Description

[Technical Field]

[0001] This invention relates to a personal care composition comprising moringa oil and coconut oil in a specific weight ratio. Surprisingly, it was found that the expression of the heme oxygenase 1 (HMOX1) gene is synergistically upregulated within this ratio range. [Background technology]

[0002] Human skin, through its barrier function, mechanical strength, and water impermeability, can provide physicochemical protection against environmental attacks. It is constantly exposed to harmful environmental and extreme conditions such as ultraviolet radiation, blue light, and pollution. Such exposure to harmful environments can lead to the excessive production of reactive oxygen species (ROS), thereby resulting in a state of oxidative stress. Oxidative stress can affect the skin in many ways, including signs of premature aging, such as wrinkles, fine lines, and uneven skin tone.

[0003] Heme oxygenase 1 (encoded by the HMOX1 gene) is known to play a protective role against oxidative stress. It is an enzyme that catalyzes the breakdown of heme into biliverdin, a precursor to bilirubin, a potent antioxidant metabolite with antioxidant and anti-inflammatory properties. Primarily functioning in heme catabolism, heme oxygenase 1 is a crucial cytoprotective enzyme with antioxidant, anti-inflammatory, and anti-apoptotic properties.

[0004] Stimulating the HMOX1 gene stimulates heme breakdown, which in turn provides anti-aging, antioxidant, and anti-inflammatory benefits. Human skin is also a site where inflammation, oxidative stress, and cell damage occur regularly.

[0005] Therefore, the inventors recognized the need to develop a personal care composition that can improve the expression of the HMOX1 gene, and developed a personal care composition containing moringa oil and coconut oil. Surprisingly, they found that when the weight ratio of moringa oil to coconut oil is at least 1.4:1, the expression of the HMOX1 gene can be significantly upregulated. [Overview of the project]

[0008] In a third aspect, the present invention relates to the use of the compositions of the present invention to provide antioxidant, anti-inflammatory, and anti-apoptotic benefits; to reduce oxidative stress; and / or to upregulate the expression of the heme oxygenase 1 gene.

[0009] All other aspects of the present invention will become more readily apparent upon consideration of the following detailed description and examples. [Modes for carrying out the invention]

[0045] [Examples] [Example 1] This example demonstrates the preparation of a personal care composition containing moringa seed oil and coconut oil. [Table 1]

[0046] A series of personal care compositions were prepared according to Table 1 using the following standard procedure.

[0047] [Example 2] This example demonstrates the ability of various personal care compositions to upregulate HMOX1 gene expression.

[0048] ArchSkin living skin equivalent (LSE) is a 3D reconstructed human epidermal model using Chinese human keratinocytes supplied by Archgene (Shanghai, China) and cultured at a gas-liquid interface in a prescribed serum-free medium. Squalene monohydroperoxide (SQOOH), a stress surrogate, was prepared in-house by exposing squalene to sunlight irradiation in the presence of a sensitizer.

[0049] The sample was diluted in phosphate-buffered saline (PBS) in a 1:1 volume ratio. SQOOH was diluted with squalene to a 1% by weight concentration. 10 μL of diluted SQOOH was applied topically to the LSE and left for 30 seconds. The LSE was then treated with 10 μL of the diluted sample for another 30 seconds and gently rinsed with 20 mL of PBS. After 24 hours, tissue was collected for gene expression analysis.

[0050] The extracted RNA was quantified using a Nanodrop spectrometer (Thermo Fisher Scientific, Waltham, MA, USA) and reverse transcribed to generate template cDNA using Evo M-MLV RT Premix for qPCR (Accurate Biotechnology, catalog: AG11706) according to the manufacturer's protocol. HMOX1 gene expression was analyzed by polymerase chain reaction (PCR) using the SYBR® Green Premix Pro Taq HS qPCR Kit (Accurate Biotechnology, AG11701). The actin beta (ACTB) gene was selected as a housekeeping gene, and all data regarding the relative expression of the target gene were normalized relative to the ACTB gene. The fold change in expression was calculated compared to a blank control (culture medium without the active substance). All tests were performed at least three times, and the fold changes in HMOX1 gene expression are shown in Table 1.

[0051] The results are summarized in Table 2. [Table 2]

[0052] The data in Table 1 clearly shows that samples 1, 2, and 3 can significantly increase HMOX1 gene expression more than samples A, B, or C. Sample 1 is significantly superior to sample 3 in increasing HMOX1 gene expression, and sample 2 is significantly superior to both samples 1 and 3 in increasing HMOX1 gene expression.
