Abstract
The present invention relates to a microfluidic method for preparing a pharmaceutical composition that contains exosomes, wherein the exosomes contain lipophilic and / or hydrophilic active substances. The method comprises the following steps in the following order: (a) extracting lipids from a cell line of human normal embryonic lung fibroblasts (MRC-5 cell line) to obtain a lipid film. (b) resuspending the lipid film in an organic solvent and mixing with the aqueous phase within a microfluidic device to obtain exosomes, where the ratio of the organic phase flow?rate to the aqueous phase flowrate is set from 1:5 to 1:3; the organic phase flow rate is preferably 200΅L / min or 400΅L / min, and the aqueous phase flow rate is preferably 100΅L / min or 1000΅L / min; (c) purifying the obtained exosomes. (d) loading of the exosomes with the active substance. (e) purifying the obtained exosomes to remove unloaded drug.
