Abstract
Description
Method for producing a compound having a bibranched N-acetyl-D-galactosamine structure
[0001] The present invention relates to a novel method for producing a biantennary N-acetyl-D-galactosamine ligand-oligonucleotide conjugate, and more particularly to an industrially advantageous novel method for producing a biantennary N-acetyl-D-galactosamine unit and novel intermediates.
[0002] A method for delivering oligonucleotides to hepatic parenchymal cells in the liver using conjugates of nucleic acid drugs in which N-acetyl-D-galactosamine (also known as GalNAc) is linked to the oligonucleotide via a linker has been reported (Patent Document 1). A novel biantennary N-acetyl-D-galactosamine unit (also known as biantennary GalNAc unit) that enables hepatic delivery of any oligonucleotide, as well as a method for producing this unit using a protecting group such as an Fmoc group, have also been reported (Patent Document 2). While there is a need for a new method for the mass synthesis of high-quality biantennary GalNAc units, the production of compounds such as biantennary GalNAc units, which generally have complex structures and many reactive sites, is extremely difficult. In particular, the selective introduction and removal of substituents and protecting groups during the production process to obtain high-quality compounds with complex structures and many reactive sites in high yields is difficult. For example, attempts have been made to selectively remove only the benzyl group from compounds containing both trityl and benzyl groups, but the yield is only about 35% (Non-Patent Document 1).

[0134] To facilitate the separation operation after the reaction and reduce the loss of the product to the aqueous layer, we investigated the use of a mixed solvent of ethyl acetate and various polar solvents. This process is a continuous process, and if the reaction could be carried out in a mixed solvent with ethyl acetate, the post-treatment solvent in the previous process, the process would be simpler. Therefore, when carrying out this reaction in a mixed solvent, we investigated the optimal mixed system of ethyl acetate and various polar solvents. Under the same reaction conditions as in Example 2-5, 5% palladium carbon powder (PE type hydrous product) (manufactured by NE Chemcat Corporation) was used. The results are shown in Table 3.
