Abstract
Description
Technology Field
[0001] The present invention relates to a topical skin composition incorporating taurine crystals containing a useful ingredient. It relates to a technique for introducing a useful ingredient during taurine recrystallization. Background Technology
[0002] Taurine is a compound with the chemical name 2-aminoethanesulfonic acid and is the most abundant sulfur-containing free amino acid found in living cells. Although taurine is different from the amino acids that make up proteins and is known to act like vitamins or hormones, or to exert various pharmacological effects including those on the brain, nervous system, circulatory system, and hepatobiliary system, it has recently been known to play an important role in skin moisturization.
[0003] As a method for producing taurine, a number of production methods are known, such as extraction from marine products or multiple organic synthesis methods. For example, Patent Document 1 describes a method for obtaining taurine with high purity, and Patent Document 2 describes a method for obtaining taurine with high yield.
[0004] As a technique for purifying taurine, Patent Document 3 describes a method for purifying taurine in which a solution containing taurine and an inorganic salt is brought into contact with a porous solid material exhibiting a molecular sieve effect, thereby adsorbing taurine onto the porous solid material to separate it from the inorganic salt, and subsequently using an organic solvent that may contain water to detach the taurine adsorbed onto the porous solid material. Furthermore, as a means for purifying taurine with high yield, a technique of performing crystallization by adding alcohol is known in the past. That is, when purifying taurine from a hot water or hot water extraction solution of a natural product (such as aquatic products) or a solution from the end of a synthesis reaction, taurine crystals are precipitated in an aqueous alcohol solution, and final taurine crystals are obtained by filtration and drying. Patent Document 4 describes a method for purifying taurine in which impurities are separated or removed by adding alcohol to a taurine-containing reaction solution obtained through a manufacturing process. As a method for purifying the obtained crystal, Patent Document 5 describes a method for purifying taurine in which the crystal precipitated by crystal is separated from the crystal mother liquor, the crystal is treated with an aqueous alcohol solution containing hydrochloric acid, and then subsequently treated with an aqueous alcohol solution not containing hydrochloric acid.
[0005] On the other hand, regarding the technology of incorporating other components into crystals, research has been conducted to use them while ensuring that each component can be stabilized, for components such as vitamins, amino acids, and peptides, which are unstable in aqueous solutions, easily decomposed by oxidation or temperature, and difficult to stably combine. Patent Document 6 indicates that when calcium pantothenate and ascorbic acid are crystallized and mixed, the stability is improved compared to when they are mixed with calcium pantothenate that has not been crystallized. Furthermore, Patent Document 7 describes a method for obtaining a stable liquid formulation by adding water-soluble vitamins in a crystalline state to edible oils such as sunflower oil, corn oil, and cottonseed oil. Additionally, Patent Document 8 describes a lipstick composition containing a crystalline vitamin B3 compound. Prior art literature

[0007] While manufacturing and purification methods for obtaining taurine crystals with high purity and high yield are known, conversely, intentionally incorporating other active ingredients or skin-beneficial ingredients into taurine crystals has not been known until now.
[0008] Furthermore, regarding the technology of incorporating other components into crystals, the composition used in such technology is a mixture of powders in the absence of water or a composition dispersed in oil, and its use is limited to non-aqueous systems.
[0009] If useful organic compounds, peptides, pigments, etc., can be stably contained within water-soluble crystals, it becomes possible to color the crystals or stably incorporate useful components into various formulations. The objective of the present invention is to provide a new means of delivering useful components into the skin. means of solving the problem
[0010] The inventors have discovered, through repeated research, that by adding an aqueous solution of dissolved taurine to a water-soluble organic solvent such as an alcohol or polyol, which has low taurine solubility, or by slowly injecting the water-soluble organic solvent into the aqueous solution to form an interface separated into two phases of the aqueous solution of taurine and the water-soluble organic solvent, it is possible to produce fine needle-shaped crystals of taurine by creating a state of extremely low taurine solubility and proceeding with recrystallization therefrom, and that hollow crystals are formed during recrystallization. During this recrystallization, it is possible to produce fine needle-shaped crystals of taurine in which useful components, such as vitamins or pigments, are supported on the crystal surface or contained in the hollow portion by dissolving or dispersing useful components in either the aqueous solution of taurine or the water-soluble organic solvent used, or both. In addition, it was discovered that in a topical skin composition containing the obtained crystals, it is possible to provide a formulation that has no rough texture of the crystals and imparts a soft, powdery sensation of fine needle-like crystals.

Effects of the invention
[0034] The external skin composition of the present invention contains taurine micro-needle crystals containing a useful ingredient, so the useful ingredient can be stably retained within the composition. Since the taurine crystals in the present invention are formed into micro-needle crystals by recrystallization, it is possible to pierce the crystals into the stratum corneum without feeling the rough texture of the crystals during use, directly transfer the useful ingredient into the skin, and then supply the included ingredient into the skin by dissolving the crystals in the moisture inside the skin. [0036] The topical skin composition according to the present invention contains microacoustic crystals of taurine containing a useful component within the crystals. In the topical skin composition, it is preferable that the microacoustic crystals of taurine are contained in at least a partially dispersed manner.
[0037] The above-mentioned taurine microneedle crystals can be manufactured by creating a state of extremely low solubility of taurine and proceeding with recrystallization (crystal growth) therefrom, by mixing an aqueous solution (A) in which taurine is dissolved with a liquid substance (B) in which taurine is low solubility (e.g., a water-soluble organic solvent), or by bringing the aqueous solution (A) in which taurine is dissolved with a liquid substance (B) in which taurine is low solubility (e.g., forming an interface by slowly injecting the liquid substance (B) into the aqueous solution (A). The obtained taurine microneedle crystals have a controlled crystal form. Additionally, when manufacturing the crystals, a hollow portion is formed within the taurine microneedle crystals, and this hollow portion is defined as the spatial volume.
[0038] Accordingly, the microacicular crystals of taurine have spaces within the microacicular crystals of taurine. It is preferable that the volume of the spaces of the microacicular crystals of taurine occupies 0.001% to 30% of the volume of the taurine crystals, and more preferable that it occupies 0.1% to 30%. Here, the size of the volume of the spaces of the microacicular crystals of taurine is calculated by interpreting microscopic images of the crystals.
[0039] The taurine microneedle crystal containing a useful component in the crystal can be prepared by dissolving or dispersing a useful component (any organic compound component, peptide, protein, or pigment, etc.) in an aqueous solution (A) or a liquid substance (B) during the recrystallization of the taurine. It can also be prepared by incorporating or supporting the useful component during the recrystallization. Here, incorporation refers to at least one of the following: entering into the taurine crystal lattice or containing the useful component in the hollow portion (space) of the taurine crystal. In the former case, the lattice constant increases in correspondence with the expansion of the taurine crystal. In the latter case, the lattice constant does not expand due to incorporation. Support refers to adsorbing or binding to the surface of the taurine crystal. In the present invention, a taurine microneedle crystal containing or supporting a useful component, that is, a taurine microneedle crystal containing a useful component in the crystal, may be referred to as a taurine microneedle crystal containing a useful component. It is desirable that the fine needle-like crystals of taurine have spaces, and that useful components are contained within those spaces.
[0040] The taurine used as a raw material in the present invention is not specifically limited to any that has been conventionally used in pharmaceuticals, quasi-pharmaceuticals, or cosmetics, and includes both synthetic products and extracts from natural products.
[0041] The microcrystals of taurine in the present invention are needle-shaped crystals, but may also be columnar crystals. Here, a needle-shaped crystal refers to a shape in which the long diameter of the crystal is three times or more the short diameter, and a columnar crystal refers to a shape in which the long diameter of the crystal is less than three times the short diameter. The micro needle-shaped or columnar crystals of taurine in the present invention are crystal forms that can be distinguished from the bulk crystals of raw taurine by microscopic observation. An example of a micro needle-shaped crystal is shown in FIG. 2, and an example of a bulk crystal is shown in FIG. 3. Hereinafter, the taurine recrystallized in the present invention will be referred to as taurine micro needle-shaped crystals.
[0042] The size of the taurine microneedle crystals is preferably such that, when used in a topical skin preparation composition, it does not leave a rough texture and provides a soft, powdery finish. It is more preferable that the thickness is 150 micrometers or less and the length is 3000 micrometers or less, more preferable that the thickness is 5 to 150 micrometers and the length is 100 to 3000 micrometers, even more preferable that the thickness is 10 to 100 micrometers and the length is 100 to 2000 micrometers, and most preferable that the thickness is 10 to 60 micrometers and the length is 150 to 2000 micrometers.
[0043] Here, thickness refers to the short diameter of the crystal, and length refers to the long diameter of the crystal. The thickness and length of the crystal can be measured using a digital microscope (e.g., the "Digital Microscope VHX-7000" manufactured by Keyence Co., Ltd.).
[0044] Crystalline forms with a thickness exceeding 150 micrometers and a length exceeding 3000 micrometers (e.g., taurine crystals and taurine crystalline powders commercially available as raw materials) are undesirable because they leave a gritty foreign sensation on the skin when applied.
[0045] In the present invention, among the fine needle-shaped crystals of taurine, particulate crystals with a thickness of less than 5 micrometers or a length of less than 100 micrometers may be included, but it is particularly preferable not to include crystals with a thickness exceeding 150 micrometers. In the present invention, conditions for recrystallization can be set so that needle-shaped crystals with a thickness exceeding 150 micrometers are not formed.
[0046] Specific recrystallization (crystal growth) conditions are a process of growing crystals in an environment in which an aqueous solution (A) in which taurine is dissolved is mixed or brought into contact with a liquid substance (B) in which taurine has low solubility.
[0047] Examples of liquid substances (B) with low solubility of taurine used for recrystallization (crystal growth) include organic solvents, which are not particularly limited but include, for example, polar organic solvents such as acetone, methyl ethyl ketone, dimethyl sulfoxide, dimethylformamide, ethyl acetate, isopropyl myristate, methylheptyl laurate, and jojoba oil; non-polar organic solvents such as dodecane, isododecane, liquid paraffin, squalane, hydrogenated polyisobutene, and olefin oligomers; alcohols such as ethanol and 1-propanol; polyhydric alcohols such as 1,3-butanediol, glycerin, and 1,2-pentanediol. Among them, in terms of the high effect of lowering the solubility of taurine by addition, the liquid substance (B) is preferably a liquid substance (water-soluble organic solvent) having a solubility in water of 1 mass% or more at 25°C, and specifically, it is preferably ethanol, 1-propanol, 2-propanol, 1-butanol, 2-butanol, ethylene glycol, diethylene glycol, 1,2-propanediol, 1,3-propanediol, 1,2-butanediol, 1,3-butanediol, isoprene glycol, diethylene glycol, dipropylene glycol, glycerin, diglycerin, 1,2-pentanediol, 1,2-hexanediol, acetone, methyl ethyl ketone, dimethyl sulfoxide, dimethylformamide, cyclohexylglycerin, or n-hexylglycerin. In terms of using the obtained microneedle crystals as a component of a topical skin preparation, the liquid substance (B) is most preferably ethanol, isopropyl alcohol, ethylene glycol, 1,2-propanediol, 1,3-propanediol, 1,2-butanediol, 1,3-butanediol, isoprene glycol, diethylene glycol, dipropylene glycol, glycerin, diglycerin, 1,2-pentanediol, 1,2-hexanediol, cyclohexylglycerin, or n-hexylglycerin. One of these or a mixture of two or more solvents may be used.
[0048] If the liquid substance (B) is not used, the crystalline form does not become fine needle-shaped crystals, and when incorporated into a topical skin composition, it results in a gritty texture, which is undesirable.
[0049] Regarding the concentration of taurine in the aqueous solution (A) in which taurine is dissolved for use in recrystallization, it is preferable to have 3 mass% or more from the perspective of forming fine needle-shaped crystals when mixed with a solvent with low solubility of taurine, and 28 mass% or less from the perspective of the solubility of taurine in water. If it is less than 3 mass%, crystals do not precipitate unless 100 mass% or more of the liquid substance (B) is added to the aqueous solution (A), and a sufficient amount of crystals is not obtained, so it is not practical in terms of cost and yield. If it exceeds 28 mass%, the recrystallization process is not practical because it becomes difficult to dissolve taurine even when heated to 80°C or higher. In this regard, the concentration of taurine in the aqueous solution (A) in which taurine is dissolved is preferably 5 to 20 mass%, more preferably 6 to 18 mass%, and most preferably 7 to 15 mass%.
[0050] In the aqueous solution (A) in which taurine is dissolved for recrystallization, one or more water-soluble organic solvents with low solubility of taurine may be added as a liquid substance (B) for the purpose of lowering the saturation concentration. In this case, water-soluble organic solvents such as glycerin, 1,3-propanediol, ethanol, isopropyl alcohol, ethylene glycol, 1,2-propanediol, 1,2-butanediol, 1,3-butanediol, isoprene glycol, diethylene glycol, dipropylene glycol, diglycerin, 1,2-pentanediol, 1,2-hexanediol, cyclohexylglycerin, n-hexylglycerin, etc. are preferred. The concentration of the liquid substance (B) (water-soluble organic solvent) is preferably 50 mass% or less, more preferably 30 mass% or less, and most preferably 25 mass% or less with respect to the total amount of the aqueous solution (A). If it exceeds 50 mass%, the solubility of taurine becomes too low, and since a sufficient amount of crystals cannot be obtained, it is not realistic in terms of cost and yield.
[0051] In the present invention, the useful components included in the microacoustic crystals of taurine are not particularly limited. Examples of the useful components include vitamins and vitamin derivatives, hydroquinone and its derivatives, whitening components, anti-inflammatory components, antioxidant components, blood circulation promoting components, cell revitalization components, coenzymes and their intermediate metabolites, sugars, plant extract components, peptides, proteins, and pigments.

[0058] It is possible to include one or more of the above useful components in the fine needle-shaped crystals of taurine.
[0059] The above useful ingredients are not limited to water-soluble ingredients but may also be oil-soluble ingredients. Furthermore, even ingredients that are prone to oxidation, particularly those that could not be incorporated previously due to changes in odor or discoloration over time, can be incorporated by incorporating them into the taurine microneedle crystals, thereby enabling their stable presence. Additionally, since the taurine microneedle crystals in the topical skin composition dissolve and the contained ingredients dissolve after the composition is applied to the skin, the contained ingredients can be applied to the skin in a fresh state. Furthermore, by incorporating bio-derived ingredients such as high molecular weight polysaccharides, peptides like glucose factor, and proteins that are difficult to penetrate the skin into the taurine crystals, the needle crystals in the composition can be directly punctured into the skin to facilitate penetration.
[0060] The amount of the useful component included can be expressed as a percentage of the amount of the useful component in the total weight of the crystal. The percentage of the useful component in the fine needle-shaped crystals of taurine is preferably 0.001 to 30 mass%, more preferably 0.001 to 25 mass%, and most preferably 0.001 to 20 mass%. If the percentage is less than 0.001 mass%, it is not desirable because the effect of the useful component included is not sufficiently obtained.
[0061] In order to suitably obtain fine needle-shaped crystals of taurine having the above-described inclusion ratio, when producing fine needle-shaped crystals of taurine, it is preferable to use the useful component in a state where it is dissolved or dispersed in an aqueous solution (A) in which taurine is dissolved or in a liquid substance (B) with low solubility of taurine. The concentration of the useful component is preferably 0.001 mass% or more in the aqueous solution (A) or the liquid substance (B) in terms of the amount of inclusion component introduced into the taurine crystals. If it is less than 0.001 mass%, it is not desirable because it is impossible to confirm that the inclusion component is contained therein. Furthermore, the upper limit of the concentration of the useful component is not particularly limited.
[0062] Microneedle crystals of taurine containing a useful component may be incorporated into a topical skin composition after being filtered and dried, or the solution from which the crystals were precipitated may be incorporated as is. Furthermore, the useful component may not only be present in the microneedle crystals of taurine but may also be dissolved or dispersed in the medium of the topical skin composition. In that case, the useful component in the microneedle crystals of taurine and the useful component dissolved or dispersed in the medium of the topical skin composition may be the same or different.
[0063] The external skin composition of the present invention can be used primarily as an external skin composition for cosmetics or quasi-drugs. The formulation of the external skin composition of the present invention is not particularly limited, but examples include liquids, lotions, cosmetic emulsions, fluids, creams, aqueous gels, ointments, etc. These formulations can be prepared by conventional methods.

[0161] Example 17
[0162] 100 mL of an aqueous solution containing 10 mass% taurine and 10 mass% glycerin was heated to 50°C and uniformly dissolved, then cooled to 40°C while stirring and left to stand. After standing, 15 mL of an ethanol solution containing 5 mass% uranine (a fluorescent substance) heated to 40°C was slowly injected. Crystals precipitated at the interface separated into two phases were filtered and separated, washed with ethanol, and dried to obtain orange-yellow crystals. Microscopic images of the obtained crystals are shown in Figures 4 to 7.
[0163] As shown in FIGS. 4 to 7, the shape of the obtained taurine microcrystals was needle-shaped. The micro needle-shaped crystals had a maximum thickness of 30 micrometers and a maximum length of 230 micrometers.
[0164] As shown in Figures 5 and 7, the obtained crystals were observed under a fluorescence microscope, and it was confirmed that uranine was contained within the crystals.

[0179] Example 19 and Comparative Example 7
[0180] Microneedle-shaped crystals containing dipotassium glycyrrhizinate were prepared, and the amount of dipotassium glycyrrhizinate penetrating into the skin was measured.
[0181] 100 mL of an aqueous solution containing 10 mass% taurine and 3 mass% dipotassium glycyrrhizinate was heated to 60°C and uniformly dissolved, then cooled to 40°C while stirring, and after standing, 15 mL of ethanol was slowly injected, and the crystals precipitated at the interface separated into two phases were filtered and separated, washed with ethanol, and dried to obtain a crystal product.

[0183] In addition, the obtained crystals were analyzed by liquid chromatography and it was confirmed that they contained 0.101 mass% of dipotassium glycyrrhizinate.
[0184] A composition of the table below containing 10 mass% of this fine needle-shaped crystal, and a composition of a comparative example in which dipotassium glycyrrhizinate was dissolved to the same concentration as this were prepared, and the amount of dipotassium glycyrrhizinate penetrating into the skin was compared when the obtained composition was applied to human skin.
[0185]
[0186] To evaluate the amount of penetration into the skin, each sample was applied to human excised skin at a rate of 10.0 mg / square centimeter using a vertical Franz-type diffusion cell with an effective diameter of 2.0 centimeters, an area of 3.14 square centimeters, and a receptor solution volume of 2.4 mL.
[0187] After 6 hours, the stratum corneum and the epidermis and dermis were separated from the sample skin by tape stripping, and the dipotassium glycyrrhizinate contained in each of the stratum corneum, epidermis and dermis, and receptor solution was quantified by high-speed liquid chromatography.
[0188] As shown in FIG. 10, regarding the composition of Example 19, compared to the composition of Comparative Example 7, the penetration amount of dipotassium glycyrrhizinate throughout the skin is about 2.5 times, and it was confirmed that by incorporating the useful component into the microacoustic crystals, the useful component penetrates the epidermis, dermis, and receptor solution.

[0292] Test Example 1
[0293] In the taurine containing various beneficial components prepared above, a test was conducted to demonstrate that the beneficial components are contained within the taurine crystals. The following is the result of X-ray diffraction of the astaxanthin-containing taurine needle crystals prepared in Example 14.
[0294] As a control, needle-shaped crystals of taurine alone were prepared by the following process.
[0295] 100 mL of an aqueous solution containing 10% taurine was heated to 50°C and uniformly dissolved, then cooled to 30°C while stirring, and after standing, 15 mL of ethanol was slowly injected. Crystals precipitated at the interface separated into two phases were filtered and separated, washed with ethanol, and dried to obtain colorless transparent crystals.
[0296] X-ray diffraction of taurine needle crystals and astaxanthin-containing taurine needle crystals was performed using an X-ray diffraction device, model number SmartLab, manufactured by Rigaku Co., Ltd. X-rays (wavelength = 0.154 nm), with an output of 45 kV and 200 mA, and a HyPix-3000 detector, measurements were performed using the 2θ / θ measurement method with a scanning speed of 10° / min for a range of 2θ from 10 to 80°.
[0297] The obtained X-ray diffraction pattern is shown in Fig. 1. The top is the astaxanthin-containing taurine crystal, and the bottom is the diffraction pattern from the taurine crystal.
[0298] From the above results, astaxanthin-containing taurine crystals and taurine crystals do not exhibit the same diffraction pattern. The diffraction angles of the 111th and 200th planes are summarized in Table 8 below. Specifically, the lattice constants were calculated from the following data using Bragg's equation for the spread of the 111th and 200th planes (a decrease in 2θ indicates a spread of the plane spacing).
