Abstract
Description
Technical Field
[0001] The present invention relates to a composition for improving skin condition or preventing deterioration of skin condition, a method for improving skin condition, a method for preventing deterioration of skin condition, and a composition for improving acne.
Background Art
[0002] Skin troubles are caused by external factors (ultraviolet rays, dryness, pollutants, etc.) and internal factors (aging, hormonal imbalance, stress, dietary habits, etc.). These factors reduce various functions of the skin, causing troubles such as acne, inflammation, dryness, and spots, and deteriorating the skin condition. Skin troubles not only impair appearance, but also cause discomfort due to itching and pain, and combined with psychological effects, can lower QOL (Quality of Life), so maintaining healthy skin is an important issue for many people. In order to maintain healthy and beautiful skin, the development of effective skin care products and skin care methods has been ongoing for many years.
[0003] In the skin care market, in order to avoid the adverse effects of synthetic chemicals on the skin, demand for naturally derived ingredients and ingredients derived from organisms other than animals (plants, algae, etc.) is increasing. Examples of naturally derived ingredients and ingredients derived from organisms other than animals include plant-derived raw materials extracted from plant flowers, leaves, roots, fruits, etc., algae-derived raw materials extracted from seaweed and freshwater algae, and microorganism-derived raw materials obtained by fermentation of microorganisms, etc. Sulfated polysaccharides derived from the freshwater blue-green alga Aphanothece sacrum are known to have excellent moisturizing properties and the like, and have been proposed as raw materials for cosmetics (for example, Patent Documents 1 to 2). It has also been reported that the use of this sulfated polysaccharide may alleviate symptoms in patients with atopic dermatitis (see Non-Patent Document 1).

Patent Document 1
[0005] [Non-Patent Document 1] Ngatu et al., “Anti-allergic and Profilaggrin(ProFLG)-mRNA expression modulatory effects of sacran,” International Journal of Biological Macromolecules, No. 105, 2017, p1532-1538 [Overview of the Initiative] [Problems that the invention aims to solve]
[0006] Among the skin problems mentioned above, acne (acne vulgaris) is a skin disease that can occur at a wide range of ages, from adolescence to young adulthood, and is often accompanied by itching and pain, causing significant discomfort. Furthermore, acne often leaves scars, causing appearance and psychological stress, and significantly impacting quality of life, thus requiring appropriate treatment. However, there are still few natural ingredients that are effective against acne, and further development is desired.
[0007] The present invention has been made in view of the above problems, and aims to provide a composition containing naturally derived active ingredients that can improve skin condition or prevent the worsening of skin condition in the event of acne and other skin problems, and a method for improvement or prevention using said composition. [Means for solving the problem]
[0008] The inventors of this invention conducted intensive research to solve the above problems and, as a result, novelly discovered that sulfated polysaccharides derived from Suizenji Nori improve skin condition or prevent the worsening of skin condition by improving acne, thus completing the present invention.
[0009] In other words, the present invention provides the following invention. (1) A composition containing sulfated polysaccharides derived from Suizenji Nori, for improving skin condition or preventing deterioration of skin condition. (2) The composition of (1) wherein the improvement in skin condition is one or more selected from the group consisting of improvement of acne, suppression of skin itchiness, improvement of skin texture, and improvement of skin smoothness. (3) The composition of (1) above, wherein the prevention of worsening of skin condition is the prevention of rough skin. (4) A method for improving the condition of healthy skin using the composition of (1) above (excluding methods for treating human beings). (5) A method for preventing deterioration of the condition of healthy skin using the composition of (1) above (excluding methods for treating humans). (6) A composition for improving acne, containing sulfated polysaccharides derived from Suizenji Nori. [Effects of the Invention]
[0010] According to the present invention, acne can be improved. In addition, it is possible to obtain an effect of suppressing itching associated with acne, an effect of improving the skin condition in which the texture and smoothness have deteriorated due to acne, and an effect of preventing skin roughness associated with acne, making it possible to tighten and improve the health of the skin. Furthermore, since Suizenji-nori-derived sulfated polysaccharides are derived from algae, they can be used safely and securely compared to synthetic ingredients derived from petroleum raw materials. In addition, Suizenji-nori-derived sulfated polysaccharides themselves are expected to have moisturizing effects, so they can be used not only as an acne treatment agent but also to improve various skin functions and skin conditions. [Modes for carrying out the invention]
[0011] The present invention will be described in more detail below. However, the present invention is not limited to the embodiments shown below. In this specification, unless otherwise specified, "parts" and "%" mean "parts by mass" and "% by mass," respectively. In this specification, unless otherwise specified, "XX or greater and YY or less" or "XX to YY" means a numerical range including the lower and upper limits which are the endpoints. When a numerical range is described in steps, the upper and lower limits of each numerical range can be combined in any way.
[0012] <Compositions for improving skin condition; Compositions for preventing deterioration of skin condition> The composition for improving skin condition and the composition for preventing deterioration of skin condition (hereinafter sometimes collectively referred to as "the composition") of the present invention contains sulfated polysaccharides derived from Suizenji-nori and is an agent (skin condition improving agent or skin condition deterioration prevention agent) that has the effect of improving skin condition and / or preventing deterioration of skin condition. In the above composition, sulfated polysaccharides derived from Suizenji-nori are contained as an active ingredient.
[0013] The skin condition improvement effect refers to the effect of improving or enhancing one or more properties of the skin. In other words, it can be said to be an effect of improving skin problems. Examples of skin problems include acne (acne vulgaris; including pimples), itchy skin, redness of the skin, excessive sebum secretion on the skin surface, and skin texture and smoothness worsened by acne, etc. By improving these skin problems, the composition of the present invention provides the effect of conditioning troubled skin and tightening troubled skin. Furthermore, the effect of preventing deterioration of skin condition refers to the effect of preventing the deterioration of one or more skin properties. More specifically, deterioration of skin properties includes the occurrence of rough skin and acne. By preventing the deterioration of these skin conditions, the composition of the present invention provides the effect of keeping the skin healthy.
[0014] (Sulfated polysaccharides derived from Suizenji-nori) The sulfated polysaccharide derived from Aphanothece sacrum (hereinafter sometimes simply referred to as "sulfated polysaccharide" or "polysaccharide") is a type of sulfated polysaccharide derived from the freshwater cyanobacterium Aphanothece sacrum obtained naturally or by aquaculture (cultivation), and generally has an average molecular weight of 2,000,000 or more. Specifically, it has a repeating structure of sugar chain units in which a sugar structure having a hexose structure and a sugar structure having a pentose structure are linked in a linear or branched form via α-glycosidic bonds or β-glycosidic bonds, the sugar chain unit contains a sulfated sugar as the sugar structure, and in the sugar chain unit, 2.7 or more hydroxyl groups per 100 hydroxyl groups are sulfated, or it is a sugar derivative in which elemental sulfur accounts for 1.5% by mass or more of all elements.
[0015] Specific examples of the sulfated polysaccharide include sulfated muramic acid and sulfated N-acetylmuramic acid.
[0016] The sulfated polysaccharide contains at least glucose, galactose, mannose, galactosamine, xylose, arabinose, glucuronic acid, galacturonic acid, fucose, and rhamnose, and a functional group selected from the functional group group including at least a sulfate group, a lactic acid group, and a methyl group may be bonded to any bonding position in these sugar structures. The sulfate group, which is a characteristic of the sulfated polysaccharide derived from Aphanothece sacrum, is preferably contained in the sulfated polysaccharide in an amount of 3 to 20% by mass, more preferably 5 to 15% by mass, and even more preferably 8 to 12% by mass.
[0017] A part of the sugar structure constituting the sulfated polysaccharide may further be bonded to a peptide or a lipid.
[0018] The molar ratio of the main sugar structures constituting the sulfated polysaccharide may be arabinose 1.1 : fucose 3.7 : rhamnose 15.4 : xylose 17.0 : mannose 10.5 : galactose 12.3 : galacturonic acid 4.6 : glucuronic acid 4.7 : glucose 28.8 : galactosamine 2.03. In addition, the number of types of sugar structures (monosaccharides) constituting the sulfated polysaccharide may be 5 or more, 7 or more, 8 or more, or 10 or more.
[0019] The average molecular weight of the sulfated polysaccharide is, for example, 500,000 or more, 800,000 or more, 1,000,000 or more, 2,000,000 or more, 5,000,000 or more, 10,000,000 or more, 20,000,000 or more, etc., and is 30,000,000 or less, 28,000,000 or less, 25,000,000 or less, 22,000,000 or less, etc. A non-contradictory combination thereof is also acceptable. For example, it may be 1,000,000 to 30,000,000, 5,000,000 to 28,000,000, 10,000,000 to 25,000,000, 20,000,000 to 22,000,000, etc.
[0020] The sulfated polysaccharide may be one extracted, isolated and purified from Aphanothece sacrum. The method for extracting the sugar derivative from Suizenji-nori is not particularly limited, and conventional methods such as those described in Patent Document 2 can be used. Specifically, for example, the following can be used: A suitable amount of Suizenji-nori is frozen and thawed, then washed with water to remove water-soluble pigments, and then lipid-soluble pigments are removed by stirring with an organic solvent such as ethanol for 24 hours. After separating the organic solvent from the Suizenji-nori from which the pigments have been removed, the solvent is dissolved in an aqueous solution of sodium hydroxide, for example, at 40 to 80°C, while stirring for 5 to 12 hours. The aqueous solution of the sugar derivative extracted by this operation is neutralized with concentrated hydrochloric acid, for example. Here, desalting may or may not be performed by immersing in 60 to 80% isopropyl alcohol, for example. After that, the solution is poured into 100% isopropanol while stirring to precipitate a gel-like sugar derivative, and then dried with hot air. Drying with hot air may be done at 85 to 100°C for 6 to 24 hours, for example. The extracted and precipitated sugar derivatives may be purified by centrifugation, filtration, alcohol washing, etc.
[0021] Sulfated polysaccharides may be commercially available products, such as "Sacran" (registered trademark) manufactured by Green Science Material (GSM) and "Sacranex" (registered trademark) manufactured by the DIC Group.
[0022] Furthermore, the sulfated polysaccharides may be present in a form that is included in the crushed dried product of Suizenji-nori (hereinafter referred to as "crushed dried product of Suizenji-nori"), insofar as they achieve the effects of the present invention. Therefore, the present invention can also provide a composition containing crushed dried Suizenji-nori containing sulfated polysaccharides as an active ingredient. The crushed dried Suizenji Nori containing the sugar derivative may be, for example, a crushed freeze-dried Suizenji Nori containing the sugar derivative, a crushed sun-dried Suizenji Nori containing the sugar derivative, or a crushed hot-air dried Suizenji Nori containing the sugar derivative, and any one of these may be used, or a combination of several may be used. Crushed dried Suizenji-nori containing sulfated polysaccharides can all be prepared by conventional methods. For example, crushed freeze-dried Suizenji-nori containing the aforementioned sugar derivatives can be prepared by freezing Suizenji-nori containing water in a low-temperature freezer, removing the water contained within the algae by vacuum freeze-drying to obtain freeze-dried Suizenji-nori, and then crushing it (for example, by grinding it with a mortar and pestle, or by grinding it into a powder using a pulverizer). After that, it may be suspended in an appropriate solvent or solution.
[0023] Furthermore, the sulfated polysaccharides may be present in an extract of Suizenji-nori (hereinafter referred to as "Suizenji-nori extract") insofar as they achieve the effects of the present invention. Therefore, the present invention can also provide a composition containing a Suizenji Nori extract containing sulfated polysaccharides as an active ingredient. Suizenji-nori extract containing sulfated polysaccharides can be prepared, for example, by the method described above.
[0024] The composition of the present invention improves skin condition or prevents the deterioration of skin condition. The target skin cells may be cells present in the epidermis or cells present in the dermis. Examples of cells present in the epidermis include keratinocytes, granular cells, spinous cells, Langerhans cells, and basal cells. Examples of cells present in the dermis include fibroblasts and pigment cells (melanocytes). In addition to the above, sebaceous glands, pores, and hair follicles can also affect skin condition and acne, so the composition of the present invention may also act on sebaceous gland-related cells such as sebaceous gland cells, hair follicle-related cells such as hair matrix cells, and hair root / hair-related cells such as dermal papilla cells. Furthermore, since commensal bacteria on the skin also affect skin condition, sebum secretion, and decomposition, the composition may also act on cells related to commensal bacteria.
[0025] The composition of the present invention may be used as is, but may also be used as a cosmetic, topical skin preparation, quasi-drug, pharmaceutical, etc., for the purpose of obtaining an effect of improving skin condition and / or preventing the deterioration of skin condition, or may be incorporated into cosmetics, topical skin preparations, quasi-drugs, or pharmaceuticals. Among these, it is preferable to use it in the form of a cosmetic or topical skin preparation.
[0026] • Cosmetics or topical skin preparations When the composition of the present invention is used in cosmetics or topical skin preparations, the cosmetics or topical skin preparations can be manufactured by conventional methods (for example, methods described in the Japanese Pharmacopoeia) using the composition itself containing the sulfated polysaccharide derived from Suizenji Nori, or, if applicable, by further mixing it with other components.
[0027] Other components include pharmaceutically acceptable carriers. The effects of improving skin condition and preventing the deterioration of skin condition are as described above. [Examples]
[0057] The present invention will be specifically described below with reference to examples, but the present invention is not limited to these examples.
[0058] [Manufacturing example] The compositions (creams) for Production Example 1 and Comparative Production Example 1 were prepared using the following procedure. First, component A, as shown in Table 1, was dispersed at room temperature and then heated to 80°C and mixed and dissolved. In a separate container, component B was heated to 80°C and mixed and dissolved. Next, component A was gradually added to component B in several batches while stirring with a homomixer at 80°C. After that, the mixture was emulsified by stirring with a homomixer at 7000 rpm for 3 minutes. Then, component C was added while stirring at 80 rpm with the stirring blade changed to a U-shaped blade, and the mixture was air-cooled to below 40°C to obtain the compositions (creams) shown in Table 1. The content of each component listed in Table 1 is in parts by mass.
[0059] [Table 1]
[0060] [Experimental Example 1] The effects of sulfated polysaccharides derived from Suizenji-nori on the skin were evaluated using the following procedure. The study involved 30 volunteers aged 15 to 42 years (average 21.1 ± 5.0 years). The subjects had combination or oily skin (U-zone sebum >130 μg / cm³). 2 The subjects were defined as individuals who reported having mild to moderate acne (approximately 2-3 stages on the IGA scale). The test sample (cream A) was applied to one side of each subject's face, and a placebo (cream B) was applied to the other side of their face. The samples were applied twice daily, morning and evening, for a total of four weeks. Data was collected before the start of the study and after four weeks.
[0061] The area and volume of acne were measured using a Nikon D5600 digital camera (product name, manufactured by Nikon Corporation). High-resolution macro photographs were taken of a 50mm wide x 50mm high area of the cheek, and the area and volume of acne after 4 weeks were calculated using image analysis, with the pre-test values set to 100%.
[0062] [Table 2]
[0063] [Experimental Example 2] The effects of sulfated polysaccharides derived from Suizenji-nori on the skin were evaluated using the following procedure. Similar to Experimental Example 1, acne on the entire cheek was evaluated, and the number and circumference of acne were calculated. The calculated value for Cream A (measured value after 4 weeks divided by the value before the start of the experiment) is set as 100% when the calculated value for Cream B (measured value after 4 weeks divided by the value before the start of the experiment) after 4 weeks is shown in Table 3 as the acne suppression ratio. Furthermore, the amount of sebum on the cheeks was measured using the oil content measuring device "Sebumeter SM 815" (product name, manufactured by Courage+Khazaka). Table 3 shows the sebum secretion suppression ratio of Cream A (measured value after 4 weeks divided by value before the start of the test) compared to the calculated value of Cream B after 4 weeks (measured value after 4 weeks divided by value before the start of the test), which is set to 100%.
[0064] [Table 3]
[0065] In addition, after the trial in Experiment Example 2, the subjects were given a questionnaire on each of the following items using a 4-point scale (1 = strongly disagree, 2 = disagree, 3 = agree, 4 = strongly agree), and their satisfaction level was calculated using the following formula. The satisfaction levels for each item of Cream A and Cream B are shown in Table 4. Satisfaction level (%) = Number of people who answered 3 or 4 / Total number of respondents
[0066] [Table 4]
[0067] [Experimental Example 3] The effects of sulfated polysaccharides derived from Suizenji-nori on the skin were evaluated using the following procedure. The study involved 32 volunteers aged 20 to 61 years (average 39.8 ± 11.6 years). Participants were defined as having sensitive skin and self-reporting moderate to very severe facial eczema (redness grade 2-4 on the Eiben-Nielson light scale). The test sample (cream A) was applied to one side of the subject's face, and a placebo (cream B) was applied to the other side of their face, twice daily, morning and evening, for a total of four weeks. After the trial, participants were given a questionnaire on each of the following items using a 4-point scale (1 = strongly disagree, 2 = disagree, 3 = agree, 4 = strongly agree), and satisfaction levels were calculated using the following formula. Table 4 shows the satisfaction levels for each item of Cream A and Cream B. Satisfaction level (%) = Number of people who answered 3 or 4 / Total number of respondents
[0068] [Table 5]
[0069] From the results above, it was confirmed that the composition according to the present invention reduces the area, volume, number, and circumference of acne, as well as the amount of sebum secreted. Furthermore, an improvement in acne was also observed in a comprehensive questionnaire administered to participating volunteers. Therefore, it has been confirmed that the composition of the present invention can improve acne, prevent and improve the worsening of skin conditions (itching, redness, uneven discoloration, discomfort, dryness), prevent and improve rough skin (decreased texture, decreased smoothness, decreased luster, etc.), and contribute to maintaining healthy skin.
