Abstract

This invention concerns the design of specific primers for the OBP2 gene DNA in cotton aphids (Aphis gossypii). The primer design in this invention was designed using the Primer3Plus program. Based on the reference DNA sequence of chromosome 2 Haplotype 1 of A. gossypii (GenBank: NC_065531.1) from the NCBI database. Several primer designs were obtained: AgosOBP2_E1 F: 5'- ACA TAA GAA CAC ACT CAC GG -3' and AgosOBP2_E1 R: 5'- ACA AAA TTG CAC TAT ACT CAG T -3' for detection of exon 1 with a target DNA length of 348 bp, AgosOBP2_E2 F: 5'- TCA TTT CAC CCC GTC GTT TCA -3' and AgosOBP2_E2 R: 5'- TCT GAC AAT CTA CTC GGT CTA TGT -3' for detection of exon 2 with a target DNA length of 502 bp, AgosOBP2_E3 F: 5'- AGG GGG TGG CTT ATA ACT G -3' and AgosOBP2_E3 R: 5'- TCT CCG TTT TTA ACA AAG CAG 3' for detection of exon 3 with target DNA length of 677 bp, as well as AgosOBP2_E4-5 F: 5'- GAT GGC TAA GAT ACG TTT CG -3' and AgosOBP2_E4-5 R: 5'- TGT TCA AAC TCA TAC TTT CCG -3' for exons 4 and 5 with a target DNA length of 408 bp.The material used was A. gossypii from chili plants. The DNA amplification achieved met the target and the chromatogram results were good.
