mRNA display antibody library and method

Abstract

To provide a composition and a method for fabricating an mRNA display library and a recombinant high affinity binder.SOLUTION: A recombinant virus includes recombinant nucleic acid including a member of an expression library encoding a separate antibody or an antibody fragment, and the member of the expression library is fabricated by: a step of fabricating or providing (1) a VH-CDR1 / 2 sub-library, (2) a plurality of VH-CDR3 sub-libraries, and (3) a VL sub-library, wherein each member of sub-libraries includes at least one random cassette having a plurality of degenerate base positions; and a step of recombining at least a part of at least two members of the VH-CDR1 / 2 sub-library, the plurality of VH-CDR3 sub-libraries, and the VL sub-library to form an expression library member in the expression library.SELECTED DRAWING: Figure 5
