AI Technical Summary

Technical Problem

Current anti-cancer therapies are inadequate for targeting cancers dependent on Myt1 activity, particularly those with CCNE1 amplification or FBXW7 mutations, as they fail to effectively inhibit cell proliferation and induce cell death.

Method used

A combination therapy using Myt1 inhibitors alongside other kinase inhibitors (e.g., WEE1, FEN1, TOP1, RRM1, RRM2, AURKB, TOP2A, ATR, TTK, SOD1, SOD2, BUB1, CDC7, SAE1, PLK1, UBA2, HDAC3, CHEK1, AURKA, MEN1, DOT1L, CREBBP, EZH2, PLK4, HASPIN, METTL3) and platinum-based DNA damaging agents or nucleoside analogs to treat cancers with CCNE1 overexpression or FBXW7 mutations.

Benefits of technology

The combination therapy synergistically induces cell death in cancer cells, effectively targeting and inhibiting cell proliferation in cancers with CCNE1 overexpression or FBXW7 mutations, enhancing treatment efficacy.

Abstract

The use of inhibitors of tyrosine and threonine specific cdc2 inhibitory kinase (Myt1) in the treatment of cancer is disclosed. In a preferred embodiment, the Myt1 inhibitor is a carboxamidopyrrolopyrazine or carboxamidopyrrolopyridine of formula I. The Myt1 inhibitor can be used in combination with various other anti-cancer agents. Such agents include WEE1 inhibitors, TOP1 or TOP2A inhibitors, RRM1 or RRM2 inhibitors, AURKA or AURKB inhibitors, ATR inhibitors, TTK inhibitors, SOD1 or SOD2 inhibitors, BUB1 inhibitors, CDC7 inhibitors, SAE1 inhibitors, PLK1 inhibitors, UBA2 inhibitors, DUT inhibitors, HDAC3 inhibitors, CHEK1 inhibitors, MEN1 inhibitors, DOT1L inhibitors, CREBBP inhibitors, EZH2 inhibitors, PLK4 inhibitors, HASPIN inhibitors, METTL3 inhibitors, nucleoside analogs, and platinum-based DNA alkylating agents.
