissolved in deionized water and diluted to 100mL volumetric flasks. Bamboo polysaccharide content was determined using the phenol-sulfuric acid method. 1 mL of sample solution was placed in a test tube, 1 mL of 5% phenol solution was added and shaken well, followed by 5 mL of concentrated sulfuric acid. After standing for 15 minutes, the absorbance was measured at 490 nm. The bamboo polysaccharide content was calculated based on the standard curve. Bamboo flavonoid content was determined using the aluminum salt colorimetric method. 2 mL of sample solution was placed in a test tube, 1 mL of 10% aluminum trichloride solution and 1 mL of 1 mol / L potassium acetate solution were added, and the volume was adjusted to 10 mL with deionized water. After standing for 30 minutes, the absorbance was measured at 415 nm. The bamboo flavonoid content was calculated based on the standard curve. The extraction rate was calculated using the formula: Extraction rate = (Mass of target substance in the product ÷ Mass of target substance in the bamboo raw material) × 100%. Each sample was measured in triplicate, and the average value was taken as the final extraction rate. The differences in bamboo polysaccharide and bamboo flavonoid extraction rates between the various examples and comparative examples were compared to verify the extraction efficiency advantage of the proposed process.
[0056] Test Item 2: Purity Test of Bamboo Polysaccharide and Bamboo Flavonoid Products. This test was performed in accordance with GB / T31740-2015 "Food Additives - Bamboo Polysaccharide" and GB / T42114-2022 "Determination of Flavonoids in Plants by High Performance Liquid Chromatography". 1.00 g of bamboo polysaccharide and bamboo flavonoid products prepared in each example and comparative example of Test Item 1 were accurately weighed. The bamboo polysaccharide product was dissolved in deionized water and diluted to 50 mL. The purity was determined by high performance liquid chromatography. The chromatographic conditions were as follows: a 4.6 mm × 250 mm, 5 μm amino column; a mobile phase of acetonitrile-water (70:30 v / v); a flow rate of 1.0 mL / min; a column temperature of 30 °C; a detection wavelength of 210 nm; and an injection volume of 20 μL. The purity of bamboo polysaccharides was calculated using the peak area normalization method. Bamboo flavonoids were dissolved in methanol and diluted to 50 mL; their purity was also determined using high-performance liquid chromatography (HPLC). The chromatographic conditions were as follows: a 4.6 mm × 250 mm, 5 μm C18 column; a mobile phase of methanol-0.1% phosphoric acid aqueous solution (v / v ratio 55:45); a flow rate of 1.0 mL / min; a column temperature of 35 °C; a detection wavelength of 270 nm; and an injection volume of 20 μL. The purity of bamboo flavonoids was calculated using the peak area normalization method. Each sample was measured in triplicate, and the average value was taken as the final purity. The purity differences between the examples and the comparative examples were compared, along with the extraction rate test results.
[0057] Test Item 3: Extraction Media Cycling Stability Test. This test was conducted in accordance with HJ / T336-2006 "Clean Production Evaluation Index System for Agricultural Product Processing" and GB / T16632-2008 "Performance Evaluation Method for Circulating Cooling Water Treatment Agents". Five batches of simultaneous extraction experiments of bamboo polysaccharides and bamboo flavonoids were repeated according to the process conditions of Examples 1-3 and Comparative Examples 1-6. For each batch, the extraction rate and product purity of bamboo polysaccharides and bamboo flavonoids were determined according to the methods of Test Items 1 and 2. The amount of extraction media replenished in step S2 of each batch and the appearance of the media after circulation, including homogeneity and the presence or absence of precipitation, were also recorded. After the experiment, the coefficient of variation of the extraction rate and purity of the five batches was calculated. The smaller the coefficient of variation, the better the media cycling stability. The differences in media replenishment amount and product performance stability between the examples and comparative examples were also compared.
[0058] Table 1: Results of bamboo polysaccharide and bamboo flavonoid extraction rates in each example and comparative example
[0059] Table 2: Purity test results of bamboo polysaccharide and bamboo flavonoid products in each example and comparative example
[0060] Table 3: Cyclic stability test results of media in each embodiment and comparative example
[0061] Note: The coefficient of variation in Table 3 is used to reflect the fluctuation of extraction rate and purity in the 5 batches of repeated experiments. The smaller the value, the better the process stability. The proportion of medium replenishment in a single batch is the proportion of the mass of new medium to be replenished when each batch is recycled to the total mass of medium. The smaller the value, the higher the medium recycling rate. In Examples 1-3, the system remained uniform and transparent throughout the 5 batches of recycling, without layering or precipitation. In Comparative Example 1, trace amounts of flocculent matter appeared in the 3rd batch and slight turbidity appeared in the 5th batch. In Comparative Example 2, there was no obvious precipitation throughout the process, but the stability of flavonoid dissolution continued to decline. In Comparative Example 3, interface layering and poor uniformity appeared in the 4th batch. In Comparative Example 4, the uniformity of the system decreased from the 2nd batch onwards, and the batch fluctuations were obvious. In Comparative Examples 5 and 6, no medium recycling was performed, and all extraction media were replaced with freshly prepared media in each batch.
[0062] Based on Examples 1-3 and Comparative Example 1, and in conjunction with Tables 1-3, it can be seen that the recycled liquid generated during the steam explosion process and its contained endogenous substances have an important synergistic effect on the extraction and separation of bamboo polysaccharides and bamboo flavonoids. Simply using deionized water with citric acid to simulate the turbidity adjustment effect of endogenous organic acids cannot replace the comprehensive effect of multiple endogenous substances in the recycled liquid, thereby affecting the extraction and separation performance of the entire process and also leading to a decrease in stability during the media circulation process.
[0063] Based on Examples 1-3 and Comparative Example 2, and in conjunction with Tables 1-3, it can be seen that the inclusion solubilizing agent plays a key mediating role in the simultaneous extraction of bamboo polysaccharides and bamboo flavonoids. It can promote the dissolution and phase distribution of flavonoids. Removing this agent will destroy the solubilizing effect of the system on flavonoids, affecting the simultaneous extraction efficiency and separation purity of the two active ingredients. It will also cause fluctuations in the stability of flavonoid dissolution during the medium circulation process.
[0064] Based on Examples 1-3 and Comparative Example 3, and in conjunction with Tables 1-3, it can be seen that the selection of phase-forming salts has a certain synergistic compatibility with the endogenous substances in the recycled liquid. A suitable phase-forming salt can assist the thermosensitive polymer in forming a stable homogeneous extraction system and promote the phase separation of active ingredients. Changing the type of phase-forming salt will reduce the synergistic effect with the endogenous substances, affect the homogeneity and phase separation efficiency of the system, and thus affect the extraction rate and product purity, as well as weaken the stability of the medium circulation.
[0065] Based on Examples 1-3 and Comparative Example 4, and in conjunction with Tables 1-3, it can be seen that the matching of the temperature and the cloud point of the system during low-temperature homogeneous synchronous extraction is the key to ensuring extraction efficiency and system stability. Extraction at a temperature higher than the cloud point of the system will disrupt the homogeneous extraction environment, increase mass transfer resistance, and hinder the full dissolution of bamboo polysaccharides and bamboo flavonoids. It will also lead to significant fluctuations in extraction efficiency and product purity between batches, affecting the system stability during the media circulation process.
[0066] As can be seen from Examples 1-3 and Comparative Example 5, and Tables 1-3, the closed-loop circulation design of the extraction medium can maintain the stability of the process system, reduce material consumption, and the recycled medium can maintain the ability to extract and separate active ingredients. Without medium circulation, even if the single extraction effect is close to the normal process, it will lead to increased fluctuations in extraction rate and purity between batches, while significantly increasing material consumption and reducing the economy and stability of the process.
[0067] As can be seen from Examples 1-3 and Comparative Example 6, and Tables 1-3, the temperature-sensitive homogeneous medium composed of temperature-sensitive polymers, phase-forming salts, and inclusion solubilizing agents, as well as the process design of temperature-controlled phase separation and endogenous material recycling, are the core of achieving simultaneous and efficient extraction and separation of bamboo polysaccharides and bamboo flavonoids. Traditional alcohol-water reflux extraction processes lack these core designs, cannot achieve selective separation and efficient enrichment of the two active ingredients, and do not have the ability to circulate the medium, thus affecting the extraction efficiency, product purity, and stability of the process.
