Abstract
This invention concerns a method for isolating a bioactive compound from the marine sponge Aaptos lobata, aaptamine, with a high degree of purity (>98%). The purification stage begins with extraction using MeOH, followed by liquid-liquid separation (between EtOAc and H2O), then the water fraction is further purified using chromatography techniques. The chromatography technique used to obtain aaptamine is by using High Performance Liquid Chromatography (HPLC) using a C18 stationary phase (Cosmosil C18, 4.6 x 250 mm) using a mixture of MeOH and H2O mobile phases and a flow rate of 1 ml / min. In the HPLC chromatogram, the aaptamine compound was obtained at a retention time (RT) of 5.53 min with a purity level of >98%. This level of purity was proven by using the NMR spectrum of the aaptamine isolate obtained which showed characteristic peaks for aaptamine without any impurity peaks.
