By mixing the adjustable fat with buffer solution to form fat nanoparticles and adding nucleic acids to these particles, the difficulty of transferring nucleic acid to cells in the prior art is solved, and efficient and safe nucleic acid distribution and therapeutic effects are achieved.

Abstract
To provide methods of producing lipid nanoparticle (LNP) formulations, to provide LNP formulations produced by the method, and to provide therapeutic and diagnostic uses related to the produced LNP formulations.SOLUTION: A method comprises: mixing a lipid solution comprising an ionizable lipid with an aqueous buffer solution comprising a first buffering agent, thereby forming a lipid nanoparticle solution comprising lipid nanoparticles; and adding a nucleic acid solution comprising a nucleic acid to the lipid nanoparticle solution, thereby forming a LNP formulation comprising the lipid nanoparticles associated with the nucleic acid.SELECTED DRAWING: None
