Abstract

This invention aims to produce a persistent drug-tolerant U87MG glioblastoma cell model for early monitoring of adaptive resistance to temozolomide chemotherapy and a method for its production. This invention relates to molecular oncology and pharmacology, specifically the development of a cancer cell model that is tolerant to chemotherapy drugs for use in research on therapeutic resistance mechanisms, drug screening, and cancer therapy development. This invention was obtained through the following stages: 1) culturing U87MG cells and 2) exposing U87MG cells to repeated TMZ with graded concentrations. To assess the results of cell culture and TMZ exposure, cell morphology observations, cell viability tests, cell proliferation tests to determine the doubling time of the cell population, and spheroid formation tests were carried out.U87MG-T67 cells produced from this invention have 80-100% viability and cell proliferation that is slowed by PDT more than 1.5 times compared to U87MG cells, which indicates that they are persistent drug-tolerant cells to TMZ. This invention has morphological characteristics of a population of cells that are stacked on top of each other, have a dilated and branched cytoplasm with interconnected cellular fibers forming a network. The spheroid formation ability in this invention is >1.4 times compared to parental U87MG cells which indicates an increase in stemness character.
