Abstract
Description
TECHNICAL FIELD
[0001] (CROSS-REFERENCE TO RELATED APPLICATIONS) This application claims the benefit of U.S. Provisional Patent Application No. 63 / 588,030 filed on October 5, 2023, the entire disclosure of which is incorporated herein by reference for all purposes.
[0002] (FIELD OF THE INVENTION) The present invention relates generally to skincare compositions. In particular, the present invention relates to a skincare composition comprising a myrrh resin extract and a retinoid. The present invention also relates to a method of treating skin using the above composition.
BACKGROUND ART
[0003] Human skin undergoes certain aging processes, some of which are attributed to intrinsic processes (e.g., chronological aging) and some to extrinsic factors (e.g., photoaging). In addition, temporary or even persistent changes to the skin, such as acne, oily or dry skin, keratosis, rosacea, photosensitivity, inflammatory, erythematous, and allergic or autoimmune reactive reactions, for example, dermatitis and photodermatosis, may occur.
[0004] Consequences of the aforementioned aging processes may include skin thinning, weakening of the interlacement of the epidermis and dermis, as well as a reduction in the number of cells and the blood vessels that supply them. These consequences are often undesirable, and individuals suffering from these issues turn to topical treatments to address them.
[0005] Retinoids have been used to treat skin conditions caused by intrinsic aging, extrinsic factors, acne or skin disorders. However, retinoids can be difficult to formulate, as many are susceptible to photodegradation, and retinoids may be overly irritating to the skin of some individuals.
[0006] Myrrh resin is a natural extract from the Commiphora genus of trees. Its uses in perfumes and medicines, such as as a disinfectant, have been known since ancient times. Topical application of myrrh resin to the skin can cause skin irritation.
[0007] One option to avoid irritation caused by retinoids on the skin is to suggest alternative compounds that activate the same or similar biological pathways as retinoids. However, this is a complex task because identifying such alternative compounds is difficult. Another option is to enhance or increase the beneficial effects of retinoids and thus limit the amount of retinoid applied topically. However, this task is also complex because identifying these compounds is difficult.
[0008] There is a continuing need and interest in identifying compounds or products that can enhance or augment the activation of retinoids to beneficial biological pathways without increasing the undesirable side effects of retinoids, such as skin irritation. Improving the potential of retinoids using known, reliable natural ingredients is particularly desirable. [Overview of the project] [Means for solving the problem]
[0009] One aspect of the present invention relates to a skincare composition comprising myrrh resin extract and a retinoid.
[0010] This embodiment can be combined with various embodiments and in any combination. Therefore, in some embodiments, the myrrh resin extract is produced from the resin of a plant of the genus Commiphora. In one or more embodiments, the myrrh resin extract is obtained by extraction in an organic solvent. In some embodiments, the myrrh resin extract is present in an amount ranging from about 0.000001% to about 10% of the total weight of the composition. In one or more embodiments, the retinoid is selected from the group consisting of retinol, retinal, retinol esters, retinoic acid, their derivatives, and combinations thereof. In one or more embodiments, the retinoid is present in an amount ranging from about 0.001% to about 10% of the total weight of the composition. In one or more embodiments, the ratio of myrrh resin to retinoid is about 100:1 to about 1:10. In some embodiments, the skincare composition further comprises components selected from the group consisting of surfactants, chelating agents, emollients, moisturizers, conditioners, preservatives, opacifiers, fragrances, and combinations of two or more of these. In one or more embodiments, the skincare composition is in the form of a solution, suspension, emulsion, lotion, cream, serum, gel, stick, spray, ointment, liquid cleanser, bar soap, shampoo, hair conditioner, paste, foam, powder, mousse, shaving cream, hydrogel, or film-forming product.
[0011] Another aspect of the present invention relates to a skincare composition comprising about 0.01% to about 2.0% by weight of myrrh resin extract and about 0.01% to about 1.0% by weight of retinol, based on the total weight of the composition.
[0012] Another aspect of the present invention relates to a method for treating skin, comprising topically applying a composition according to any of the above embodiments to the skin.
[0013] Another aspect of the present invention relates to a method for treating signs of skin aging, comprising topically applying a skincare composition according to any of the above embodiments to skin that requires treatment for skin aging.
[0014] Another aspect of the present invention relates to a method for enhancing the anti-aging effect of retinoids in the skin layers of the skin where it is needed, and comprising topically applying a skincare composition comprising myrrh resin extract and retinoids to the skin.
[0015] Another aspect of the present invention relates to a method for increasing CRABP2 or AQP3 expression in skin cells, comprising topically applying a skincare composition comprising myrrh resin extract and a retinoid to the skin.
[0016] This embodiment can be combined with various embodiments and in any combination. Therefore, in some embodiments of the methods disclosed above, the retinoid is selected from the group consisting of retinol, retinal, retinol esters, retinoic acid, their derivatives, and combinations thereof. In one or more embodiments, retinol is present in an amount ranging from about 0.001% to about 10% of the total weight of the composition.
[0017] In one or more embodiments, the myrrh resin extract is present in an amount ranging from about 0.001% to about 10% of the total weight of the composition.
[0018] In one or more embodiments, a skincare composition applied topically to the skin comprises about 0.01% to about 2.0% by weight of myrrh resin extract and about 0.01% to about 1.0% by weight of retinol, based on the total weight of the composition.
[0019] These and other features and advantages of the present invention will be readily apparent from the following "Modes for Carrying Out the Invention". [Modes for carrying out the invention]
[0020] As used herein, a skincare product or skincare composition is a product that can be applied to the user's skin or hair and provides benefits to that user. These benefits may include, for example, hydration of the skin or hair, or delivery of one or more therapeutic or beneficial agents to the skin or hair, as described in more detail below.
[0021] To provide a more accurate explanation, some of the quantitative expressions presented herein are not qualified with the term “approximately”. Whether the term “approximately” is explicitly used or not, all quantities given herein are meant to refer to actual given values, and also to approximate values of such given values that are reasonably estimated on the basis of ordinary skill in the art, including approximations of such given values by experimental and / or measurement conditions. General conventions in scientific and technical literature apply: the last decimal place of a number indicates its precision. Where no other margin of error is given, the maximum margin is confirmed by applying the convention of rounding to the last decimal place; for example, for a measurement of 3.5%, the margin of error is 3.45 to 3.54.
[0022] All percentages (%) described herein are by weight unless otherwise specified.
[0023] One aspect of the present invention is, Myrrh resin extract, This invention relates to a skincare composition containing retinoids.
[0024] Surprisingly, this combination of myrrh resin extract and retinoids has been found to provide increased CRABP2 or AQP3 expression in skin cells without the drawbacks, such as skin irritation, conventionally associated with natural extracts such as retinoids. This increase in CRABP2 or AQP3 expression is thought to have beneficial effects on the skin. For example, it can be used to reduce signs of skin aging or to increase skin hydration.
[0025] "Extract" means a substance or a mixture of substances that is removed from myrrh resin by a chemical process. [0111] The present invention will be better understood through the following embodiments, which are essentially illustrative and not intended to limit to any specific combination of elements. [Examples]
[0112] method EpiDerm Full Thickness Skin 3D Equivalents (EFT-400) were purchased from MatTek Corporation (Ashland, MA 01721, lot number 34272, kit N and kit P). Upon receipt, the skin equivalents were transferred to new 6-well plates (MatTek Corporation, Ashland, MA 01721) containing 2.5 mL of EFT-400 medium (MatTek Corporation, Ashland, MA) and equilibrated overnight.
[0113] The following day, skin equivalents were topically treated with 4 μL of the following: vehicle, 0.04% retinol, 0.04% retinol + 0.05% myrrh resin extract, or 0.04% retinol + 0.1% myrrh resin extract.
[0114] Propylene glycol / ethanol (PgETOH) (3:7) (w / w) was used as the vehicle. Retinol (BASF 50C) was diluted in PgETOH to a 0.4% stock concentration. The 0.4% retinol was further diluted to prepare 0.08% retinol. Since retinol is 50% active, the percentage of active retinol in the final treatment solution was 0.04% retinol. Myrrh resin extract (Firmenich) was weighed to 16.1 mg and diluted in 1.6 mL of PgETOH for a 1% myrrh resin extract stock solution.
[0115] To prepare a 0.04% retinol + 0.1% myrrh resin extract treatment solution, combine 100 μL of 0.4% retinol stock solution, 50 μL of 1% myrrh resin extract stock solution, and 350 μL of PgETOH. To prepare a 0.04% retinol + 0.05% myrrh resin extract treatment solution, combine 100 μL of 0.4% retinol stock solution, 25 μL of 1% myrrh resin extract stock solution, and 375 μL of PgETOH.
[0116] After 24 hours of treatment, the EFT-400 skin equivalent was excised from the insert and dissolved in 600 μL of lysis buffer consisting of 100 parts of RLT buffer (RNA purification kit, trademark RNEASY Mini kit, Qiagen, Valencia, CA) and 1 part of 2-mercaptoethanol in a reinforced tube with a screw cap and O-ring seal. Tissue grinding beads (trademark PRECELLYS CKMix50, Bertin Corp, Rockville, MD) were also added to the tube. The tube was shaken for 4 cycles at 7200 RPM for 30 seconds each, with a 40-second pause between cycles. RNA was extracted from the solution using the RNEASY Mini Kit (Qiagen, Valencia, CA) according to the manufacturer's instructions, and the RNA was eluted in 30 μL of RNase-free water. Reverse transcription (RT) was performed using the Invitrogen SuperScript IV ViLo kit (ThermoFisher Scientific, Bridgewater, NJ).
[0117] Gene expression was performed using gene expression assays sold under the brand name TAQMAN for cellular retinoic acid-binding protein 2 (CRABP2), aquaporin-3 (AQP3), interleukin-8 (IL8), and polymerase (RNA) II polypeptide A (POLR2A), as well as using the Fast Advanced Master Mix (ThermoFisher Scientific, Bridgewater, NJ), also sold under the brand name TAQMAN. qPCR analysis was performed using the TAQMAN Fast Advanced Master Mix on a real-time PCR system sold under the brand name QUANTSTUDIO 7 Flex System (ThermoFisher Scientific, Bridgewater, NJ).
[0118] The expression of these genes was normalized against the expression of the human POLR2A "housekeeping" gene. Because this "housekeeping" gene is invariant in expression across tissues, cells, and experimental procedures, it is widely used as a control for normalizing specific gene expression levels. The normalization method involves measuring the expression of an internal reference gene or the "housekeeping" gene, taking into account potential errors in RNA / cDNA loading and variations in reverse transcription efficiency.
[0119] The magnification change was calculated by comparing it with the vehicle control, and a two-sided, two-sample Student's t-test (Microsoft Office Excel 2007, Microsoft, Redmond, WA, USA) was performed.
[0120] The results are shown in Tables 1-3 as the average magnification change relative to the vehicle.
[0121] "Untreated" means that the gene expression assay was performed using a vehicle:propylene glycol / ethanol (PgETOH) that does not contain retinol or myrrh resin extract.

[0124] [Table 2]
[0125] [Table 3]
[0126] Conclusion: CRABP2 is well-established in the literature as a highly sensitive marker for the biological activity and potency of retinoids (Elder JT, Cromie MA, Griffiths CEM, Chambon P, Voorhees JJ. Stimulus-selective induction of CRABP-II mRNA: A marker for retinoic acid action in human skin. J Invest Dermatol. 1993;100(4)), and is associated with the conferral of anti-aging benefits for the skin (Bielli A, Scioli MG, D'Amico F, Tarquini C, Agostinelli S, Costanza G, Doldo E, Campione E, Passeri D, Coniglione F, Orlandi A. Cellular retinoic acid binding protein-II expression and its potential role in skin aging. Aging (Albany NY). 2019 Mar 18;11(6):1619-1632).
[0127] Surprisingly, the combination of 0.04% retinol and 0.05% myrrh resin extract showed significantly higher CRABP2 induction than retinol alone.
[0128] This data indicates that the combination of retinol and myrrh resin extract results in increased retinol-like properties and therefore anti-aging benefits. Each biomarker is associated with anti-aging benefits, and such additional benefits from the combination are highly advantageous.
[0129] Alongside the increase in CRABP2 induction, no increase in IL8 was observed in most combinations. Retinoids are known to cause irritation and stinging upon application, which is considered IL8 induction in vitro. Our results show that the combinations resulted in increased retinol-like activity without the associated inflammatory side effects.
[0130] Aquaporin 3 (AQP3) is a passive water transporter involved in skin hydration and important for maintaining water homeostasis in the skin. AQP3 is indirectly induced by retinol. The above results demonstrate that the combination of retinol and myrrh resin extract shows a significantly increased induction of AQP3 compared to retinol alone. Our results show that this combination exhibits an increased benefit in skin hydration.
[0131] It has been observed that the active dose of compounds that provide in vitro benefits, such as myrrh resin extract or retinol, may generally require a 10-fold increase in vivo to exhibit similar benefits.
