Abstract

To provide a technology for more reliably achieving a higher cell viability rate in the preparation of CAR-T cells while employing the transposon method for CAR gene transfer. The present invention provides a method for preparing genetically modified T cells that express a chimeric antigen receptor, comprising the following steps (i) to (iv): (i) preparing non-proliferating cells that retain a viral peptide antigen by culturing a monocyte-reduced T cell-containing cell population in the presence of a viral peptide antigen and subjecting the cell population to a treatment that eliminates its proliferation ability; (ii) extracting a target antigen from the monocyte-reduced T cell-containing cell population by a transposon method. (ii) obtaining genetically modified T cells into which a specific chimeric antigen receptor gene has been introduced; i) The non-proliferating cells prepared in step (i) and the genetically modified T cells obtained in step (ii) and co-culturing; (iv) recovering the cells after culturing.
