Description

Technical Field

[0001] The present invention relates to an active ingredient that has excellent biological safety and is incorporated into external skin preparations (quasi-drugs, cosmetics, etc.).

Background Art

[0002] In recent years, an increasing number of people experience subjective symptoms such as erythema and itching on the skin due to the excessive response of the in-vivo immune system to various environmental factors and stress. For this reason, active ingredients for external skin preparations for antipruritic, analgesic, or anti-stress use are in demand, but no active ingredient that exhibits sufficient effectiveness and has excellent biological safety has been found yet.

Disclosure of the Invention

Problems to be Solved by the Invention

[0003] As a result of intensive studies aimed at solving the above problems, the present inventors found that an extract of Damask rose flowers exerts an effect of significantly suppressing the production of Nerve Growth Factor (NGF), which is a cause of hyperesthesia in the epidermis or dermis. Conventionally, it has been known from the techniques described in, for example, cited documents 1 to 2 that extracts of Damask rose flowers have an antioxidant effect, a whitening effect, or a skin turnover improving effect, but it was not known that extracts of Damask rose flowers exert an effect of suppressing the production of nerve growth factor.

Means for Solving the Problems

[0005] The present invention is a nerve growth factor production inhibitor comprising, as an active ingredient, a solvent extract of Damask rose belonging to the genus Rosa of the family Rosaceae.

Effects of the Invention

[0006] According to the present invention, a topical skin preparation for improving rough skin, relieving itching, relieving pain, or for sensitive skin can be provided based on a component derived from natural products, which has excellent biocompatibility and inhibits the production of nerve growth factor (NGF). Furthermore, a topical skin preparation for improving itchy skin diseases (such as atopic dermatitis) can also be provided. [Brief explanation of the drawing]

[0007] [Figure 1] Figure 1 shows the results of a test evaluating the effect of inhibiting nerve growth factor production. [Modes for carrying out the invention]

[0008] Preferred embodiments of the present invention will be described in detail below. The extract material used in this invention is Rosa damascena, a member of the Rosaceae family, and any variety (including hybrids and subspecies) of Rosa damascena may be used.

[0009] The materials used in this invention may include the whole plant, leaves, flowers, stems, seeds, fruits, roots, etc., of the Damask rose, but the use of the whole plant or flowers is preferred.

[0010] To prepare the extract, first, the Damask rose (e.g., whole plant, flowers, etc.) is washed with water if necessary to remove foreign matter, then either left as is or dried, and finely chopped or pulverized as needed, and then brought into contact with the extraction solvent for extraction. Extraction can be carried out by contacting the extraction solvent according to conventional methods such as immersion, but supercritical fluid extraction can also be used in addition to immersion.

[0011] Examples of extraction solvents include water; lower alcohols such as methanol, ethanol, and propanol; polyhydric alcohols such as ethylene glycol, propylene glycol, 1,3-butylene glycol, and glycerin; esters such as ethyl acetate, butyl acetate, and methyl propionate; ketones such as acetone and methyl ethyl ketone; ethers such as ethyl ether and isopropyl ether; and hydrocarbon solvents such as n-hexane, toluene, and chloroform. These can be used individually or in combination of two or more.

[0012] Among these extraction solvents, hydrophilic solvents such as water, lower alcohols, or polyhydric alcohols are preferred in the present invention, from the viewpoint of skin irritation and effectiveness, as well as their broad applicability to cosmetics. Preferred examples of using these hydrophilic solvents include, for example, the use of water, lower alcohols (especially ethanol), or polyhydric alcohols (especially 1,3-butylene glycol) alone, or a mixed solvent of water and lower alcohols (especially ethanol), or a mixed solvent of water and polyhydric alcohols (especially 1,3-butylene glycol, glycerin), but among these, water alone or a mixed solvent of water and 1,3-butylene glycol are particularly preferred.

[0013] When using a mixed solvent, the mixing ratio is preferably in the range of 1:10 to 20:1 by volume (hereinafter the same) for a mixed solvent of water and 1,3-butylene glycol, 1:10 to 25:1 for a mixed solvent of water and ethanol, and 1:10 to 20:1 for a mixed solvent of water and glycerin.

[0014] Furthermore, the weight ratio of the dried part of the Damask rose (e.g., the whole plant or flowers) to the extraction solvent is preferably in the range of 1:1 to 1:50, and more preferably in the range of 1:5 to 1:35.

[0015] While there are no particular limitations on the pH of the extract during preparation, it is generally preferable to have a pH in the range of 3 to 9. In this sense, if necessary, the extraction solvent may be adjusted to the desired pH by adding an alkaline adjusting agent such as sodium hydroxide, sodium carbonate, or potassium hydroxide, or an acidic adjusting agent such as citric acid, hydrochloric acid, phosphoric acid, or sulfuric acid.

[0016] Extraction conditions such as extraction temperature and extraction time vary depending on the type of solvent and pH used. For example, when using water, 1,3-butylene glycol, or a mixture of water and 1,3-butylene glycol as the solvent, the extraction temperature is preferably in the range of 0°C to 80°C, more preferably in the range of 0°C to 20°C, and the extraction time is preferably in the range of 1 to 168 hours (1 hour to 1 week), more preferably in the range of 1 to 120 hours (1 hour to 5 days).

[0017] Furthermore, prior to or in parallel with the extraction process of the present invention, the Damask rose may be subjected to hydrolysis as needed. This may improve the storage stability of the Damask rose extract and allow for more effective use of the extract as a cosmetic ingredient.

[0018] When the extract is subjected to enzymatic hydrolysis, one or more enzymes selected from any of the following groups may be used: proteolytic enzymes such as actinase, papain, and pepsin; starch-degrading enzymes such as glucoamylase, α-amylase, and β-amylase; fibrinolytic enzymes such as cellulase, hemicellulase, and pectinase; and lipases. However, it is more preferable to use a combination of one or more enzymes selected from each of these groups.

[0019] The amount of enzyme added is preferably in the range of 0.01 to 10% by weight in total, and more preferably in the range of 0.1 to 2.0% by weight, relative to the solid content, for example, the whole plant or flower part of the Damask rose.

[0020] The extract prepared as described above may generally be adjusted to a pH of 3 to 8, and used as it is as a cosmetic compounding ingredient, or may be used after being concentrated to a desired concentration by vacuum concentration or the like. Further, the extract may be made into a dried product by a conventional method such as a spray drying method.

[0021] Further, the extract prepared as described above may be refrigerated for a certain period of time to enhance storage stability and the like, and then the supernatant may be used as a cosmetic compounding ingredient.

[0022] The extract of the present invention obtained as described above contains various active ingredients. For example, it was confirmed by measurement using liquid chromatography (manufactured by Hitachi High-Technologies Corporation; LaChrom ELITE) that the extract contains gallic acid, chlorogenic acid, and caffeic acid as organic acids.

[0023] Examples of uses of the external skin preparation containing the Damask rose extract of the present invention include cosmetics (including quasi-drugs), for example, basic cosmetics such as milky lotions, creams, lotions, essences, and packs; makeup cosmetics such as lipsticks, foundations, liquid foundations, makeup pressed powders, blushes, and face powders; cleansing cosmetics such as facial cleansers, body shampoos, hair shampoos, and soaps; hair restorers, and further bath additives and the like, but are of course not limited thereto.

[0024] When the Damask rose extract of the present invention is used in cosmetics, the blending amount of the Damask rose extract, as the solid content of the extract, is generally in the range of 0.002 to 1.0% by weight, preferably 0.02 to 0.2% by weight for basic cosmetics; generally in the range of 0.002 to 1.0% by weight, preferably 0.02 to 0.2% by weight for makeup cosmetics; and generally in the range of 0.002 to 10.0% by weight, preferably 0.02 to 7.0% by weight for cleansing cosmetics.

In addition, as a comparative group, a control group was established in which only 50% 1,3-butylene glycol solution was added to the culture medium (adjusted to a final concentration of 1,3-butylene glycol of 2% of the total volume of the culture medium) instead of the extract from Production Example 1, and a control group was established in which 50% 1,3-butylene glycol solution and 100 μM epinephrine (LKT lab) were added to the culture medium. Cells from the test group were collected with 0.5 mL of Trizol reagent (Invitrogen) 3 hours after addition. 100 μL of chloroform (Wako Pure Chemical Industries, Ltd.) was added to the collected cells and mixed by stirring. The mixture was then centrifuged at 12,500 rpm at 4°C for 15 minutes using a centrifuge (TOMY MX-160), and 200 μL of the aqueous layer was collected. 500 μL of isopropanol (Wako Pure Chemical Industries, Ltd.) was added to the collected aqueous layer and mixed by stirring. The mixture was then centrifuged at 12,500 rpm at 4°C for 15 minutes to obtain a total RNA precipitate. 1 mL of 75% ethanol was added to the total RNA and mixed by stirring. The precipitate was then collected by centrifugation at 12,500 rpm at 4°C for 15 minutes. The recovered total RNA was reverse transcribed using a designated kit (PrimeScript RT reagent Kit with gDNA Eraser (Perfect Real Time) [Takara Bio Inc.]) to synthesize cDNA. Using the synthesized cDNA as a sample, the expression of various genes and the expression of the internal standard G3PDH gene were detected using Thermal Cycler Dice® Real Time System Single [Takara Bio Inc.] and SYBR® Premix Ex Taq™ II (Tli RNaseH Plus) [Takara Bio Inc.].Here, G3PDH (glyceraldehyde-3-phosphate dehydrogenase) is one of the housekeeping genes (genes that are commonly expressed in a constant amount in many tissues and cells, and are always expressed and essential for cell maintenance and proliferation), and because its expression level is always constant, it is used as an internal standard in PCR experiments. The test results compare the expression levels of each gene in each test group when the expression level of the G3PDH gene is kept constant. In this test system, the relative expression levels of each gene in the test group and the control group were calculated when the gene expression level in the control group was set to 100.

[0062] The results of Test Example 1 are shown in Figure 1. First, as shown in Figure 1, it was confirmed that the expression level of the NGF gene was significantly increased in the control group to which epinephrine was added as a stressor substance. On the other hand, it was confirmed that the expression of the NGF gene was significantly suppressed in the test group to which the extract according to the present invention was added compared to the control group.
