Description

Detailed Description of the Invention

[0001] This application claims priority to a prior application, filed on May 14, 2021 with the National Intellectual Property Office of the People's Republic of China, bearing application number 202110533087.9 and entitled "Acid addition salts and crystalline forms, compositions and medicinal uses of salts of ROCK inhibitors," and a prior application, filed on May 7, 2022 with the National Intellectual Property Office of the People's Republic of China, bearing application number 202210500178.7 and entitled "Acid addition salts and crystalline forms, compositions and medicinal uses of salts of ROCK inhibitors," both of which are incorporated herein by reference in their entirety. [Technical field] The present invention relates to the field of medicinal chemistry, specifically to acid addition salts and crystalline forms, compositions and medicinal uses of ROCK inhibitors. [Background technology] Idiopathic interstitial pulmonary fibrosis (IPF) is a chronic, diffuse interstitial lung disease of unknown etiology that has changed from usual interstitial pneumonia, and its histopathological and imaging findings often show usual interstitial pneumonia. Because of its complex pathogenesis and irreversible progression of the disease, early diagnosis is difficult, and once diagnosed, the survival rate of patients decreases significantly over time, with the 3-year survival rate being 50% and the 5-year survival rate being only 20%, which is lower than the survival rate of many cancers (e.g., leukemia, breast cancer, colon cancer, uterine tumors, kidney cancer, etc.), and it is said to be a "cancer that is not cancer." Currently, there is no reliably effective treatment for IPF. According to the results of recent randomized controlled clinical trials, drugs such as pirfenidone and nintedanib can be used appropriately in accordance with the actual clinical conditions in China. However, treatment with nintedanib is only recommended for IPF patients with mild to moderate pulmonary dysfunction. Further study is needed to determine whether nintedanib is effective in treating IPF patients with severe pulmonary dysfunction, and the treatment course of drug administration.

[0002] Rho GTPases, discovered in 1985, belong to the Ras superfamily and share 25% homology with Ras. The main members of Rho GTPases currently found in mammalian histiocytes are Rho (A, B, C), Rac (1, 2, 3), Cdc42 (Cdc42Hs / G25K, TC10, Tcl), Rho D, Rho G, Chp (1, 2), Rnd (Rho E / Rnd3, Rnd1 / Rho6, Rnd2 / Rho7), Rho H / TTF, Rif, Wrch1 and Rho BTB (1, 2), of which Rho (A, B, C) is one of the most prominent members of Rho GTPases. ROCK (Rho-associated protein kinase), also known as Rho kinase, belongs to the serine / threonine protein kinase family, has a molecular weight of approximately 160 kD, and is the downstream target effector molecule of Rho whose function has been most thoroughly investigated to date. ROCK includes isoforms ROCK1 (ROKβ, p160-ROCK) and ROCK2 (ROKα). The two isoforms share 65% amino acid sequence identity and high similarity in the kinase domain (92% identical). ROCK is distributed throughout the body, and ROCK1 is more highly expressed in non-neuronal tissues (blood, small intestine, thymus, etc.), whereas ROCK2 is more highly expressed in the brain, heart, and colon.

[0003] ROCK is involved in the development of multiple cardiovascular and cerebrovascular diseases, including hypertension, atherosclerosis, ischemic stroke, heart disease, diabetic nephropathy, eye diseases, tumors, neuropathic diseases, radiation injury, and autoimmune diseases. For example, the Rho / ROCK signaling pathway is involved in the activation of NAD(P)H oxidase, which induces oxidative stress, cardiac microvascular injury, and C-reactive protein-induced atherosclerotic thrombosis; high glucose can activate the Rho / ROCK pathway, induce the expression of visfatin and type I procollagen in cardiac fibroblasts, and cause cardiac fibroblast hyperproliferation to induce diabetic cardiomyopathy; activation of the Rho / ROCK signaling pathway can regulate the NF-κB signaling pathway and upregulate inflammatory genes to induce the development of diabetic nephropathy; the Rho / ROCK signaling pathway affects the metastasis of cancer cells by changing the permeability of biological membranes; when the spinal cord is injured, activation of Rho induces growth cone atrophy, causing axon regeneration impairment, and induces the inhibitory effect of chondroitin sulfate proteoglycans on neuronal growth.

[0004] In addition, the Rho / ROCK signaling pathway is also involved in the occurrence and development of fibrosis. Activation of the Rho / ROCK signaling pathway can improve the degree of ischemic myocardial fibrosis, and the expression of Rho and ROCK in cardiac tissues of rats with acute myocardial fibrosis is obviously elevated. Activation of the Rho / ROCK signaling pathway can induce actin phosphorylation and cause cell fibrosis. Both in vivo and in vitro experimental results demonstrate that the physiological and pathological damage of the cardiopulmonary caused by long-term radiation exposure is linked to fibrosis induced by the Rho / ROCK pathway. The formation of endothelial adhesion fibronectin and focal adhesion, reduction of endothelial cell migration, and endothelial dysfunction caused by ionizing radiation are linked to the reorganization of actin cytoskeleton and the formation of stress fibers induced by the activation of the Rho / ROCK signaling pathway.

[0005] Lung injury in IPF primarily targets alveolar epithelial cells (ACEs), whose death triggers wound-healing responses, including activation of innate immunity, vascular leakage and extravascular blood coagulation, fibroblast recruitment, proliferation and activation, extracellular matrix synthesis and cross-linking, alveolar collapse and epithelial cell regeneration. ROCK signaling can fundamentally regulate the movement of these cells, especially epithelial cells, endothelial cells and fibroblasts, involved in the healing response. The pivotal role of ROCK in these responses further suggests the possibility of treating pulmonary fibrosis with ROCK inhibitors.

[0006] At present, drugs that treat many diseases, including fibrosis, via the ROCK inhibitory pathway are not yet commercially available. The development of new drugs requires the chemical and biological properties of the lead compound to be well optimized. In addition, the compound must have the desired pharmacokinetic and pharmacodynamic characteristics. The laborious development process usually requires extensive testing. The process of determining the optimal compound often requires the preparation of thousands of structurally similar compounds. Therefore, improving ROCK kinase inhibitors and developing new scaffold compounds with ROCK1 and / or ROCK2 kinase inhibitory activity is of positive significance to the treatment of the above diseases. At the same time, the development of solid drug forms of these compounds suitable for pharmaceutical use, such as solid forms with improved stability, hygroscopicity and / or efficacy, in order to achieve good effects in the stages of drug production and drug use, is a technical problem that must be urgently solved. [0200] 1 H NMR (400 MHz, DMSO-d6) δ 11.69 (s, 1H), 7.61 (d, J = 12.9 Hz, 1H), 7.46 (s, 1H), 7.31 (dd, J = 11.5, 6.1 Hz, 2H), 7.24 (t, J = 8.0 Hz, 1H), 6.93 - 6.87 (m, 2H), 6.80 (dd, J = 7.3, 1.9 Hz, 1H), 4.59 (s, 2H), 4.31 (d, J = 5.8 Hz, 2H), 3.99 (t, J = 8.7 Hz, 2H), 3.74 (s, 3H), 3.12 (t, J = 8.5 Hz, 2H). Example 1: Preparation of the hydrochloride salt An appropriate amount (20 mg to 30 mg) of compound A was weighed out and placed in a sample bottle at room temperature. Then, 0.2 mL of different solvents were added as shown in Table 5, and 1 M hydrochloric acid was added to form salts, followed by stirring at room temperature for 3 to 6 hours. If no precipitate was formed, a poor solvent (MTBE) was added to promote the precipitation of precipitates. If solids were present, samples were collected by filtration, dried under vacuum at 50 °C for about 3 hours, and analyzed by XRPD, TGA, DSC, and 1 H-NMR characterization was performed. The specific information of the raw materials and the results are shown in Table 5. The XRPD pattern and analysis of the hydrochloride salt crystalline form I prepared in Group 1 are shown in Figure 1 and Table 5'.

[0201] [Table 7]

[0202] [Table 8]

[0203] In this example, a hydrochloride crystalline form designated as hydrochloride crystalline form I was obtained. 1 The H-NMR and IC characterization results are summarized in Table 6 and Figures 2 and 3.

[0204] The hydrochloride salt crystalline form I had a weight loss of about 5.4% up to 110°C. 1 H-NMR analysis showed that the sample had no residual organic solvents, and the weight loss in the TGA was attributed to dehydration (approximately 1 equivalent of HO). The DSC pattern showed multiple heavy thermodynamic events, including a broad endothermic peak with a peak temperature of approximately 97 °C, which was attributed to dehydration. IC analysis showed that the sample contained approximately 1 equivalent of chloride ion, giving a salt formation ratio of 1 / 1. The hydrochloride salt crystalline form I was determined to be a hydrate.

[0205] [Table 9]

[0206] [Table 10]

[0207] Example 2: Preparation of p-toluenesulfonic acid salt An appropriate amount (20 mg to 30 mg) of compound A was weighed out and placed in a sample bottle at room temperature, then dissolved or suspended in 0.2 mL of a solvent selected from Table 8, and then p-toluenesulfonic acid solid or 1 M p-toluenesulfonic acid in methanol was added to react and form a salt, and the solution or suspension was stirred at room temperature for 5 to 15 hours. If no precipitate was formed, a poor solvent was added to promote the precipitation of the precipitate. If a solid was present, it was filtered, washed, and the sample was collected and dried at 50°C under vacuum for about 3 hours, and analyzed by XRPD, TGA, DSC, and the like. 1 H-NMR characterization was performed, and the specific information and results are shown in Table 8. The XRPD pattern and analysis of the p-toluenesulfonate salt crystalline form I prepared in Group 1 are shown in FIG. 4 and Table 8'.

[0208] [Table 11]

[0209] [Table 12]

[0210] In this example, p-toluenesulfonate crystalline form I was obtained. 1 The H-NMR characterization results are summarized in Table 9 and Figures 5-6.

[0211] p-Toluenesulfonate crystalline form I has a weight loss of about 2.5% up to 105°C, 1 H-NMR analysis showed that the weight loss in TGA was due to the removal of water and organic solvents since the sample contained 0.5% EtOH and 0.1% MTBE organic solvents. Two thermodynamic events appeared in the DSC pattern, a broad endothermic peak with a peak temperature of about 89 °C was determined to be due to dehydration, and a second endothermic peak with a peak value of about 127 °C was determined to be due to melting. Therefore, p-Toluenesulfonate crystalline form I was determined to be a hydrate.

[0212] [Table 13]

[0213] Example 3: Preparation of benzenesulfonate salt An appropriate amount (20 mg to 30 mg) of compound A base was weighed out and placed in a sample bottle at room temperature, then dissolved or suspended in 0.2 mL of a solvent selected from Table 10, and then solid benzenesulfonic acid or a 1 M methanol solution of benzenesulfonic acid was added to react and form a salt, and the solution or suspension was stirred at room temperature for 5 to 15 hours. If no precipitate was formed, a poor solvent was added to promote the precipitation of the precipitate. If a solid was present, it was filtered, a sample was collected, and dried at 50°C under vacuum for about 3 hours, and analyzed by XRPD, TGA, DSC, and the like. 1 H-NMR characterization was performed, and the specific information and results are shown in Table 10. The XRPD pattern and analysis of the benzenesulfonate salt crystalline form I prepared in Group 1 are shown in Figure 7 and Table 10'.

[0214] [Table 14]

[0215] [Table 15]

[0216] In this example, a benzenesulfonate crystalline form named benzenesulfonate crystalline form I was obtained. 1 H-NMR, TGA and DSC characterizations were performed, and the relevant characterization results are summarized in Table 11 and Figures 8 to 9.

[0217] The benzenesulfonate salt, Form I, has a weight loss of about 3.0% up to 117°C. 1H-NMR analysis showed that the sample contained 0.4% EtOH and 0.2% MTBE organic solvents, so the weight loss in the TGA was attributed to the removal of water and organic solvent residues. The broad overlapping endothermic peaks with a peak temperature of about 114 °C in the DSC pattern were attributed to dehydration (about 1 equivalent of H2O). The benzenesulfonate salt crystalline form I was determined to be a hydrate.

[0218] [Table 16]

[0219] Example 4: Preparation of the maleate salt 30 mg of compound A was weighed out and placed in a sample bottle at room temperature, then dissolved or suspended in 0.2 mL of a solvent selected from Table 12, and maleic acid solid or 1 M maleic acid in methanol was added to react and form a salt, and the solution or suspension was stirred at room temperature for 15 hours. If no precipitate was formed, a poor solvent was added to promote the precipitation of the precipitate. If a solid was present, it was filtered, the sample was collected, and dried under vacuum at 50°C for about 3 hours to obtain the product, which was then analyzed by XRPD, TGA, DSC, and the like. 1 H-NMR characterization was performed, and the specific information and results are summarized in Table 12. The XRPD patterns and analysis of the maleate salt crystalline form I and maleate salt crystalline form II prepared in Group 2 and Group 1 are shown in Figure 10 and Table 12', Figure 11 and Table 12'', respectively.

[0220] [Table 17]

[0221] [Table 18]

[0222] [Table 19]

[0223] In this example, two maleate crystalline forms were obtained, identified and named maleate crystalline forms I and II, respectively. 1 H-NMR, TGA and DSC characterizations were performed, and the relevant characterization results are summarized in Table 13 and Figures 12, 13, 14-1 and 14-2.

[0224] Maleate crystalline form I has a weight loss of about 8.4% up to 100°C, 1 H-NMR analysis showed that the sample contained approximately 13% EA, so the weight loss in the TGA was attributed to the removal of EA. The DSC pattern showed broad overlapping endothermic peaks with a peak temperature of approximately 83 °C, which was attributed to the removal of organic solvents. The maleate salt crystalline form I was determined to be an EA solvate.

[0225] Maleate Form II has a weight loss of about 3.0% up to 104°C; 1 H-NMR analysis showed that the sample contained approximately 2.0% MTBE, so the weight loss in the TGA was attributed to the removal of MTBE and water. The DSC pattern showed broad overlapping endothermic peaks with a peak temperature of approximately 108 °C, which was attributed to the removal of organic solvents and water.

[0226] [Table 20]

[0227] Example 5: Preparation of the oxalate salt 30 mg of compound A was weighed out and placed in a sample bottle at room temperature, then dissolved or suspended in 0.2 mL of a solvent selected from Table 14, and 1 M oxalic acid in methanol was added to react and form a salt, and the solution or suspension was stirred at room temperature for about 15 hours. If no precipitate was formed, a poor solvent was added to promote the precipitation of the precipitate. If a solid was present, the sample was collected by filtration, dried under vacuum at 50°C for about 5 hours, and analyzed by XRPD, TGA, DSC, and 1 H-NMR characterization was performed, and the specific information and results are shown in Table 14. The XRPD pattern and analysis of the oxalate crystalline form I prepared in Group 3 are shown in Figure 15 and Table 14'.

[0228] [Table 21]

[0229] [Table 22]

[0230] In this example, an oxalate crystalline form was obtained, designated as oxalate crystalline form I. For the oxalate samples of group 3 1 H-NMR, TGA, DSC, and IC characterization were performed, and the relevant characterization results are summarized in Tables 15 to 16 and Figures 16 and 17.

[0231] Form I of the oxalate salt has a weight loss of about 0.5% by 150°C; 1 H-NMR analysis showed that there was no residual organic solvent in the sample, so the weight loss in the TGA was attributed to the removal of adsorbed water. The DSC pattern showed a sharp endothermic peak with a peak temperature of about 206 °C, which was attributed to melting associated with the decomposition of the sample. IC results showed that the acid-base ratio of the sample was about 1 / 1. The oxalate crystalline form I was determined to be anhydrous.

[0232] [Table 23]

[0233] [Table 24]

[0234] Example 6: Amplification of Oxalate Crystal Form I Approximately 300 mg of compound A was weighed and dissolved in 100 μL of MeOH, followed by the addition of 78 mg of oxalic acid. An additional 2 mL of 2-Me-THF was then added, and the suspension was stirred for 3 days. After filtration, the sample was collected and dried under vacuum at 50 °C for approximately 15 h. The sample was analyzed by XRPD, IC, TGA, DSC, and NMR. 1 H-NMR characterization was performed.

[0235] Approximately 200 mg of oxalate crystalline form I was obtained, with a yield of approximately 53%. 1 H-NMR analysis was performed. IC results showed that the acid-base ratio of the oxalate was 1:1. TGA data showed that the sample had a weight loss of about 1.9% up to 150°C. Nuclear magnetic analysis showed that the sample contained 2.1% 2-Me-THF. The DSC pattern contained an endothermic peak with a peak temperature of 203°C, which was attributed to melting due to the decomposition of the sample. Detailed characterization results are shown in Table 17 and Figures 18 to 20.

[0236] [Table 25]

[0237] Example 7: Preparation of the fumarate salt and its crystalline form I An appropriate amount of compound A was weighed out and placed in a sample bottle at room temperature, then dissolved or suspended in 0.2 mL of a solvent selected from Table 18, and then solid fumaric acid was added to react and form a salt, and the solution or suspension was stirred at room temperature for a certain time (about 3 hours to 15 hours). If no precipitate was formed, a poor solvent was added to promote the precipitation of the precipitate. If a solid was present, the sample was collected by filtration, dried under vacuum at 50 °C for about 5 hours, and analyzed by XRPD, TGA, DSC, and 1 H-NMR characterization was performed, and specific information and results are shown in Table 18.

[0238] [Table 26]

[0239] The XRPD and analytical patterns of the fumarate crystalline product obtained in Group 4 are shown in FIG. 21 and Table 19'. 1 H-NMR, TGA and DSC characterizations were performed and the relevant characterization results are summarized in Table 19 and Figures 22-23.

[0240] The fumarate salt crystalline form I exhibits almost no weight loss up to 150°C, 1 H-NMR analysis showed that the sample contained 0.5 equivalents of fumaric acid (i.e., the fumarate salt was a salt formed from compound A and fumaric acid in a molar ratio of 1:0.5). A sharp endothermic peak with a peak temperature of about 157°C appeared in the DSC pattern, which was attributed to melting due to decomposition of the sample. The fumarate salt crystalline form I was determined to be anhydrous.

[0241] [Table 27]

[0242] [Table 28]

[0243] Example 8: Preparation of Fumarate Crystalline Form II 6.56 g of crude compound A was added to 46 mL of a mixed solvent with a volume ratio of ACN:THF=2:1, and the solid showed no sign of dissolving after heating to 60-70°C and refluxing. The solid still did not dissolve after adding 1.10 g (0.55 eq) of fumaric acid. The mixture was stirred at 60-70°C for 1 h, cooled naturally to 10-20°C and stirred for 16 h, filtered, and the filter cake was spun dry to obtain 7.32 g of a yellow solid fumarate salt. XRPD, TGA, DSC, and 1 H-NMR characterization was performed, and the relevant characterization results are summarized in Table 20 and Figures 24 to 27. Among them, the molar ratio of fumaric acid to compound A was 0.5:1, the anhydrous crystalline form was obtained, the chemical purity was 99.3%, and the crystallinity was high, which was designated as fumarate crystalline form II.

[0244] The PLM results showed that the fumarate crystalline form II obtained in this example was composed of particles with irregular morphology ranging from 10 μm to 80 μm, and the PSD results showed that its Dv(10) was 8.52 μm and Dv(90) was 43.2 μm.

[0245] [Table 29]

[0246] [Table 30]

[0247] Another example of preparing Fumarate Crystal Form II is shown in Table 20-1.

[0248] [Table 31]

[0249] Note: [1] It should be understood by those skilled in the art that the seed crystals of fumarate salt Form II are added using the fumarate salt Form II obtained by production, for example, referring to the fumarate salt Form II obtained in Groups 1 to 4 in Table 20.

[0250] Example 9: Fumarate Crystal Form I - Amplified Preparation Approximately 300 mg of compound A and 50.21 mg of fumaric acid were weighed and dispersed in 4.5 mL of a mixed solvent (EA / MeOH, volume ratio 35 / 1). Then, the mixture was stirred at room temperature overnight (about 15 h). After filtration, the sample was collected and dried under vacuum at 50°C for about 3 hours to obtain about 315 mg of fumarate salt crystalline form I, with a yield of about 90%, and the sample was analyzed by XRPD, DSC, TGA, DVS, and the like. 1 H-NMR analysis was carried out.

[0251] The TGA data showed that there was almost no weight loss up to 100°C. Nuclear magnetic analysis showed that the sample had no residual organic solvents. The DSC pattern contained an endothermic peak with a peak temperature of 159°C, which was attributed to melting due to the decomposition of the sample. Detailed characterization results are shown in Table 21 and Figures 30-32.

[0252] [Table 32]

[0253] Example 10: Preparation of the tartrate salt 30 mg of compound A was weighed out and placed in a sample bottle at room temperature, then dissolved or suspended in 0.2 mL of a solvent selected from Table 22, and L-tartaric acid solid was added to react and form a salt, and the solution or suspension was stirred at room temperature overnight (about 15 hours). If no precipitate was formed, a poor solvent was added to promote the precipitation of the precipitate. If a solid was present, the sample was collected by filtration, dried under vacuum at 50°C for about 3 hours, and analyzed by XRPD, TGA, DSC, and 1 H-NMR characterization was performed, and the specific information and results are shown in Table 22. The XRPD pattern and analysis of the tartrate salt obtained as prepared in Group 1 are shown in Figure 33, Table 22'.

[0254] [Table 33]

[0255] [Table 34]

[0256] In this example, a tartrate salt crystalline form was obtained, designated as tartrate salt crystalline form I. For the tartrate salt samples, 1 H-NMR, TGA and DSC characterizations were performed and the relevant characterization results are summarized in Table 23 and Figures 34-35.

[0257] The tartrate salt crystalline form I had a weight loss of only 0.8% before 150°C, 1 H-NMR analysis showed that the sample contained 0.8% MTBE and 1 equivalent of acid. The DSC pattern showed a sharp endothermic peak with a peak temperature of approximately 135 °C, which was attributed to the melting of the sample. Tartaric acid crystalline form I was determined to be anhydrous.

[0258] [Table 35]

[0259] Example 11 Preparation of Free Base Crystalline Form I Take 30 mg of compound A, add 0.2 mL of ethyl acetate to obtain a suspension, stir at room temperature for 15 hours, filter, and the collected sample was dried in vacuum at 50°C for 3 hours to obtain free base crystalline form I. The XRPD detection pattern and analysis are shown in Figure 36, and the DSC-TGA detection result is shown in Figure 37, DSC: 157°C. 1 The H-NMR pattern is shown in Figure 37-2.

[0260] [Table 36]

[0261] Example 12 Preparation of Free Base Crystalline Form II 300 mg of compound A was taken and dispersed in 2 mL of mixed solvent (EtOH / Water, v / v=1 / 10), stirred at room temperature for 3 days, and filtered. The filtered collected sample was dried in vacuum at 50° C. for about 15 hours to obtain free base crystalline form II. The XRPD detection pattern and analysis of the obtained crystalline form II are shown in FIG. 38, and the DSC-TGA detection result is shown in FIG. 39. 1 The H-NMR pattern is shown in Figure 39-1.

[0262] [Table 37]

[0263] Example 13: Hygroscopicity measurements of free base crystalline form II, oxalate crystalline form I, fumarate crystalline form I and fumarate crystalline form II Approximately 40 mg of each of the free base crystalline form II, the oxalate crystalline form I, the fumarate crystalline form I and the fumarate crystalline form II was weighed and subjected to the hygroscopicity evaluation in the DVS tray after peeling according to the dynamic moisture sorption / desorption analysis (DVS) method described above.

[0264] The DVS data showed that the weight of free base crystalline Form II increased by 1.3% / 1.5% respectively in the range of 0.0%RH~80% / 90%RH, indicating that it was slightly hygroscopic, and the detailed characterization results are shown in Figure 40. The XRPD pattern of free base crystalline Form II did not change before and after the DVS measurement, and the crystalline form did not change.

[0265] The DVS data showed that the weight of oxalate Form I increased by 2.3% / 3.1% respectively in the range of 0.0%RH~80% / 90%RH, indicating that the sample was slightly hygroscopic, and the detailed characterization results are shown in Figure 41. The XRPD patterns of oxalate Form I before and after DVS measurement were consistent, and the crystal form did not change.

[0266] The DVS data showed that the weight gain of fumarate crystalline form I was less than 0.2% in the range of 0.0% RH to 80% / 90% RH, indicating that it was not hygroscopic. Detailed characterization results are shown in Figure 42. The XRPD patterns of fumarate crystalline form I before and after DVS measurement were consistent, indicating that the crystalline form did not change.

[0267] DVS data showed that fumarate salt Form II absorbed about 0.2% water at 0.0%RH~95%RH and was not hygroscopic. The XRPD patterns of fumarate salt Form II before and after DVS measurement were consistent, and the crystal form did not change. Detailed characterization results are shown in Figure 43 and Figure 44.

[0268] Example 14: Solubility measurements of free base crystalline form II, oxalate crystalline form I, fumarate crystalline form I and fumarate crystalline form II The solubilities of the free base crystalline Form II, the oxalate crystalline Form I, the fumarate crystalline Form I and the fumarate crystalline Form II were measured in biorelevant media at 37°C.

[0269] 15 mg of free base crystalline form II, oxalate crystalline form I, fumarate crystalline form I and fumarate crystalline form II were weighed and dispersed in 5.0 mL of biorelevant medium. The mixture was shaken at 37°C with a rotation speed of 100 rpm, and 1 mL of the dispersion was taken out and filtered at 0.5 hours, 2 hours and 24 hours, respectively. The solubility of the filtrate was measured by HPLC, the pH value was measured by a pH meter, and the crystalline forms of the filter cake were subjected to XRPD characterization. The relevant characterization results are summarized in Table 26.

[0270] [Table 38]

[0271] In the three biorelevant media, the fumarate crystalline form I had higher solubility (0.5 h) than the free base crystalline form II, with the 0.5 h solubility of the two solid forms in SGF medium being the highest (1.0 mg / mL vs. 1.9 mg / mL), almost 10 times higher than that of the oxalate crystalline form I (0.07 mg / mL). Both salt forms dissociated to the free base in the solubility measurements.

[0272] In the solubility measurements, the 24-h crystal form of free base Form II in biorelevant media did not change. The 24-h crystal form of oxalate Form I in FeSSIF and SGF did not change, but partially dissociated to free base Form II in FaSSIF at 24 h. Fumarate Form I dissociated to free base Form II at 0.5 h in FaSSIF and FeSSIF media.

[0273] The solubility measurements showed that the solubility of the fumarate salt crystalline form I in three biorelevant media at 0.5 hours was approximately twice that of the free base crystalline form II.

[0274] Example 15: Stability Measurement of the Solid Forms of Free Base Crystalline Form II, Oxalate Crystalline Form I, Fumarate Crystalline Form I, and Fumarate Crystalline Form II Appropriate amounts of free base crystalline form II, oxalate crystalline form I, fumarate crystalline form I and fumarate crystalline form II were taken and placed under two conditions of 60°C / closed and 40°C / 75% RH open for 1 week and 2 weeks, and the samples at 0 days, 1 week and 2 weeks were dissolved in diluent to prepare approximately 1.0 mg / mL solutions to analyze chemical stability by HPLC, and the solid samples after 1 week or 2 weeks were analyzed for physical stability by XRPD measurement. The relevant characterization results are summarized in Tables 27 to 28.

[0275] [Table 39]

[0276] [Table 40]

[0277] The stability results showed that the free base crystalline form II and the fumarate crystalline form I were physically and chemically stable for one week under two conditions, 60°C / closed and 40°C / 75%RH open, while the oxalate crystalline form I was decomposed.

[0278] The 14-day stability test results showed that the free base crystalline form II and the fumarate crystalline form I were physically and chemically stable for 2 weeks under two conditions: 60°C / closed and 40°C / 75% RH open.

[0279] Example 16: Comparison of pharmacokinetic studies of free base crystalline form II and fumarate crystalline form I Twelve SD rats (purchased from Beijing Weitong Lihua Laboratory Animal Technology Co., Ltd.), half female and half male, weighing 180 g–280 g, were used in the experiment. They were randomly divided into four groups, with three rats in each group: groups 1 and 3 were female rats, and groups 2 and 4 were male rats.

[0280] The free base crystal form II and the fumarate crystal form I of compound A were dissolved in 5% TPGS and administered intragastrically at a drug concentration of 20 mg / mL (calculated as compound A), with a volume of 10 mL / kg, a dose of 20 mg / kg, and a QD (once a day) frequency. The concentration of compound A in plasma was detected.

[0281] The data were analyzed by a noncompartmental model using WinNonlin (version 5.2.1 Pharsight, Mountain View, CA) to obtain PK parameters (C0, C1, C2, C3, C4, C5, C6, C7, C8, C9, C10, C11, C12, C13, C14, C15, C16, C17, C18, C19, C20, C21, C22, C23, C24, C25, C36, C37, C48, C49, C59, C61, C72, C73, C74, C75, C81, C92, C93, C10, C11, C12, C13, C24, C14, C15, C25, C36, C48, C59, C61, C72, C73, C74, C75, C81, max , T max , AUC 0-last , AUC inf , T 1 / 2 , CL, Vz, and other parameters were selected.

[0282] Please refer to Table 29 for each pharmacokinetic parameter.
