Description

[Technical field]

[0001] The present invention relates to novel esterases, in particular esterases with improved activity and / or improved thermostability compared to parent esterases. The present invention also relates to the use of said novel esterases for degrading polyester-containing materials, such as plastic products. The esterases of the present invention are particularly suitable for degrading polyethylene terephthalate and polyethylene terephthalate-containing materials.

[0002] background Esterases are capable of catalyzing the hydrolysis of various polymers, including polyesters. In this context, esterases have shown promising effects in many industrial applications, including as detergents for dishwashing and laundry applications, as degradative enzymes for processing biomass and food, as biocatalysts in the detoxification of environmental pollutants, or for the treatment of polyester fabrics in the textile industry. The use of esterases as degradative enzymes for the hydrolysis of polyethylene terephthalate (PET) is particularly interesting. Indeed, PET is used in many technical fields, such as in the manufacture of clothing, carpets, or in the form of thermosetting resins for the manufacture of packaging or automotive plastics, etc. As a result, the accumulation of PET in landfill sites is becoming an increasingly ecological problem.

[0003] Enzymatic degradation of polyesters, especially PET, is considered an interesting solution to reduce the accumulation of plastic waste. Indeed, enzymes can accelerate the hydrolysis of polyester-containing materials, especially plastic products, even down to the monomer level. Moreover, the hydrolyzates (i.e., monomers and oligomers) can be reused as materials for synthesizing new polymers.

[0004] In this context, several esterases have been identified as potential degradative enzymes for polyesters, and some mutants of such esterases have been developed. Among the esterases, cutinases (also known as cutin hydrolases) (EC 3.1.1.74) are of particular interest. Cutinases have been identified from various fungi (PE Kolattukudy in "Lipases", Ed. B. Borg-strom and HL Brockman, Elsevier 1984, 471-504), bacteria and plant pollen. Recently, metagenomics approaches have identified further esterases.

[0005] However, there remains a need for esterases with improved activity and / or improved thermostability compared to known esterases to make the polyester degradation process more efficient and therefore more competitive.

[0006] Summary of the Invention The present invention provides novel esterases that exhibit improved activity and / or improved thermostability compared to the parent or wild-type esterase having the amino acid sequence shown in SEQ ID NO: 1. The wild-type esterase corresponds to amino acids 36-293 of the amino acid sequence of the metagenomic cutinase described in Sulaiman et al., Appl Environ Microbiol. 2012 Mar. and referenced in SwissProt as G9BY57. The esterases of the present invention are particularly useful in methods for degrading plastic products, especially plastic products containing PET.

The term "cutinase" or "cutin hydrolase" refers to an esterase classified under EC 3.1.1.74 according to enzyme nomenclature, which is capable of catalyzing the chemical reaction of the production of cutin monomers from cutin and water.

[0021] The term "wild-type protein" or "parent protein" refers to the non-mutated version of a naturally occurring polypeptide. In the present case, parent esterase refers to the esterase having the amino acid sequence shown in SEQ ID NO:1.

[0022] The terms "mutant" and "variant" refer to a polypeptide derived from SEQ ID NO:1 and containing at least one modification or alteration, i.e., substitution, insertion and / or deletion, at one or more (e.g., multiple) positions, and having polyesterolytic activity. Mutants can be obtained by various techniques well known in the art. In particular, examples of techniques for altering a DNA sequence encoding a wild-type protein include, but are not limited to, site-directed mutagenesis, random mutagenesis and synthetic oligonucleotide construction. Thus, as used herein, the terms "modified" and "altered" with respect to a particular position means that the amino acid at this particular position has been modified compared to the amino acid at this particular position in the wild-type protein.

[0023] "Substitution" means that an amino acid residue is replaced by another amino acid residue. Preferably, the term "substitution" refers to the replacement of an amino acid residue by another amino acid residue selected from the 20 naturally occurring standard amino acid residues, amino acid residues that are rarely found in nature (e.g., hydroxyproline, hydroxylysine, allohydroxylysine, 6-N-methyllysine, N-ethylglycine, N-methylglycine, N-ethylasparagine, alloisoleucine, N-methylisoleucine, N-methylvaline, pyroglutamine, aminobutyric acid, ornithine, norleucine, norvaline) and non-natural amino acid residues that are often synthesized (e.g., cyclohexylalanine). Preferably, the term "substitution" refers to the replacement of an amino acid residue by another amino acid residue selected from the 20 naturally occurring standard amino acid residues (G, P, A, V, L, I, M, C, F, Y, W, H, K, R, Q, N, E, D, S and T). The sign "+" indicates a combination of substitutions. In this specification, the following terminology is used to designate substitutions: L82A refers to the substitution of the amino acid residue at position 82 of the parent sequence (leucine, L) with alanine (A). A121V / I / M refers to the substitution of the amino acid residue at position 121 of the parent sequence (alanine, A) with one of the following amino acids: valine (V), isoleucine (I) or methionine (M). Substitutions can be conservative or non-conservative. Examples of conservative substitutions are within the groups of basic amino acids (arginine, lysine and histidine), acidic amino acids (glutamic acid and aspartic acid), polar amino acids (glutamine, asparagine and threonine), hydrophobic amino acids (methionine, leucine, isoleucine, cysteine and valine), aromatic amino acids (phenylalanine, tryptophan and tyrosine) and small amino acids (glycine, alanine and serine).

[0024] Unless otherwise specified, positions disclosed herein are numbered with reference to the amino acid sequence shown in SEQ ID NO:1.

[0025] As used herein, the term "sequence identity" or "identity" refers to the number (or fraction expressed as a percentage) of matches (identical amino acid residues) between two polypeptide sequences. Sequence identity is determined by comparing sequences when aligned to maximize overlap and identity while minimizing sequence gaps. In particular, sequence identity can be determined using any of a number of mathematical global or local alignment algorithms, depending on the length of the two sequences. Sequences of similar length are preferably aligned using a global alignment algorithm (e.g., the Needleman and Wunsch algorithm; Needleman and Wunsch, 1970), which optimally aligns the sequences over their entire length, while sequences of substantially different lengths are preferably aligned using a local alignment algorithm (e.g., the Smith and Waterman algorithm (Smith and Waterman, 1981) or the Altschul algorithm (Altschul et al., 1997; Altschul et al., 2005)). Alignment for purposes of determining % amino acid sequence identity can be accomplished in a variety of ways that are within the skill of the art, for example, using publicly available computer software available on internet websites such as http: / / blast.ncbi.nlm.nih.gov / or http: / / www.ebi.ac.uk / Tools / emboss / . Those skilled in the art can determine appropriate parameters for measuring alignment, including any algorithms needed to achieve maximal alignment over the full length of the sequences being compared. For purposes herein, % amino acid sequence identity values refer to values generated using the pairwise sequence alignment program EMBOSS Needle, which uses the Needleman-Wunsch algorithm to generate an optimal global alignment of two sequences, where all search parameters are set to default values, i.e., scoring matrix=BLOSUM62, gap open=10, gap extend=0.5, end gap penalty=false, end gap open=10 and end gap extend=0.5.

[0026] "Polymer" refers to a compound or mixture of compounds whose structure is composed of multiple monomers (repeating units) linked by covalent chemical bonds. In the context of the present invention, the term polymer includes natural or synthetic polymers composed of a single type of repeating unit (i.e., homopolymers) or a mixture of different repeating units (i.e., copolymers or heteropolymers). According to the present invention, "oligomer" refers to a molecule containing from 2 to about 20 monomers.

[0027] In the context of the present invention, "polyester-containing material" or "polyester-containing product" refers to a product, such as a plastic product, that contains at least one polyester in crystalline, semi-crystalline or completely amorphous form. In a particular embodiment, polyester-containing material refers to any item made from at least one plastic material, such as plastic sheets, tubes, rods, profiles, shapes, films, bulk blocks, etc. These contain at least one polyester and, optionally, other substances or additives, such as plasticizers, mineral or organic fillers. In another particular embodiment, polyester-containing material refers to a molten or solid plastic compound or plastic blend suitable for making a plastic product. In another particular embodiment, polyester-containing material refers to a textile, fabric or fiber that contains at least one polyester. In another particular embodiment, polyester-containing material refers to a plastic waste or fiber waste that contains at least one polyester.

[0028] In this description, the term "polyester" includes, but is not limited to, polyethylene terephthalate (PET), polytrimethylene terephthalate (PTT), polybutylene terephthalate (PBT), polyethylene isosorbide terephthalate (PEIT), polylactic acid (PLA), polyhydroxyalkanoate (PHA), polybutylene succinate (PBS), polybutylene succinate adipate (PBSA), polybutylene adipate terephthalate (PBAT), polyethylene furanoate (PEF), polycaprolactone (PCL), poly(ethylene adipate) (PEA), polyethylene naphthalate (PEN), and blends / mixtures of these polymers.

[0029] Novel esterases The present invention provides novel esterases with improved activity and / or improved thermostability compared to parent esterases. In particular, the inventors have designed novel enzymes that are particularly suitable for use in industrial processes. The esterases of the present invention are particularly suitable for degrading polyesters, especially PET, including PET-containing materials and especially plastic products containing PET. In certain embodiments, the esterases show both improved activity and improved thermostability.

[0030] It is therefore an object of the present invention to provide an esterase that exhibits improved activity compared to the esterase having the amino acid sequence shown in SEQ ID NO:1.

[0031] In particular, the inventors have identified specific amino acid residues in SEQ ID NO:1 that are intended to contact the polymer substrate in the X-ray crystal structure (i.e., the folded 3D structure) of the esterase, which can be advantageously modified to facilitate contact between the substrate and the esterase, increase adsorption of the polymer and / or thereby improve the activity of the esterase towards this polymer.

[0032] In the context of the present invention, the term "enhanced activity" or "enhanced degradation activity" refers to an improved ability of an esterase to degrade polyesters at a given temperature and / or to adsorb to polyesters, compared to the ability of the esterase of SEQ ID NO:1 to degrade the same polyester at the same temperature. In particular, the esterase of the present invention has an improved PET degradation activity. Such an improvement can be at least 10%, preferably at least 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, 100%, 110%, 120%, 130% or more higher than the PET degradation activity of the esterase of SEQ ID NO:1. In particular, the degradation activity is a depolymerization activity resulting in monomers and / or oligomers of polyesters, which can be further recovered and optionally reused.

[0033] The "degradation activity" of an esterase can be evaluated by a person skilled in the art according to methods known to those skilled in the art. For example, the degrading activity can be evaluated by measuring the depolymerization activity rate of a particular polymer, measuring the rate of degrading a solid polymer compound dispersed in an agar plate, or measuring the depolymerization activity rate of a polymer in a reactor. In particular, the degrading activity can be evaluated by measuring the "specific degrading activity" of an esterase. The "specific degrading activity" of an esterase for PET corresponds to the TA / hour of PET μmol / min or mg equivalents generated per mg of esterase in the first period of the reaction (i.e., the first 24 hours), and is determined from the linear part of the hydrolysis curve of the reaction, such curve being set by multiple samplings performed at different times during the first 24 hours. As another example, the "degradation activity" can be evaluated by measuring the percentage of oligomers and / or monomers released after a defined period of time when a polymer or a polymer-containing plastic product is contacted with a degrading enzyme under appropriate conditions of temperature, pH and buffer.

[0034] The ability of an enzyme to adsorb to a substrate can be evaluated by a person skilled in the art according to methods known per se in the art. For example, the ability of an enzyme to adsorb to a substrate can be measured from a solution containing the enzyme, where the enzyme is pre-incubated with the substrate under suitable conditions.

[0035] The inventors have also identified target amino acid residues in SEQ ID NO:1 that can be advantageously modified to improve the stability (i.e., improved thermostability) of the corresponding esterase at elevated temperatures, advantageously above 50°C, preferably above 70°C.

[0036] It is therefore an object of the present invention to provide novel esterases which exhibit improved thermostability compared to that of the esterase having the amino acid sequence shown in SEQ ID NO:1.

[0037] In the context of the present invention, the term "improved thermostability" denotes an improved ability of an esterase to resist changes in its chemical and / or physical structure at elevated temperatures, in particular at temperatures between 50°C and 90°C, compared to the esterase of SEQ ID NO:1.

[0038] In particular, thermostability can be assessed by evaluation of the melting temperature (Tm) of the esterase. In the context of the present invention, "melting temperature" refers to the temperature at which half of the considered enzyme population is unfolded or misfolded. Typically, the esterases of the present invention exhibit an increase in Tm of about 1°C, 2°C, 3°C, 4°C, 5°C, 10°C, 12°C or more compared to the Tm of the esterase of SEQ ID NO:1. In particular, the esterases of the present invention can have an extended half-life at temperatures between 50°C and 90°C compared to the esterase of SEQ ID NO:1.

[0039] The melting temperature (Tm) of an esterase can be measured by a person skilled in the art according to a method known per se in the art. For example, DSF can be used to quantify the change in thermal denaturation temperature of an esterase, thereby determining its Tm. Alternatively, Tm can be evaluated by analyzing protein folding using circular dichroism. Preferably, Tm is measured using DSF or circular dichroism as mentioned in the experimental section. In the context of the present invention, the comparison of Tm is performed by Tm measured under the same conditions (e.g. pH, nature and amount of polyester, etc.).

[0040] Alternatively, thermostability can be assessed by measuring the esterase and / or polyester depolymerization activity of the esterase after incubation at different temperatures and comparing it to the esterase and / or polyester depolymerization activity of the parent esterase. The ability to perform multiple polyester depolymerization assays at different temperatures can also be evaluated. A rapid and valuable test can consist of evaluating the ability of the esterase to degrade solid polyester compounds dispersed in agar plates after incubation at various temperatures by halo diameter measurements.

[0041] It is therefore an object of the present invention to provide an esterase which (i) has the amino acid sequence as set forth in SEQ ID NO:1, (ii) contains 1 to 15 amino acid substitutions selected from the group consisting of A17T, T27S, S48T, L82I, F90L, Y92F, G135A, A140S, N143I, S145T, A149G, S164P, V167Q, S206T, N213P or T252S, wherein the positions are numbered with reference to the amino acid sequence as set forth in SEQ ID NO:1, and (iii) exhibits improved polyesterolytic activity and / or improved thermostability compared to the esterase of SEQ ID NO:1.

[0079] The invention also encompasses a nucleic acid encoding an esterase of the invention, wherein the sequence of said nucleic acid, or at least a part of said sequence, has been engineered using optimized codon usage.

[0080] Alternatively, the nucleic acid of the invention can be deduced from the sequence of the esterase of the invention and the codon usage can be adapted depending on the host cell in which the nucleic acid will be transcribed. These steps can be carried out according to methods well known to those skilled in the art, some of which are described in the reference manual Sambrook et al. (Sambrook et al., 2001).

[0081] The nucleic acids of the invention can further comprise additional nucleic acid sequences, i.e. regulatory sequences, i.e. promoters, enhancers, silencers, terminators, signal peptides, etc., which can be used to cause or regulate expression of the polypeptide in a selected host cell or system.

[0082] Furthermore, the present invention relates to an expression cassette comprising a nucleic acid of the invention operably linked to one or more control sequences which direct the expression of said nucleic acid in a suitable host cell.

[0083] As used herein, the term "expression" refers to any process involved in the production of a polypeptide, including, but not limited to, transcription, post-transcriptional modification, translation, post-translational modification, and secretion.

[0084] The term "expression cassette" refers to a nucleic acid construct that comprises a coding region, ie, a nucleic acid of the invention, and a regulatory region, ie, a region that contains one or more control sequences operably linked thereto.

[0085] Typically, the expression cassette comprises or consists of the nucleic acid of the invention operably linked to a control sequence, such as a transcription promoter and / or a transcription terminator. The control sequence can comprise a promoter recognized by a host cell or an in vitro expression system for the expression of the nucleic acid encoding the esterase of the invention. The promoter contains a transcription control sequence that mediates the expression of the enzyme. The promoter can be any polynucleotide that exhibits transcriptional activity in the host cell, including mutant promoters, truncated promoters and hybrid promoters, and can be obtained from a gene encoding an extracellular or intracellular polypeptide, either homologous or heterologous to the host cell. The control sequence can also be a transcription terminator. The transcription terminator is recognized by the host cell to terminate transcription. The terminator is operably linked to the 3' end of the nucleic acid encoding the esterase. Any terminator that is functional in the host cell can be used in the present invention. Typically, the expression cassette comprises or consists of the nucleic acid of the invention operably linked to a transcription promoter and a transcription terminator.

[0086] The present invention also relates to a vector comprising a nucleic acid or an expression cassette as defined above.

[0087] As used herein, the term "vector" or "expression vector" refers to a DNA or RNA molecule containing an expression cassette of the present invention that is used as a vehicle to transfer recombinant genetic material into a host cell. The main types of vectors are plasmids, bacteriophages, viruses, cosmids and artificial chromosomes. The vector itself is generally a DNA sequence consisting of an insert (heterologous nucleic acid sequence, transgene) and a larger sequence that serves as the "backbone" of the vector. The purpose of the vector to transfer genetic information to the host is typically to isolate, amplify or express the insert in the target cell. Vectors called expression vectors (expression constructs) are specifically adapted for the expression of heterologous sequences in the target cell and generally have a promoter sequence that drives the expression of the heterologous sequence that encodes a polypeptide. Generally, regulatory elements present in expression vectors include a transcription promoter, a ribosome binding site, a terminator and an operator that is optionally present. Preferably, the expression vector also contains an origin of replication for autonomous replication in the host cell, a selection marker, a limited number of useful restriction enzyme sites and the potential for high copy number. Examples of expression vectors are cloning vectors, modified cloning vectors, specifically designed plasmids and viruses. Expression vectors that provide appropriate levels of polypeptide expression in different hosts are well known in the art. The choice of vector will typically depend on the compatibility of the vector with the host cell into which the vector is introduced. Preferably, the expression vector is a linear or circular double-stranded DNA molecule.

[0088] Another object of the invention is to provide a host cell comprising a nucleic acid, an expression cassette or a vector as described above. Thus, the invention relates to the use of a nucleic acid, an expression cassette or a vector of the invention for transforming, transfecting or transducing a host cell. The choice of vector will typically depend on the compatibility of the vector with the host cell into which the vector needs to be introduced.

[0089] According to the invention, the host cell can be transformed, transfected or transduced in a transient or stable manner. The expression cassette or vector of the invention is introduced into the host cell such that the cassette or vector is maintained as a chromosomal integrant or as a self-replicating extrachromosomal vector. The term "host cell" also encompasses any progeny of a parent host cell that is not identical to the parent host cell due to mutations that occur during replication. The host cell can be any cell useful for the production of the variants of the invention, for example, a prokaryotic or eukaryotic organism. Prokaryotic host cells can be either gram-positive or gram-negative bacteria. The host cell can also be a eukaryotic cell, for example, a yeast, fungal, mammalian, insect or plant cell. In a particular embodiment, the host cell is selected from the group of Escherichia coli, Bacillus, Streptomyces, Trichoderma, Aspergillus, Saccharomyces, Pichia, Vibrio or Yarrowia.

[0090] The nucleic acids, expression cassettes or expression vectors of the invention can be introduced into host cells by any method known to those skilled in the art, such as, for example, electroporation, conjugation, transduction, competent cell transformation, protoplast transformation, protoplast fusion, biolistic "gene gun" transformation, PEG-mediated transformation, lipid-assisted transformation or transfection, chemically-mediated transformation, lithium acetate-mediated transformation, liposome-mediated transformation.

[0091] Optionally, more than one copy of a nucleic acid, cassette or vector of the invention can be inserted into the host cell to increase production of the variant.

[0092] In a particular embodiment, the host cell is a recombinant microorganism. Indeed, the present invention allows the engineering of microorganisms with improved ability to degrade polyester-containing materials. For example, the sequences of the present invention can be used to complement wild-type strains of fungi or bacteria known to be capable of degrading polyesters in order to improve and / or enhance the performance of the strains.

[0093] Production of esterase Another object of the invention is to provide a method for producing an esterase of the invention comprising expressing a nucleic acid encoding the esterase and, optionally, recovering the esterase.

[0094] In particular, the present invention relates to an in vitro method for producing an esterase of the present invention comprising (a) contacting a nucleic acid, cassette or vector of the present invention with an in vitro expression system, and (b) recovering the esterase produced. In vitro expression systems are well known to those skilled in the art and are commercially available.

[0095] Preferably, the method of manufacture comprises: (a) culturing a host cell containing a nucleic acid encoding an esterase of the invention under conditions suitable for expression of the nucleic acid; (b) recovering the esterase from the cell culture.

[0096] Advantageously, the host cell is a recombinant Bacillus, a recombinant E. coli, a recombinant Aspergillus, a recombinant Trichoderma, a recombinant Streptomyces, a recombinant Saccharomyces, a recombinant Pichia, a recombinant Vibrio or a recombinant Yarrowia.

[0097] The host cells are cultured in a nutrient medium suitable for production of the polypeptide using methods known in the art. For example, the cells can be cultured in shake flask cultures or by small- or large-scale fermentation (including continuous, batch, fed-batch, or solid-phase fermentation) in laboratory or industrial fermenters conducted in suitable media under conditions that allow the enzyme to be expressed and / or isolated. Cultivation is carried out in suitable nutrient media prepared from commercial sources or according to published compositions (e.g., catalogues of the American Type Culture Collection).

[0098] If the esterase is secreted into the nutrient medium, it can be directly recovered from the culture supernatant. Conversely, it can be recovered from the cell lysate or after permeabilization. The esterase can be recovered using any method known in the art. For example, the esterase can be recovered from the nutrient medium by conventional techniques. These techniques include, but are not limited to, recovery, centrifugation, filtration, extraction, spray drying, evaporation or precipitation. Optionally, the esterase can be partially or completely purified by various techniques known in the art. These techniques include, but are not limited to, chromatography (e.g., ion exchange, affinity, hydrophobic, chromatofocusing and size exclusion), electrophoretic techniques (e.g., preparative isoelectric focusing), differential solubility (e.g., ammonium sulfate precipitation), SDS-PAGE or extraction to obtain a substantially pure polypeptide.

[0099] The esterase can be used as it is in purified form, alone or in combination with further enzymes, to catalyze the enzymatic reactions involved in the decomposition and / or recycling of polyester and / or polyester-containing materials, such as plastic products containing polyester. The esterase can be in soluble form or on a solid phase. In particular, the esterase can be bound to cell membranes or lipid vesicles or synthetic supports, such as glass, plastic, polymers, filters, membranes, etc., in the form of beads, columns, plates, etc.

[0100] composition A further object of the present invention is to provide a composition comprising the esterase of the present invention or a host cell or an extract thereof. In the context of the present invention, the term "composition" encompasses any kind of composition comprising the esterase of the present invention or a host cell.

[0101] The composition of the present invention may contain 0.1% to 99.9% by weight, preferably 0.1% to 50% by weight, more preferably 0.1% to 30% by weight, and even more preferably 0.1% to 5% by weight of the esterase, based on the total weight of the composition. Alternatively, the composition may contain 5 to 10% by weight of the esterase of the present invention.

[0102] The composition can be liquid or dry, for example in the form of a powder, hi some embodiments, the composition is a lyophilizate.

[0103] The composition may further comprise an excipient and / or a reagent, etc. Suitable excipients include buffers commonly used in biochemistry, agents for adjusting pH, preservatives such as sodium benzoate, sodium sorbate or sodium ascorbate, preservatives, protectants or stabilizers such as starch, dextrin, gum arabic, salts, sugars such as sorbitol, trehalose or lactose, glycerol, polyethylene glycol, polypropylene glycol, propylene glycol, sequestering agents such as EDTA, reducing agents, amino acids, carriers such as solvents or aqueous solutions, etc. The composition of the present invention can be obtained by mixing the esterase with one or more excipients.

[0104] In certain embodiments, the composition comprises 0.1% to 99.9% by weight, preferably 50% to 99.9% by weight, more preferably 70% to 99.9% by weight, and even more preferably 95% to 99.9% by weight of excipients based on the total weight of the composition. Alternatively, the composition may comprise 90% to 95% by weight of excipients.

[0105] In certain embodiments, the composition may further comprise additional polypeptides exhibiting enzymatic activity. The amount of the esterase of the invention will be easily adapted by the skilled person depending, for example, on the nature of the polyester to be degraded and / or the additional enzymes / polypeptides contained in the composition.

[0106] In a particular embodiment, the esterase of the invention is solubilized in an aqueous medium together with one or more excipients, in particular excipients capable of stabilizing or protecting the polypeptide from degradation. For example, the esterase of the invention can finally be solubilized in water together with further components, such as glycerol, sorbitol, dextrin, starch, glycols, such as propanediol, salts, etc. The resulting mixture can then be dried to obtain a powder. Methods for drying such mixtures are well known to those skilled in the art and include, but are not limited to, lyophilisation, freeze-drying, spray drying, supercritical drying, downdraft evaporation, thin layer evaporation, centrifugal evaporation, conveyor drying, fluidized bed drying, drum drying, or any combination thereof.

[0107] In certain embodiments, the composition is in powder form and comprises an esterase and a stabilizing / solubilizing amount of glycerol, sorbitol or dextrin, e.g., maltodextrin and / or cyclodextrin, starch, glycol, e.g., propanediol, and / or salt.

[0108] In a particular embodiment, the composition of the invention comprises at least one recombinant cell expressing the esterase of the invention or an extract thereof. "Extract of a cell" refers to any fraction obtained from a cell, such as cell supernatant, cell debris, cell wall, DNA extract, enzyme or enzyme preparation or any preparation obtained from a cell by chemical, physical and / or enzymatic treatment, essentially free of living cells. A preferred extract is an enzyme-active extract. The composition of the invention can comprise one or more recombinant cells of the invention or an extract thereof and optionally one or more additional cells.

[0109] In an embodiment, the composition consists of or comprises a culture medium of a recombinant microorganism that expresses and secretes an esterase of the invention. In a particular embodiment, the composition comprises such a culture medium that has been lyophilized.

[0110] Use of esterase It is a further object of the present invention to provide a method of using the esterase of the present invention for degrading and / or recycling polyester or polyester-containing materials under aerobic or anaerobic conditions. The esterase of the present invention is particularly useful for degrading PET and PET-containing materials.

[0111] Therefore, an object of the present invention is the use of an esterase of the present invention or a corresponding recombinant cell or extract or composition thereof for the enzymatic degradation of polyesters.

[0112] In a particular embodiment, the polyester targeted by the esterase is selected from polyethylene terephthalate (PET), polytrimethylene terephthalate (PTT), polybutylene terephthalate (PBT), polyethylene isosorbide terephthalate (PEIT), polylactic acid (PLA), polyhydroxyalkanoate (PHA), polybutylene succinate (PBS), polybutylene succinate adipate (PBSA), polybutylene adipate terephthalate (PBAT), polyethylene furanoate (PEF), polycaprolactone (PCL), poly(ethylene adipate) (PEA), polyethylene naphthalate (PEN) and blends / mixtures of these materials, preferably polyethylene terephthalate.

[0113] In a preferred embodiment, the polyester is PET and at least monomers (e.g., monoethylene glycol or terephthalic acid) and / or oligomers (e.g., methyl-2-hydroxyethyl terephthalate (MHET), bis(2-hydroxyethyl) terephthalate (BHET), 1-(2-hydroxyethyl) 4-methyl terephthalate (HEMT), and dimethyl terephthalate (DMT) are recovered.

[0114] It is also an object of the present invention to use an esterase according to the invention or a corresponding recombinant cell or an extract or composition thereof for the enzymatic degradation of at least one polyester of a polyester-containing material.

[0115] Another object of the present invention is to provide a method for degrading at least one polyester of a polyester-containing material, comprising contacting the polyester-containing material with an esterase or a host cell or a composition of the present invention, thereby degrading at least one polyester of the polyester-containing material.

[0116] Advantageously, the polyester is depolymerized to monomers and / or oligomers.

[0117] In particular, the present invention provides a method for degrading PET in a PET-containing material by contacting the PET-containing material with an esterase or host cell or composition of the present invention, thereby degrading the PET.

[0118] In an embodiment, at least one polyester is decomposed into repolymerizable monomers and / or oligomers, which can be advantageously recovered for reuse. The recovered monomers / oligomers can be used for recycling (e.g., repolymerization of polyester) or methanation. In a particular embodiment, at least one polyester is PET, and monoethylene glycol, terephthalic acid, methyl-2-hydroxyethyl terephthalate (MHET), bis(2-hydroxyethyl) terephthalate (BHET), 1-(2-hydroxyethyl) 4-methyl terephthalate (HEMT), and / or dimethyl terephthalate (DMT) are recovered.

[0119] In an embodiment, the polyester of the polyester-containing material is completely degraded.

[0120] The time required to degrade a polyester-containing material can vary depending on the polyester-containing material itself (i.e. the nature and origin of the polyester-containing material, its composition, form, etc.), the type and amount of esterase used, and various process parameters (i.e. temperature, pH, further agents, etc.) A person skilled in the art can easily adapt the process parameters to the polyester-containing material and the expected degradation time.

[0121] Advantageously, the degradation method is carried out at a temperature comprised between 20°C and 90°C, preferably between 40°C and 80°C, more preferably between 50°C and 70°C, more preferably between 60°C and 70°C. In a particular embodiment, the degradation process is carried out at 65°C. In another particular embodiment, the degradation method is carried out at 70°C. More generally, the temperature is kept below the inactivation temperature, which corresponds to the temperature at which the esterase is inactivated (i.e. loses more than 80% of its activity compared to its activity at its optimum temperature) and / or at which the recombinant microorganism no longer synthesizes the esterase. In particular, the temperature is kept below the glass transition temperature (Tg) of the target polyester.

[0122] Advantageously, the process is carried out in a continuous flow process at a temperature which allows the esterase to be used and / or reused several times.

[0123] Advantageously, the decomposition process is carried out at a pH comprised between 5 and 11, preferably between 6 and 9, more preferably between 6.5 and 9, even more preferably between 6.5 and 8.

[0124] In certain embodiments, the polyester-containing material can be pretreated prior to contact with the esterase to physically alter its structure to increase the contact surface between the polyester and the esterase.

[0125] Another object of the present invention is to provide a method for producing monomers and / or oligomers from a polyester-containing material, comprising exposing the polyester-containing material to an esterase of the present invention or a corresponding recombinant cell or extract or composition thereof, and optionally recovering the monomers and / or oligomers.

[0126] The monomers and / or oligomers resulting from the depolymerization can be recovered sequentially or continuously. Depending on the starting polyester-containing material, one type of monomer and / or oligomer or several different types of monomers and / or oligomers can be recovered.

[0127] The process of the present invention is particularly useful for producing monomers selected from monoethylene glycol and terephthalic acid and / or oligomers selected from methyl-2-hydroxyethyl terephthalate (MHET), bis(2-hydroxyethyl) terephthalate (BHET), 1-(2-hydroxyethyl) 4-methyl terephthalate (HEMT) and dimethyl terephthalate (DMT) from PET and / or plastic products comprising PET.

[0128] The recovered monomers and / or oligomers can be further purified using any suitable purification method and can be prepared into a repolymerizable form. Examples of purification methods include stripping processes, separation with aqueous solutions, steam selective condensation, post-bioprocessing filtration and concentration of the medium, separation, distillation, vacuum evaporation, extraction, electrodialysis, adsorption, ion exchange, precipitation, crystallization, concentration and acid addition dehydration and precipitation, nanofiltration, acid catalyzed treatment, semi-continuous or continuous mode distillation, solvent extraction, evaporative concentration, evaporative crystallization, liquid / liquid extraction, hydrogenation, azeotropic distillation processes, adsorption, column chromatography, simple vacuum distillation, and microfiltration, in combination or alone.

[0129] The recovered repolymerizable monomers and / or oligomers can be reused, for example, to synthesize polyesters. Advantageously, polyesters of the same nature are repolymerized. However, it is possible to mix the recovered monomers and / or oligomers with other monomers and / or oligomers, for example to synthesize new copolymers. Alternatively, the recovered monomers can be used as chemical intermediates to produce new compounds of interest.

[0130] The present invention also relates to a method for surface hydrolysis or surface functionalization of polyester-containing materials, comprising exposing the polyester-containing material to an esterase of the present invention or a corresponding recombinant cell or extract or composition thereof. The method of the present invention is particularly useful for improving the hydrophilicity or water absorbency of polyester materials. Such improved hydrophilicity may be of particular interest in textile manufacturing, electronics and biomedical applications.

[0131] A further object of the present invention is to provide a polyester-containing material comprising the esterase of the present invention and / or a recombinant microorganism expressing and secreting said esterase. By way of example, methods for producing such polyester-containing materials comprising the esterase of the present invention are disclosed in patent applications WO 2013 / 093355, WO 2016 / 198650, WO 2016 / 198652, WO 2019 / 043145 and WO 2019 / 043134.

[0132] Therefore, an object of the present invention is to provide a polyester-containing material containing the esterase and / or recombinant cell and / or composition or extract thereof of the present invention and at least PET. According to an embodiment, the present invention provides a plastic product comprising PET and the esterase of the present invention having PET decomposition activity.

According to an embodiment, the present invention provides a plastic product comprising PBS and the esterase of the present invention having PBS decomposition activity.

[0135] Therefore, another object of the present invention is to provide a polyester-containing material containing the esterase and / or recombinant cell and / or composition or extract thereof of the present invention and at least PCL. According to an embodiment, the present invention provides a plastic product comprising PCL and the esterase of the present invention having PCL decomposition activity.

[0136] Classically, the esterase of the present invention can be used in detergent, food, animal feed and pharmaceutical applications. Detergent compositions include, but are not limited to, hand or machine laundry detergent compositions, such as laundry additive compositions and rinse additive fabric softener compositions suitable for pre-treatment of stained fabrics, detergent compositions for use in general household hard surface cleaning operations, and detergent compositions for hand or machine dishwashing operations. In a particular embodiment, the esterase of the present invention can be used as a detergent additive. Thus, the present invention provides a detergent composition comprising the esterase of the present invention. In particular, the esterase of the present invention can be used as a detergent additive to reduce pilling and graying effects during textile washing.

[0137] The present invention is also directed to a method for using the esterase of the present invention in animal feed, as well as to feed compositions and feed additives comprising the esterase of the present invention. The terms "feed" and "feed composition" refer to any compound, preparation, mixture or composition suitable for or intended for consumption by an animal. In another particular embodiment, the esterase of the present invention is used to hydrolyze proteins and produce hydrolysates comprising peptides. Such hydrolysates can be used as feed compositions or feed additives.

[0138] Working Example Example 1 - Construction, expression and purification of esterases construction The esterases of the invention were generated using the plasmid construct pET26b-LCC-His. This plasmid consists of the cloning of the gene encoding the esterase of SEQ ID NO:1, optimized for Escherichia coli expression, between the NdeI and XhoI restriction sites. To generate the esterase variants, two site-directed mutagenesis kits from Agilent (Santa Clara, California, USA) were used, following the supplier's recommendations: QuikChange II Site-Directed Mutagenesis Kit and QuikChange Lightning Multi Site-Directed.

[0139] Expression and purification of esterases Cloning and recombinant expression were carried out in 50 mL of LB-Miller or ZYM autoinducing medium (Studier et al., 2005- Prot. Exp. Pur. 41, 207-234) using successive strains Stellar™ (Clontech, California, USA) and E. coli One Shot® BL21 DE3 (Life technologies, Carlsbad, California, USA). Induction in LB-Miller medium was carried out with 0.5 mM isopropyl β-D-1-thiogalactopyranoside (IPTG, Euromedex, Souffelweyersheim, France) at 16°C. Cultures were stopped by centrifugation (8000 rpm, 20 min, 10°C) in an Avanti J-26 XP centrifuge (Beckman Coulter, Brea, USA). The cells were suspended in 20 mL of Talon buffer (20 mM Tris-HCl, 300 mM NaCl, pH 8). The cell suspension was then sonicated for 2 min at 30% of amplitude (2 sec ON and 1 sec OFF cycles) with an FB 705 sonicator (Fisherbrand, Illkirch, France). A centrifugation step was then achieved in an Eppendorf centrifuge at 11000 rpm, 10°C, for 30 min. The soluble fraction was collected and subjected to affinity chromatography. The purification step was completed with Talon® Metal Affinity Resin (Clontech, CA, USA). Protein elution was performed with a step of Talon buffer supplemented with imidazole. The purified protein was dialyzed against Talon buffer and then quantified using the Bio-Rad protein assay according to the manufacturer's instructions (Lifescience Bio-Rad, France) and stored at +4°C.

[0140] Example 2 - Evaluation of esterase decomposition activity The degradation activity of the esterase was determined and compared to that of the esterase of SEQ ID NO:1.

[0141] Several methods for assessing specific activity were used. (1) Specific activity and final yield based on PET hydrolysis (2) Activity based on the decomposition of solid polyester (3) Activity based on PET hydrolysis in reactors over 100 mL

[0142] 2.1 Specific activity and final yield based on PET hydrolysis 100 mg amorphous PET (powder, prepared according to WO 2017 / 198786, reaching a crystallinity of less than 20%) was weighed and introduced into a 100 mL glass bottle. 1 mL of an esterase preparation containing the esterase of SEQ ID NO:1 (as a reference control) or the esterase of the invention, prepared at 0.02 or 0.03 mg / mL in Talon buffer (20 mM Tris-HCl, 0.3 M NaCl, pH 8) was introduced into the glass bottle. Finally, 49 mL of 0.1 M potassium phosphate buffer (pH 8) was added.

[0143] Depolymerization was initiated by incubating each vial at 60°C, 65°C or 70°C and 150 rpm in a Max Q 4450 incubator (Thermo Fisher Scientific, Inc. Waltham, Mass., USA).

[0144] The initial rate of the depolymerization reaction (mg equivalents of TA produced / h) was determined by sampling at different time points during the first 72 h and analyzed by ultra-high performance liquid chromatography (UHPLC). If necessary, the samples were diluted with 0.1 M potassium phosphate buffer (pH 8). Then, 150 μL of methanol and 6.5 μL of 6 N HCl were added to 150 μL of sample or diluent. After mixing and filtering through a 0.45 μm syringe filter, the samples were loaded into the UHPLC to monitor the release of terephthalic acid (TA), MHET, and BHET. The chromatographic system used was an Ultimate 3000 UHPLC system (Thermo Fisher Scientific, Inc. Waltham, MA, USA) including a pump module, an autosampler, a column oven thermostated at 25 °C, and a UV detector at 240 nm. The column used was a Discovery® HS C18 HPLC column (150 × 4.6 mm, 5 μm, equipped with a precolumn, Supelco, Bellefonte, USA). TA, MHET and BHET were at 1 mM H 2 SO 4 A gradient of MeOH in water (30%-90%) was used for separation at 1 mL / min. Injections consisted of 20 μL of sample. TA, MHET and BHET were measured according to calibration curves made from commercial TA and BHET and self-synthesized MHET under the same conditions as the samples. The specific activity of PET hydrolysis (mg equivalents of TA / h / mg enzyme) was determined in the linear part of the hydrolysis curve of the reaction, such a curve being set up by samplings made at different times during the first 72 h. The equivalents of TA correspond to the sum of the measured TA and the TA contained in the measured MHET and BHET. The PET hydrolysis yield was determined from 72 h to 96 h using the same equivalent TA measurements.

[0145] 2.2 Activity based on the decomposition of solid polyester The induced cells, semi-purified protein extracts or purified proteins can be used as compositions containing the esterases of the invention to assess the activity of such esterases.

[0146] The induced cells correspond to samples of cell cultures obtained either after cultivation in ZYM autoinducing medium or after induction with IPTG in LB-Miller medium (described in Example 1).

[0147] Semi-purified protein extracts were obtained either after cultivation in ZYM autoinducing medium or after induction with IPTG in LB-Miller medium (described in Example 1) according to the following protocol: Cultures were stopped by centrifugation (8000 rpm, 20 min, 10° C.) in an Avanti J-26 XP centrifuge (Beckman Coulter, Brea, USA). Cell pellets were suspended in lysis buffer (20 mM Tris-HCl, 300 mM NaCl, pH 8). Cells were disrupted by a 2-h freeze / thaw cycle at −80° C., followed by 1-h incubation at 28° C. with the addition of 1 μL Lysonase Bioprocessing Reagent (Merck Millipore, Darmstadt, Germany) and vortex homogenization every 15 min. Lysates were cleared by centrifugation (2250×g, 15 min, 4° C.). To generate semi-purified fractions, lysates were treated at 70° C. for 1 h and clarified by centrifugation (2250×g, 15 min, 4° C.). Protein concentrations of fractions were quantified using the Bio-Rad protein assay according to the manufacturer's instructions (Lifescience Bio-Rad, France).

[0148] The purified protein was obtained as described in Example 1.

[0149] Samples of the composition were placed either on the surface or in wells formed in agar omnitrays containing PET or another solid polyester compound (e.g., PBAT or analogues) prepared as follows: Preparation of agar plates containing PET was achieved by solubilizing 500 mg PET in hexafluoro-2-propanol (HFIP) and pouring this medium into 250 mL of an aqueous solution. After evaporation of the HFIP at 52°C under 140 mbar, the solution was mixed v / v with 0.2 M potassium phosphate buffer (pH 8) containing 3% agar. Approximately 30 mL of the mixture was used to prepare each omnitray and stored at 4°C.

[0150] The surface area or diameter of the halos formed by polyester degradation by wild-type esterase and the mutants of the invention were measured and compared after 2 to 24 hours at 60°C, 65°C or 70°C.

[0151] 2.3 Activity based on PET hydrolysis in the reactor 0.69 μmol–2.07 μmol purified esterase, prepared in 80 mL of 100 mM potassium phosphate buffer (pH 8), was mixed with 20 g amorphous PET (prepared according to WO 2017 / 198786, reaching a crystallinity of less than 20%) in a 500 mL Minibio reactor (Applikon Biotechnology, Delft, The Netherlands). Temperature regulation at 65 °C was performed by water bath immersion and constant stirring was maintained at 250 rpm using one marine impeller. The pH of the PET depolymerization assay was regulated to pH 8 by 6 N NaOH and guaranteed by a my-Control bio controller system (Applikon Biotechnology, Delft, The Netherlands). Base consumption was recorded during the assay and can be used for characterization of the PET depolymerization assay.

[0152] The final yield of the PET depolymerization assay was determined either by measuring the weight of the residual PET or by measuring the equivalents of TA and EG produced or by base consumption. The weight measurement of the residual PET was assessed at the end of the reaction by filtering the reaction volume through a 12-15 μm grade 11 ashless filter paper (Dutscher SAS, Brumath, France) and drying such residue and then weighing. The measurement of the equivalents of TA and EG produced was realized using the UHPLC method described in 2.1 and the % hydrolysis was calculated based on the ratio of the molar concentration (TA+MHET+BHET) at a given time to the total amount of TA contained in the initial sample. The PET depolymerization produced acidic monomers that could be neutralized with base to maintain the pH in the reactor. The measurement of the equivalents of TA produced was calculated using the corresponding molar base consumption and the % hydrolysis was calculated based on the ratio of the molar concentration at a given time of the equivalents of TA to the total amount of TA contained in the initial sample.

[0160] V2 to V6 each have the exact amino acid sequence of SEQ ID NO:1, except for the combination of substitutions listed.

[0161] Example 3 - Evaluation of the thermostability of the esterases of the invention The thermostability of the esterases of the present invention was determined and compared to that of the esterase of SEQ ID NO:1.

[0162] Various methods were used to estimate thermal stability. (1) Circular dichroism of proteins in solution (2) Residual esterase activity after incubation of the protein under defined temperature, time and buffer conditions (3) Residual polyester depolymerization activity after incubation of the protein under the specified temperature, time and buffer conditions (4) the ability to degrade solid polyester compounds (e.g., PET or PBAT or analogs) dispersed in agar plates after incubation of the protein under defined temperature, time and buffer conditions; (5) Ability to perform polyester depolymerization assays multiple times under defined conditions of temperature, buffer, protein concentration, and polyester concentration. (6) Differential Scanning Fluorometry (DSF)

[0163] A detailed protocol for such a method is provided below.

[0164] 3.1 Circular dichroism Circular dichroism (CD) was performed to measure the melting temperature (T m ) (Tm=84.7°C) was performed with a Jasco 815 instrument (Easton, USA) to compare with the Tm of the esterase of the present invention. Technically, 400 μL of protein sample was prepared at 0.5 mg / mL in Talon buffer and used for CD. A first scan from 280 to 190 nm was realized to determine the two CD intensities maxima corresponding to the correct folding of the protein. A second scan was then performed from 25°C to 110°C with a wave length corresponding to such an intensity maximum, providing a specific curve (sigmoidal 3 parameters y=a / (1+e^((x-x0) / b))) that was analyzed by Sigmaplot version 11.0 software to determine the Tm when x=x0. The obtained T m reflects the thermal stability of a given protein. m The higher the , the more stable the mutant is at high temperatures.

[0165] 3.2 Residual esterase activity 1 mL of a 40 mg / L (in Talon buffer) solution of the esterase of SEQ ID NO:1 or the esterase of the present invention was incubated at various temperatures (65, 70, 75, 80 and 90° C.) for 10 days. Samples were taken periodically and diluted 1-500 times with 0.1 M potassium phosphate buffer (pH 8.0) to realize the paranitrophenol-butyrate (pNP-B) assay. 20 μL of sample was mixed with 175 μL of 1 M potassium phosphate buffer (pH 8.0) and 5 μL of pNP-B solution (40 mM) in 2-methyl-2-butanol. The enzyme reaction was carried out at 30° C. for 15 min under stirring, and the absorbance at 405 nm was obtained by a microplate spectrophotometer (Versamax, Molecular Devices, Sunnyvale, CA, USA). The activity of pNP-B hydrolysis (initial rate expressed in μmol pNPB / min) was determined using a calibration curve for liberated paranitrophenol in the linear part of the hydrolysis curve.

[0166] 3.3 Residual polyester depolymerization activity 10 mL of a solution of 40 mg / L (in Talon buffer) of the esterase of SEQ ID NO:1 and the esterase of the present invention, respectively, were incubated at various temperatures (65, 70, 75, 80 and 90°C) for 1 to 30 days. 1 mL of sample was periodically taken and transferred to a bottle containing 100 mg amorphous PET (prepared according to WO 2017 / 198786, to reach a crystallinity of less than 20%) micronized to 250-500 μm and 49 mL of 0.1 M potassium phosphate buffer (pH 8.0) and incubated at 65°C. 150 μL of buffer was periodically sampled. If necessary, the sample was diluted with 0.1 M potassium phosphate buffer (pH 8). Then, 150 μL of methanol and 6.5 μL of 6 N HCl were added to the sample or 150 μL of diluent. After mixing and filtering through a 0.45 μm syringe filter, the samples were loaded onto a UHPLC to monitor the release of terephthalic acid (TA), MHET and BHET. The chromatographic system used was an Ultimate 3000 UHPLC system (Thermo Fisher Scientific, Inc. Waltham, MA, USA) including a pump module, an autosampler, a column oven thermostated at 25° C. and a UV detector at 240 nm. The column used was a Discovery® HS C18 HPLC column (150×4.6 mm, 5 μm, with precolumn, Supelco, Bellefonte, USA). TA, MHET and BHET were dissolved in 1 mM H 2 SO 4 A gradient of MeOH in water (30%-90%) was used for separation at 1 mL / min. Injections consisted of 20 μL of sample. TA, MHET and BHET were measured according to calibration curves made from commercial TA and BHET and self-synthesized MHET under the same conditions as the samples. The activity of PET hydrolysis (μmol PET hydrolyzed / min or mg equivalent TA produced / h) was determined in the linear part of the hydrolysis curve, which was set up by sampling at different times during the first 24 h. The equivalent TA corresponds to the sum of the measured TA and the TA contained in the measured MHET and BHET.

[0167] 3.4 Decomposition of solid polyester 1 mL of a solution of 40 mg / L (in Talon buffer) of the esterase of SEQ ID NO:1 and the esterase of the invention, respectively, was incubated at different temperatures (65, 70, 75, 80 and 90° C.) for 1 to 30 days. 20 μL of the enzyme preparation was regularly placed in wells made in an agar plate containing PET. The preparation of an agar plate containing PET was realized by solubilizing 500 mg PET in hexafluoro-2-propanol (HFIP) and pouring this medium into 250 mL of an aqueous solution. After evaporation of the HFIP at 52° C. under 140 mbar, the solution was mixed v / v with 0.2 M potassium phosphate buffer (pH 8) containing 3% agar. Approximately 30 mL of the mixture is used to prepare each Omnitray and stored at 4° C.

[0168] The diameter or surface area of the halos formed by polyester degradation by wild-type esterase and the mutants of the invention were measured and compared after 2 to 24 hours at 60° C., 65° C. or 70° C. The half-life of the enzyme at a given temperature corresponds to the time required for the diameter or surface area of the halo to be reduced by half.

[0169] 3.5 Multiple depolymerization of polyester The ability of the esterases to perform successive polyester depolymerization assays was evaluated in an enzyme reactor. A Minibio 500 reactor (Applikon Biotechnology BV, Delft, The Netherlands) was started with 100 mL of 10 mM potassium phosphate buffer (pH 8) containing 3 g amorphous PET (prepared according to WO 2017 / 198786, reaching a crystallinity of less than 20%) and 3 mg LC-esterase. Agitation was set at 250 rpm using a marine impeller. The bioreactor was thermostated at 60°C, 65°C or 70°C by immersion in an external water bath. The pH was regulated at 8 by addition of 3 M KOH. The different parameters (pH, temperature, agitation, addition of base) were monitored by BioXpert software V2.95. 1.8 g amorphous PET was added every 20 hours. 500 μL of reaction medium was sampled periodically.

[0170] The amounts of TA, MHET and BHET were determined by HPLC as described in Example 2.3. The amount of EG was determined using an Aminex HPX-87K column (Bio-Rad Laboratories, Inc, Hercules, California, United States) thermostated at 65°C. The eluate was diluted with 0.6 mL / min. -1 5 mM K at 2 HPO 4 Injections were 20 μL. Ethylene glycol was monitored using a refractometer.

[0171] The % hydrolysis was calculated based on the ratio of the molar concentration (TA+MHET+BHET) at a given time to the total amount of TA contained in the initial sample or based on the ratio of the molar concentration (EG+MHET+2×BHET) at a given time to the total amount of EG contained in the initial sample. The degradation rate is calculated in mg total free TA per hour or mg total EG per hour.

[0172] The half-life of the enzyme was estimated as the incubation time required to obtain a 50% loss in degradation rate.

[0173] 3.6 Differential Scanning Fluorometry (DSF) DSF was used to assess the thermal stability of wild-type protein (SEQ ID NO:1) and its mutants by determining the melting temperature (Tm), the temperature at which half of the protein population is unfolded. Protein samples were prepared at a concentration of 14 μM (0.4 mg / mL) and stored in Buffer A consisting of 20 mM Tris HCl (pH 8.0), 300 mM NaCl. A 5000× stock solution of SYPRO Orange dye in DMSO was first diluted to 250× with water. Protein samples were loaded onto a white clear 96-well PCR plate (Bio-Rad cat# HSP9601), where each well contained a final volume of 25 μl. The final concentrations of protein and SYPRO Orange dye in each well were 5 μM (0.14 mg / ml) and 10×, respectively. The volumes loaded per well were as follows: 15 μL Buffer A, 9 μL 0.4 mg / mL protein solution, and 1 μL 250× Sypro Orange diluted solution. The PCR plate was then sealed with optical quality sealing tape and spun at 2000 rpm for 1 minute at room temperature. DSF experiments were then performed using a CFX96 Real-Time PCR System configured to use 450 / 490 excitation and 560 / 580 emission filters. Samples were heated from 25 to 100°C at a rate of 0.3°C / sec. A single fluorescence measurement was taken every 0.03 sec. Melting temperatures were determined from the peak of the first derivative of the melting curve using Bio-Rad CFX Manager software.

[0174] The esterase of SEQ ID NO:1 and the esterase of the present invention were then compared based on their Tm values. Due to the high reproducibility between experiments on the same protein from different preparations, a ΔTm of 0.8°C was considered significant for comparing the variants. The Tm values correspond to the average of at least three measurements. The Tm of the esterase of SEQ ID NO:1 is estimated at 84.7°C.

[0175] The thermostabilities of the esterases (variants) of the invention are summarized in Table 3 below, expressed as Tm values, and evaluated according to Example 2.6, where the increase in Tm compared to the esterase of SEQ ID NO:1 is indicated in brackets.
