Liver organoid model for hyperbilirubinemia and methods for producing and using same

Abstract

Disclosed herein is an improved method for using hemoglobin metabolites such as bilirubin to mature pluripotent stem cell derived liver organoids.Furthermore, by adjusting the amount of bilirubin used, liver organoids can be used as a model for hyperbilirubinemia.The scalability and ease of handling of these liver organoids make them excellent targets for drug screening against diseases such as hyperbilirubinemia.Also disclosed herein is the use of exogenous L-gulonolactone oxidase to improve the viability of bilirubin-treated liver organoids.
