Abstract
Description
[Technical Field]
[0001] This technology generally relates to the use of rhamnolipids to improve and / or promote the maintenance of a healthy scalp. More specifically, this technology relates to compositions and methods for treating and / or preventing the dandruff cycle by applying compositions comprising specific mixtures of rhamnolipids to the human scalp. In certain embodiments, application of the compositions described herein, according to the methods and regimens described herein, inhibits the growth, colonization, and / or reduces the viability of skin-causing microorganisms, such as the lipophilic yeast Malassezia. [Background technology]
[0002] The human scalp is typically characterized by the development of dense terminal hairs and numerous sebaceous glands and microorganisms. This environment contributes to the establishment of specific microenvironments involved in the development of skin pathological conditions, including, for example, seborrheic dermatitis and dandruff, psoriasis, atopic dermatitis, alopecia, and scalp aging.
[0003] Seborrheic dermatitis is a common chronic inflammatory skin disease affecting the scalp, characterized by erythema and oily scales (scaly rash). The non-inflammatory form of this disease affects approximately 50% of the world's population (with dry and desquamating scalp symptoms) and is commonly known as dandruff.
[0004] The dandruff cycle is initiated by an imbalance of endogenous factors, including host immunity, skin microbiome, sebaceous gland activity, and / or epidermal barrier damage. One or more of these conditions can create favorable conditions that lead to Malassezia infection. Malassezia fungi are lipophilic yeasts that account for over 80% of the fungal population found on human skin.
[0005] Malassezia furfur is the primary causative fungal species of dandruff and its associated symptoms and problems. Other species include M. dermatis, M. equi, M. globosa, M. japonica, M. obtusa, M. ovalis (also known as Pityrosporum ovale), M. pachydermatis, M. restricta, M. slooffiae, M. sympodialis, or combinations thereof. Once an infection is established, seborrheic dermatitis develops, leading to damage to the epidermal barrier function, increased sebum secretion, further colonization of Malassezia fungi (and secretion of metabolites), and ultimately an amplification of the inflammatory response. This results in a long-term, sometimes chronic, decline in scalp health and aesthetics. Other conditions such as psoriasis, atopic dermatitis, and accelerated scalp aging have also been reported to be associated with skin damage, oxidative stress, microbial infection, microbiome imbalance, and immunological response mechanisms.
[0006] Current market solutions involve the use of highly irritating and less selective chemical components such as coal tar, zinc pyrithione, selenium sulfide, salicylic acid, or a limited range of antifungal agents, which lose effectiveness over time. One limitation of these conventional chemicals is that while they are effective in suppressing fungal infections and / or removing dandruff from the scalp by exfoliation for aesthetic purposes, these chemicals fail to provide a comprehensive, sustainable, and natural solution to the dandruff cycle.
[0007] Therefore, there is an ongoing need for new compositions, methods, and treatment regimens that promote the maintenance of a healthy scalp. More specifically, there is a need for compositions and methods for treating and / or preventing the dandruff cycle, particularly for inhibiting or preventing the proliferation, colonization, and / or reducing the survival rate of the lipophilic yeast Malassezia.
[0008] Furthermore, formulating products with ingredients based on renewable raw materials is a recent trend. These ingredients are considered "environmentally friendly" or "natural" because they originate from renewable and / or sustainable sources. As a result, they are more environmentally friendly than ingredients derived from fossil fuels or other non-renewable sources. Ingredients with a high Bio-Renewable Carbon Index (BCI), such as over 80, indicate that the ingredient contains carbon primarily from plant, animal, or marine-based sources.
[0009] Rhamnolipid is a surfactant glycolipid produced by various bacterial species and can be prepared by fermentation based on recycled raw materials, making it an example of an "environmentally friendly" ingredient. It would be desirable to provide compositions and methods of use containing active ingredients derived from renewable sources such as rhamnolipid that can be used to promote the maintenance of a healthy scalp. Providing such compositions containing naturally derived rhamnolipid would help achieve sustainability goals by ensuring sustainable consumption through the use of bio-based antimicrobial materials.
[0010] The applicant determined that certain rhamnolipid compositions can satisfy the above objectives while also promoting the UN Sustainable Development Goals ("SDGs"). The rhamnolipid compositions of this technology contribute to better health and well-being by providing equivalent or better efficacy in maintaining a healthy scalp (e.g., in preventing and / or treating dandruff). Advantageously, the rhamnolipid compositions described herein are bio-based, renewablely sourced active materials obtained from a bacterial fermentation process that generates biodegradable waste with minimal environmental impact. These advantages further enhance SDG Goal 3 (Good Health and Well-being) and SDG Goal 12 (Responsible Consumption and Production). [Overview of the Initiative]

The methods disclosed in the present specification include applying the composition to the scalp of a mammal, the composition comprises at least one rhamnolipid, and applying the composition to the scalp of the mammal inhibits or reduces the growth, colonization and / or viability of Malassezia fungi as well as the progression in the dandruff cycle.
[0023] As defined in the present specification, "rhamnolipid" is a glycolipid having a lipid moiety comprising one or more, typically linear, saturated or unsaturated B-hydroxy-carboxylic acid moieties and a sugar moiety of one or more rhamnose units. The term Zn-rhamnolipid or zinc rhamnolipid is a salt of the rhamnolipid described in the present specification, and the salt is zinc.
[0024] The sugar moiety and the lipid moiety are linked via a B-glycosidic bond between the 1-OH group of the rhamnose moiety of the sugar moiety and the 3-OH group of the B-hydroxy-carboxylic acid of the lipid moiety. Thus, the carboxylic acid of one carboxylic acid moiety defines the terminus of the rhamnolipid. When two or more rhamnose moieties are contained in the rhamnolipid, each of the rhamnose moieties not linked to the lipid moiety is linked to another rhamnose moiety via a 1,4B-glycosidic bond. In embodiments where two or more B-hydroxy-carboxylic acids are present in the rhamnolipid, the B-hydroxy-carboxylic acid moieties are selected independently of each other. In some embodiments, the B-hydroxycarboxylic acid moieties may be the same. In some embodiments, they are different from each other.
[0025] The present technology generally relates to compositions for maintaining a healthy scalp, and compositions comprising specific mixtures of rhamnolipids in their salt forms. The rhamnolipids may have the following structure (I):

[0056] The technology and its advantages described herein will be better understood by referring to the following examples. These examples are provided to illustrate specific embodiments of the technology. By providing these examples, the inventors do not limit the scope and spirit of the technology.
[0057] Example 1 - Preparation of a rhamnolipid composition After fermentation, the entire culture containing rhamnolipid undergoes several purification steps. The specific combination of steps depends on the rhamnolipid active ingredient and purity requirements for the target application. The specific rhamnolipid mixtures used herein are purified by a solvent extraction process. As used herein, the acronym SEP-RM refers to Solvent Extraction Purified-Rhamnolipid Mixture (or composition).
[0058] For example, the SEP-RM rhamnolipid compositions used herein are prepared according to at least the following processing steps: (1) fermentation of suitable rhamnolipid-producing bacteria, (2) biomass separation, (3) sterilization, (4) purification (e.g., filtration), (5) acidification, (6) bleaching, (7) washing, (8) solvent extraction, (9) neutralization and dilution, and (10) final finishing steps(s). As discussed below, the solvent extraction process / step is intended to result in a higher purity rhamnolipid mixture / composition for personal care applications, as described herein. Additional solvent extraction steps are required, for example, to achieve a lighter color and milder odor profile required for the personal care market.
[0059] At the end of fermentation, the total culture typically contains rhamnolipid along with fermentation biomass and other by-products. To separate the biomass solids, the culture can be centrifuged. The resulting centrifuged culture is then subjected to sterilization (e.g., high-temperature sterilization), and subsequently, the centrifuged and sterilized culture is purified by filtration to remove suspended solids.
[0060] Further purification is achieved by treating the purified culture medium with acid, which converts the rhamnolipid into a water-insoluble form that settles at the bottom and separates from the bulk aqueous phase. This highly concentrated acidified rhamnolipid is then isolated (referred to as acidified concentrated clarified culture medium (ACCB)). The ACCB is then treated with a bleaching agent and subsequently washed with water to remove residual bleaching agent and other water-soluble impurities. At this point, the decolorized and washed ACCB is concentrated (e.g., with approximately 45% or more active ingredients, or approximately 35% to 55% active ingredients) and purer (e.g., with a purity of approximately 75% or more, or approximately 65% to 85% purity).
[0061] Solvent Extraction Process – To achieve better purity, color, and odor, the decolorized acidified concentrated purified culture medium (ACCB) undergoes a solvent extraction process. This is generally carried out by dissolving the decolorized and washed ACCB in an organic solvent, preferably ethyl acetate. The typically formed lag layer is separated from the bulk solution. Activated carbon is then added to the rhamnolipid solution in ethyl acetate for further decolorization and deodorization. The slurry is filtered, and the resulting solution is stripped under vacuum to remove the ethyl acetate. The highly concentrated ACCB obtained as a residue is then neutralized and diluted (e.g., to about 20% to about 30% active ingredients, preferably about 25% active ingredients) to obtain crude SEP-RM. As a final purification step, the crude SEP-RM is washed with ethyl acetate to extract the defoamer, and the final product is optically clear SEP-RM with about 25% active ingredients and about 85% purity.
[0062] Rhamnolipid, a zinc salt, was prepared by adding an equimolar amount of ZnCl2 to the active component of rhamnolipid (such as SEP-RM) in an aqueous solution and mixing at ambient temperature for approximately 20 minutes. The solution was then centrifuged to separate it into layers. The upper layer was decanted, and the lower layer was washed with approximately 20 g of DI water and centrifuged. The washing procedure was repeated, the lower layer was collected, and dried in a vacuum oven at 50°C to obtain Zn-rhamnolipid salt.
[0063] Monoramnolipid:diramnolipid ratio and congener distribution The monorhamnolipid:diramnolipid ratio and congener distribution of the SEP-RM rhamnolipid composition of this disclosure can be determined using HPLC (high-performance liquid chromatography). As will be understood by those skilled in the art, HPLC separates rhamnolipid congeners by chain length and the number of rhamnose moieties (more polar congeners elute first). Fractions are collected and analyzed by mass spectrometry to identify each peak. The congener distribution is reported as an area percentage (the active ingredient is calculated in comparison to a known standard).

Malassezia versicolor was supplied to each set of wells using well-grown and prepared cultures at a level of 2.5E+05 cfu / ml. Resazurin, a blue fluorescent dye used as a redox indicator in cell viability and growth assays, was added to each sample and control well. The mixture was kept at 20–25°C and thoroughly mixed. The plate was then incubated at 30±2°C for up to 3 days until mature biological activity was detected from the control, first visually (resazurin changes color upon metabolic treatment) and then quantitatively (by using a fluorescence-compatible plate reader). The tests were performed using 0.3x concentration Modified Dixon (MD), which is related to the scalp and is a nutrient matrix favored by microorganisms.
[0087] As described below, the rhamnolipid prepared above (SEP-RM / Lot B, having 47.6% mono-RL and 52.4% di-RL) demonstrated efficacy at 1% (w / w based on the total weight of the composition) (47% inhibition against a 2.5E+05 cfu / mL M. furfur challenge). The SEP-RM / Lot B rhamnolipid performed significantly better than the comparative sample (a commercially available rhamnolipid product with a mono:di-rhamnolipid ratio of 4:96), which showed only 19% inhibition (against a 2.5E+05 cfu / mL M. furfur challenge). When the co-compound was added to 1% SEP-RM / lot B (i.e., the sample described above) containing 47.6% mono-RL and 52.4% di-RL, the performance increased to 100% inhibition, which is the maximum detectable value (for a M. furfur challenge of 2.5E+05 cfu / ml).
[0088] The test results are shown below. [Table 19]
[0089] Domestic brand anti-dandruff products were also tested according to the protocol described above (this example and Example 3). As shown below, the efficacy of the SEP-RM rhamnolipid formulation (mentioned above) is consistent with the performance of three domestic brand anti-dandruff products (0.3x concentration) and quaternary ammonium compounds (1839-220) at 0.3x RTU. [Table 20]
[0090] Example 5: Zn-rhamnolipid activity To evaluate the efficacy of the Zn-rhamnolipid compositions disclosed herein against M. furfur, the 96-well plate liquid culture method described in Example 3 was used. The test samples contained a mixture of rhamnolipids prepared using solvent extraction as described above and converted to Zn-rhamnolipid salts. The Zn-rhamnolipid mixture was added as an active ingredient at a concentration of 1.0% to either water or a base shampoo composition containing 10% by weight of sodium lauryl sulfate, 3% by weight of cocamidopropyl betaine, and 3% by weight of sodium laureth sulfate, and a total of 100% by weight of water as active ingredients. Comparative samples were prepared from a domestic brand (H&S) dandruff shampoo product containing 1% zinc pyrithione, as well as from Zn-rhamnolipid samples prepared from commercially available rhamnolipid compositions containing approximately 3% monorhamnolipid and 97% dirhamnolipid converted to Zn-rhamnolipid salts as described above. Water alone, the base composition alone, and an aqueous zinc pyrithione solution were used as controls. The sample dilutions and test results are shown below. [Table 21]
[0091] Example 6 - Zn-rhamnolipid combined with an additional antifungal agent The efficacy of Zn-rhamnolipid in combination with different antifungal active ingredients in water or base shampoo compositions described in Example 5 was evaluated using the 96-well plate liquid culture method described in Example 3. The antifungal active ingredients tested were salicylic acid and zinc pyrithione. The active ingredient concentrations and test results of the tested samples are shown below. [Table 22]
