AI Technical Summary
There is a need for natural ingredients that can enhance the sun protection factor (SPF) of sunscreen compositions, particularly to increase UVA and UVB protection, as existing sunscreen activators are limited and challenging to improve SPF effectively.
Incorporating sialylated human milk oligosaccharides, such as 3'sialyllactose and 6'sialyllactose, with UVA filters like butyl methoxydibenzoylmethane and diethylamino hydroxybenzoyl hexyl benzoate in a composition, preferably in an oil-in-water emulsion, to enhance SPF properties.
The combination significantly increases the SPF of sunscreen compositions, providing improved protection against UVA and UVB rays, addressing skin aging and cancer risks, while being cosmetically acceptable for topical application.
Abstract
Description
Detailed description of the invention
[0001] The present invention relates to a composition containing at least one UVA sunscreen activator and having improved sun protection index (SPF) properties, and to a method for improving sun protection index (SPF) properties.
[0002] The use of compositions containing UV sunscreen activators (also called sunscreens) is very common today. Sunscreen activators in such compositions protect keratinous substances, particularly skin, from damage caused by UV rays, including damage from both UVA and UVB rays.
[0003] UVA rays have the lowest energy of all UV rays. These rays cause aging of skin cells and can indirectly damage the DNA of cells. UVA rays are mainly associated with long-term skin damage such as wrinkles, but are also thought to play a role in some skin cancers. UVB rays have shorter wavelengths. UVB radiation is associated not only with skin burns but also with skin cancer. Unprotected exposure to UVA and UVB rays damages the DNA of skin cells, causing genetic defects or mutations that can lead to skin cancer and premature aging.
[0004] Therefore, the need for sunscreens that offer high levels of UVA and UVB protection is increasing in order to achieve ultimate skin protection.
[0005] The standard measure for determining the amount of protection a composition containing sunscreen activators provides against UV radiation is the sun protection factor (SPF). However, given the limited number of sunscreen activators approved worldwide, increasing the SPF of compositions containing sunscreen activators can be challenging.
[0006] Therefore, there is still a need for preferably natural ingredients that can increase the SPF of the sunscreen active ingredients in the composition.
[0007] Surprisingly, it was found that certain human milk oligosaccharides can significantly increase the SPF of compositions containing certain UVA filters when combined with UVB and / or broadband UV filters.
[0008] Accordingly, in the first embodiment, the present invention relates to a composition for keratinous substances (e.g., hair or skin) comprising at least one UVA filter selected from the group consisting of butyl methoxydibenzoylmethane and diethylamino hydroxybenzoyl hexyl benzoate, and at least one sialylated human milk oligosaccharide. Preferably, the composition is in oil-in-water form (O / W emulsion).
[0009] The present invention also relates to a method for enhancing the sun protection index (SPF) properties of a composition comprising at least one UVA filter selected from the group consisting of butyl methoxydibenzoylmethane and diethylamino hydroxybenzoyl hexyl benzoate, the method comprising adding at least one sialylated human milk oligosaccharide to the composition in an amount effective for enhancing the SPF properties of the composition.
[0010] The present invention also relates to the use of sialylated human milk oligosaccharides to enhance the sun protection index (SPF) of a composition comprising at least one UVA filter selected from the group consisting of butyl methoxydibenzoylmethane and diethylamino hydroxybenzoyl hexyl benzoate.
[0011] The present invention also relates to a method for preparing a composition having improved sun protection index (SPF) properties, comprising adding to the composition, during the preparation of the composition, at least one UVA filter selected from the group consisting of butyl methoxydibenzoylmethane and diethylamino hydroxybenzoyl hexyl benzoate, and at least one sialylated human milk oligosaccharide. It is well understood that the improved properties are compared to a composition without sialylated human milk oligosaccharides having all the definitions and preferences given herein.
[0012] The present invention further relates to a method for producing a composition, the method comprising combining at least one UVA filter selected from the group consisting of butyl methoxydibenzoylmethane and diethylamino hydroxybenzoyl hexyl benzoate with at least one sialylated human milk oligosaccharide in the composition.
[0013] The use of the composition of the present invention as a sunscreen composition for protecting the skin from the effects of skin aging, particularly wrinkles (especially from UV-induced skin aging), and for sun protection, is also in accordance with the present invention.
[0014] Finally, the subject matter of the present invention is a method for the cosmetic treatment of keratinous substances, particularly skin, where compositions as defined herein are applied particularly to such keratinous substances, particularly skin. This method is particularly suitable for protecting the skin from the harmful effects of ultraviolet radiation, such as sunburn and / or photoaging.
[0015] The terms “UV filter” and “sunscreen activator” are used interchangeably herein and refer to compounds that absorb light in the UVB and / or UVA range (also known as UVA, UVB and broadband (i.e., UVA & UVB) filters). Preferably, such compounds have an E1 / 1 value of at least 150, more preferably at least 180, and most preferably at least 190 (i.e., λ max It has a UV absorbance of 1% at a concentration with an absorption layer thickness of 1 cm.
[0016] The term "keratinous material" means skin (body, face, eye contours, scalp), hair, eyelashes, eyebrows, body hair, nails, and / or lips. Preferably, in all embodiments, the keratinous material is skin.
[0017] The term "human milk oligosaccharides (HMOs)" refers to a family of unconjugated glycans that are very abundant in human breast milk and possess unique structural diversity. Originally, HMOs were found to promote the growth of Bifidobacterium species in the gut and are proposed to be prebiotic "bifidobacterial factors" or human milk glycans that are uniquely found in the stool of breastfed infants compared to infants fed formula.
[0018] Butyl methoxydibenzoylmethane (CAS number: 70356-09-1; abbreviated as BMDBM), also known as avobenzone, is sold, for example, by DSM Nutritional Products Ltd. under the trade name "Parsol® 1789".
[0019] Diethylaminohydroxybenzoyl hexyl benzoate (CAS number: 302776-68-7; abbreviated as DHHB), also known as hexyl 2-[4-(diethylamino)2-hydroxybenzoyl]benzoate, is sold, for example, by BASF under the trade name Uvinul® A plus.
[0020] The SPF according to the present invention is understood to be measured by the in vitro SPF method outlined in ISO 24443 (Determination of sunscreen UVA photoprotection in vitro, 01.06.2012) and illustrated in the examples.
[0021] HMOs generally consist of five monosaccharides: glucose (Glc), galactose (Gal), N-acetylglucosamine (GlcNAc), fucose (Fuc), and sialic acid (Sia), primarily along with N-acetylneuraminic acid (Neu5Ac) if sialic acid (Sia) is not the only form. More than 200 different HMOs have been identified to date, but among the most important are 3'-sialyl lactose sodium salt (3'SL) and 6'-sialyl lactose sodium salt (6'SL).
[0022] HMO can be isolated from breast milk or produced chemically or biochemically. HMOs are available from various manufacturers.
[0023] For the purposes of the present invention, the source of sialylated HMO is not essential. It is clear that HMOs from various sources can be used.
[0024] Particularly preferred sialylated HMOs according to the present invention are sialyllactose and its salts (preferably the sodium salt), for example, in particular 3'sialyllactose (3'SL), 6'sialyllactose (6'SL) and their respective sodium salts (CAS numbers: 35890-39-2 (3'sialyllactose); 128596-80-5 (3'sialyllactose sodium salt); 35890-39-2 (6'sialyllactose); 157574-76-0 (6'sialyllactose sodium salt)).

[0121] The following examples are provided to further illustrate the composition and effects of the present invention. These examples are merely illustrative and are not intended to limit the scope of the present invention in any way.
[0122] [Examples] [Brief Explanation] The 3SL and 6SL of HMO were evaluated for their SPF boosting effect. For this purpose, various sunscreens were prepared according to standard methods in the art as outlined in Table 1a.
[0123] [In Vitro SPF Measurement] The in vitro SPF test was performed on four PMMA plates (HD6 plates manufactured by Helioscreen, 5 cm × 5 cm, roughness of Sa 6 μm). 32.5 mg of each formulation (i.e., 1.3 mg / cm 2 ) was uniformly applied onto the PMMA plates and dried for 30 minutes.
[0124] In vitro SPF was measured using a Labsphere 2000 UV Transmittance Analyzer. Each PMMA plate was measured nine times at different points on the plate, yielding 36 data points. The results were calculated as the average of these 36 data points.
