Isolation method for extremely halophilic bacteria

Abstract

To provide a method for isolating extreme halophilic cells from an extreme halophilic culture, which is more efficient than conventional methods.SOLUTION: Provided is a method for isolating extreme halophilic cells from a culture solution in which the extreme halophilic cells are cultured, comprising the following steps: (1) adding an organic solvent having a lower specific gravity than water and in which extreme halophilic cells are insoluble to the culture solution of the extreme halophilic cells and mixing them, to obtain a mixed solution; and (2) isolating the extreme halophilic cells in an intermediate layer by layer-separating the mixed solution into the intermediate layer containing the intracellular component of the extreme halophilic cells, which is insoluble in a water-soluble component phase (lower layer) and a fat-soluble component phase (upper layer).
