[0001] This application claims the benefit of priority to U.S. Provisional Application No. 62 / 741,772 filed October 5, 2018, the full text of which is by reference.
[0002] Technology field
[0003] The present disclosure relates to a formulation for an anti-FGFR2 (fibroblast growth factor receptor 2) antibody, e.g., an anti-FGFR2-IIIb antibody, which, in some embodiments, can be stored for a long period in a form that is immediately available for use, e.g., as a liquid, and in some embodiments can be administered intravenously, e.g., by intravenous (IV) infusion. Background Technology
[0004] The present disclosure relates to specific formulations of antibodies that bind to FGFR2, e.g., the spliced form FGFR2-IIIb (fibroblast growth factor receptor 2 IIIb). FGFR2 is one of four FGFR proteins (FGFR1, 2, 3, and 4). FGFR is characterized by multiple alternative splicing of its mRNA, resulting in various isoforms (Ornitz et al., J. Biol. Chem. 271:15292, 1996; see also Swiss-Prot P21802 and isoforms P21802-1 to P21802-20 for the sequences of FGFR2 and its isoforms). In the case of FGFR2, selective splicing results in, for example, the isoforms FGFR2-IIIb and FGFR2-IIIc (or directly FGFR2b and FGFR2c). The FGFR2-IIIb form of FGFR2 (or denoted as K-sam-II) is a high-affinity receptor for both FGF1 and KGF family members (FGF7, FGF10, and FGF22), whereas FGFR2-IIIc (also denoted as K-sam-I) binds well to both FGF1 and FGF2 but does not bind to KGF family members (Miki et al., Proc. Natl. Acad. Sci. USA 89:246, 1992). FGFR2-IIIb is the only receptor for KGF family members (Ornitz et al., 1996, op. cit Therefore, it is also designated as KGFR.
[0005] Inhibitors of FGFR2 may include antibodies. For example, U.S. Patent No. 8,101,723 B2 describes, for example, monoclonal antibodies that bind to human FGFR2-IIIb but bind less well or not to FGFR2-IIIc, and vice versa. U.S. Patent Publication No. 2015-0050273 A1 describes specific afucosylated antibodies that bind to FGFR2-IIIb. The present disclosure describes formulations that may be used for antibodies described, for example, in U.S. Patent No. 8,101,723 B2 and U.S. Patent Publication No. 2015-0050273 A1.

[0089] Post-Foucauldron port- FGFR2 antibodies
[0090] In some embodiments, the anti-FGFR2 antibody described herein has a fucose-deficient carbohydrate structure attached (directly or indirectly) to the Fc region, i.e., the antibody is defucosylated. In some embodiments, the defucosylated antibody is an IgG1 or IgG3 antibody defucosylated from Asn297.
[0091] In this specification, an antibody is considered "defucosylated" when a plurality of such antibodies comprises at least 95% defucosylated antibodies. The amount of fucose can be determined by calculating the average amount of fucose within the sugar chains in Asn297 against the sum of all sugar structures attached to Asn297 (e.g., complex, hybrid, and high-mannose structures). Non-limiting exemplary methods for detecting fucose in antibodies include MALDI-TOF mass spectrometry (see, e.g., WO 2008 / 077546), HPLC measurement of emitted fluorescence-labeled oligosaccharides (e.g., Schneider et al., "N-Glycan analysis of monoclonal antibodies and other glycoproteins using UHPLC with fluorescence detection," Agilent Technologies, Inc. (2012)); [Lines, J. Pharm . Biomed .Analysis , 14: 601-608 (1996)]; [Takahasi, J. Chrom [Refer to ., 720: 217-225 (1996)]), capillary electrophoresis of emitted fluorescence-labeled oligosaccharides (e.g., [Ma et al., Anal. Chem . [See , 71: 5185-5192 (1999)]), and HPLC using detection by pulse amperometrics to measure monosaccharide composition (e.g., literature [Hardy, et al., Analytical Biochem.Includes [see , 170: 54-62 (1988)]. In some embodiments, in the composition of the defucosylated antibody of this specification, fucose cannot be detected by one or more of these methods.
[0092] Asn297 refers to an asparagine residue located at position approximately 297 in the Fc region (EU numbering of Fc region residues); however, in a given antibody sequence, Asn297 may also be located approximately ±3 amino acids upstream or downstream of position 297, i.e., between positions 294 and 300, due to small sequence variations of the antibody. In the FGFR2-IIIb antibody described herein, Asn297 is sequence QY N It is found in ST and is underlined in bold at sequence number 2 of the sequence table shown below.
[0093] Fucosylated variants may have improved ADCC functionality. For example, US Patent Publication US 2003 / 0157108 (Presta, L.); Refer to US 2004 / 0093621 (Kyowa Hakko Kogyo Co., Ltd). For examples of publications related to "defucosylated" or "fucose-deficient" antibodies, see US 2003 / 0157108; WO 2000 / 61739; WO 2001 / 29246; US 2003 / 0115614; US 2002 / 0164328; US 2004 / 0093621; US 2004 / 0132140; US 2004 / 0110704; US 2004 / 0110282; US 2004 / 0109865; WO 2003 / 085119; WO 2003 / 084570; WO 2005 / 035586; WO 2005 / 035778; WO2005 / 053742; WO2002 / 031140; Literature[Okazaki et al. Examples of cell lines capable of producing defucosylated antibodies include Lec13 CHO cells deficient in protein fucosylation (Literature [Ripka et al. Arch. Biochem. Biophys. 249:533-545 (1986)]; US patent application US 2003 / 0157108 A1 (Presta, L.); and WO 2004 / 056312 A1 (Adams et al .), particularly Example 11), and knockout cell lines, e.g., cell lines deficient in the functional alpha-1,6-fucosyltransferase gene FUT8, e.g., knockout CHO cells (e.g., literature [Yamane-Ohnuki] et al. Biotech. Bioeng. 87: 614 (2004)]; [Kanda, Y. et al., Biotechnol. Bioeng. Includes , 94(4):680-688 (2006)]; and WO2003 / 085107).
[0094] The anti-FGFR2 antibodies of this specification may also have a bisected oligosaccharide, for example, in which a two-antenna-type oligosaccharide attached to the Fc region of the antibody is bisected by GlcNAc. Such antibodies may have reduced fucosylation and / or improved ADCC function. Examples of such antibodies are, for example, WO 2003 / 011878 (Jean-Mairet et al .); US Patent 6,602,684(Umana et al .); and US 2005 / 0123546(Umana et al It is described in .). In some embodiments, the FGFR2 antibody has at least one galactose residue in the oligosaccharide attached to the Fc region. Such an antibody may have an enhanced CDC function. Such an antibody is, for example, WO 1997 / 30087 (Patel et al It is described in WO 1998 / 58964 (Raju, S.); and WO 1999 / 22764 (Raju, S.).
[0095] In a specific embodiment of the present invention, a defucosylated FGFR2 antibody mediates ADCC more effectively in the presence of human effector cells than an antibody having the same amino acid sequence containing fucose. Generally, ADCC activity can be determined using the in vitro ADCC assay disclosed in U.S. Publication 2015-0050273 A1, but other assays or methods for determining ADCC activity are considered, for example, in animal models.
[0096] In some embodiments, the FGFR2 antibody comprises the heavy chain and light chain sequences of SEQ ID NOs 2 and 3. In other embodiments, the antibody comprising the heavy chain and light chain sequences of SEQ ID NOs 2 and 3 is defucosylated.

In some embodiments, the gastric cancer including FGFR2 amplification overexpresses FGFR2IIIb to a greater extent than FGFR2IIIc. In some embodiments, the gastric cancer including FGFR2 amplification expresses FGFR2IIIb at a normalization level greater than 2, 3, 5, or 10 times the normalization level of FGFR2IIIc expression. In some embodiments, the expression level is normalized to GUSB. In some embodiments, the gastric cancer overexpresses FGFR2IIIb but does not include FGFR2 gene amplification. In some embodiments, the overexpression is mRNA overexpression. In some embodiments, the overexpression is protein overexpression.
[0140] FGFR2IIIb gene amplification can be determined by any suitable method in the art, including but not limited to in situ hybridization (ISH). In some embodiments, FGFR2 amplification comprises more than 3 FGFR2:CEN10 (chromosome 10 centromeres).
[0141] FGFR2IIIb mRNA overexpression can be determined by any suitable method in the art, including but not limited to methods including quantitative PCR (qPCR). The term "FGFR2IIIb mRNA overexpression" means elevated levels of FGFR2IIIb mRNA regardless of the cause of the elevated levels (i.e., whether the elevated levels are the result of increased transcription and / or decreased degradation of mRNA, other mechanisms, or a combination of mechanisms).
[0142] FGFR2IIIb protein overexpression may be determined by any suitable method in the art, including but not limited to antibody-based methods, such as immunohistochemistry (IHC). In some embodiments, IHC staining is scored according to methods in the art. The term “FGFR2IIIb protein overexpression” means an elevated level of FGFR2IIIb protein regardless of the cause of the elevated level (i.e., whether the elevated level is the result of increased translation and / or decreased degradation of the protein, other mechanisms, or a combination of mechanisms). In some embodiments, 1+, 2+, or 3+ staining of tumor cells by IHC indicates FGFR2IIIb overexpression. For example, overexpression may be determined by an IHC signal of 1+, 2+, or 3+ in at least 10% of tumor cells, e.g., at least 20%, 30%, 40%, or 50% of tumor cells. In some embodiments, 2+ or 3+ staining of tumor cells by IHC indicates FGFR2IIIb overexpression. [0143] In some embodiments, FGFR2 overexpression may be recorded as an "H score." For example, in some such embodiments, the tumor is a bladder cancer tumor. To determine the H score, the first membrane staining intensity may be determined for cells in a fixed field to obtain a score of 0, 1+, 2+, or 3+ via IHC, and the H score may be calculated using the following formula: 1 × (%) of cells visualized with an IHC intensity of 1+ + 2 × (%) of cells visualized with an IHC intensity of 2+ + 3 × (%) of cells visualized with an IHC intensity of 3+. Theoretically, the H score may range from 0 to 300, and is 300 when all cells in the field have an IHC stain of 3+. In some embodiments, the patient to be treated has an initial H score for FGFR2, e.g., FGFR2b (e.g., FGFR2IIIb), greater than 20, e.g., greater than 30, greater than 40, greater than 50, or greater than 100, or in the range of 20 to 300, 20 to 100, 20 to 50, 20 to 40, or 20 to 30. In some embodiments, the patient has an H score greater than 10 or is within the range of 10 to 20 or 15 to 20. In other embodiments, the patient has an H score of 0 to 10, which may indicate a lack of FGFR2 overexpression. In some such embodiments, the patient is a bladder cancer patient.

[0148] Examples
[0149] The embodiments discussed below are intended to be purely illustrative of the invention and should not be construed as limiting the invention in any way. While efforts have been made to ensure accuracy regarding the numbers used (e.g., quantities, temperatures, etc.), some experimental error and deviation must be taken into account.
[0150] Examples 1: Formulation Materials and methods used in the study
[0151] I. Antibody production
[0152] The anti-FGFR2-IIIb antibody used in the following examples was generated from Chinese hamster ovary (CHO) cell lines lacking the FUT8 gene (α1,6-fucosyltransferase), and various lots that underwent changes during the fermentation and purification processes were used for antibody development. The anti-FGFR2-IIIb antibody is a defucosylated humanized IgG1 monoclonal antibody. A knowledge-based formulation development approach was used to identify an appropriate composition that provides maximum stability for the protein. To this end, both the endogenous properties of the molecule and exogenous formulation components that may affect the stability of the protein were considered. Studies were conducted to identify appropriate components for producing a liquid formulation of 10 to 20 mg / ml that can be diluted for IV infusion.
[0153] Raw materials: L-histidine (PN H3911), L-histidine.HCl (PN H5659), and sodium chloride (PN S9888) were obtained from Sigma Aldrich. L-arginine.HCl (PN 2067-06), citric acid (PN 0119), sodium citrate (PN 3627), and polysorbate 20 (PN 4116) were obtained from JT Baker (Thermo Fischer Scientific).
[0154] Containers / Capsules: 3 cc Type I West Pharmaceutical glass vials (PN 68000368) and 13 mm caps (PN 19700116) from West Pharmaceutical Services, Inc. (West Pharmaceutical Services, Inc.; Pennsylvania, USA), polypropylene tubes, and glass HPLC vials were used to fill the formulation.
[0155] II. General formulation procedure
[0156] The formulation was prepared by dialyzing the drug substance with a target formulation buffer using an MWCO 20 kD dialysis membrane. The formulation was filtered in a laminar flow hood using a 0.2 µm filter unit and filled into an appropriate container / stopper system for stability evaluation. Individual vials were withdrawn at predetermined times under specified storage conditions for analysis.
[0157] III. Analysis method
[0158] Stability samples were analyzed using the initial development analysis described below.
[0159] Visual inspection Visual evaluation was performed on a black and white background under fluorescent lighting.
[0160] Protein concentrationProtein concentration was determined by UV absorbance at 280 nm using a theoretical absorption coefficient of 1.43 cm⁻¹ [g / ℓ]⁻¹. Samples were diluted within the linear absorbance range using appropriate formulation buffers and measured against Dulbecco phosphate-buffered saline (DPBS) water. OD measurements were performed on a Beckman Coulter DU800 UV-Vis spectrophotometer (Beckman Coulter, Incorporated, California, USA).
[0161] pH : The buffer pH was determined using a calibrated Beckman Coulter pHi560 pH meter (Beckman Coulter, Incorporated, California, USA).
[0162] osmotic pressure The buffer osmotic pressure was measured by vapor pressure using the Wescor VAPRO® system (Wescor, Incorporated, Utah, USA).
[0163] Differential Scanning Calorimetry ( DSC ) analyze High-sensitivity differential scanning calorimetry (HSDSC) analysis was performed using VP-DSC (MicroCal, Northampton, Massachusetts, USA). Approximately 0.5 mg of each sample was loaded into a sample cell, and an equal amount of matching dialysis buffer was loaded into a reference cell. Samples were scanned at a rate of 1°C / min at 20 to 90°C or 100°C. Data were analyzed using Origin 7.0 data analysis software (OriginLab, Massachusetts, USA).
[0164] ion exchange Chromatography (IEX)Cation exchange chromatography is a method for separating protein variants based on net surface charge. While the isoelectric point (pI, the pH at which the protein has no net charge) of a protein is determined by its primary amino acid sequence, the net charge of the protein variant is based on the protein's isoelectric charge and the pH of the performance buffer. For example, in a performance buffer at pH 7, which is about 1 unit below the antibody's pI of 8.4, the protein has a net positive charge and electrostatically binds to an oppositely weakly charged acidic carboxylate functional group located on the ethylvinylbenzene-divinylbenzene substrate and the cross-linked Propac WCX-10 column resin. Using a high-performance liquid chromatography system equipped with UV detection at 280 nm, protein variants are separated and eluted from the column when an increasing salt gradient is applied, where variants with the weakest ionic interactions begin to elute first, and variants with stronger ionic interactions elute later at higher salt concentrations. The isoproteins eluted sequentially into three regions: acidic, major, and basic, with the highest peak being the major isoprotein peak. Based on the peak regions, each region had a calculated relative peak percentage. A weak cation exchange HPLC method was performed using a Dionex WCX-10 PROPAC™ 4×250 mm column (Thermo Fisher Scientific). Samples were run on an Agilent 1200 Series HPLC, and chromatograms were integrated using Chemstation software (Agilent Technologies, California, USA). Mobile phase A was 10 mM HEPES at pH 7.0, and mobile phase B was 10 mM HEPES at pH 7.0 and 100 mM sodium chloride. A gradient method was used at 40 to 90% B for 28 minutes, and UV detection was performed at 280 nm. Acidity, basicity, and major peak percentages were recorded from the total area percentage of each peak-associated absorbance.
[0165] Size-Exclusion Chromatography (SEC) Size exclusion chromatography is a method for separating proteins based on their size. This method uses porous resins to separate molecules of different sizes. Larger molecules elute earlier because they cannot access the resin pores and pass through the column without obstruction. Smaller molecules can access the resin pores, and the tortuosity within the resin particles can increase the path length through the column. Therefore, smaller molecules elute after larger molecules. Two methods were used for size exclusion chromatography. For the first method, a Tosoh G3000SWXL 7.8×300 mm column was used with 100 mM sodium phosphate and 700 mM arginine at pH 6.8 as the mobile phase for 30 minutes at 0.5 mL / min (Sigma-Aldrich, Incorporated, Missouri, USA). The second method utilized a Sepax Zenix SEC-300 7.8×200 mm column with 100 mM sodium and 400 mM sodium chloride at pH 6.8 as the mobile phase for 12 minutes at 1 mL / min (Sepax Technologies, Incorporated, Delaware, USA). Samples were run on Agilent 1100 and 1200 series HPLCs, and chromatograms were integrated using ChemStation software with detection set at 280 nm. The percentage of aggregates, low molecular weight (LMW), and major peaks were recorded as the percentage of the total area of the absorbance associated with each peak.
[0166] light scattering : Ultraviolet absorbance at 280 nm using a spectrophotometer (UV A 280 ) was measured. Beer's law, that is, A = ε l cThe concentration of the antibody samples was calculated using (ε = absorption coefficient; l = length of the solution through which light passes (path length of the cuvette) (cm), and c = concentration of the solution). The theoretical absorption coefficient was used to determine the antibody concentration (e.g., the theoretical absorption coefficient for the anti-FGFR2 antibody is 1.43 (OD·mL) (mg·cm)). The concentration (unit: mg / mL) for each sample preparation was calculated as follows: Concentration = (Absorbance at A280 - Absorbance at A350) × (Dilution factor) / (Absorbance coefficient × Path length). The mean and standard deviation were calculated for each sample.
[0167] Examples 2: Solubility in water
[0168] The anti-FGFR2-IIIb antibody was concentrated to approximately 100 mg / mL and dialyzed in water. The sample was then further concentrated to 180 mg / mL. The anti-FGFR2-IIIb antibody was soluble in water at 180 mg / mL. It did not reach maximum solubility.
[0169] Examples 3: Hang- FGFR2 - IIIb Initial freeze / thaw stability of antibodies
[0170] A freeze-thaw study was performed on the anti-FGFR2-IIIb antibody. The antibody was concentrated to 11.6 mg / mL and formulated in 1× PBS at pH 7.4, with or without 0.01% polysorbate 20. The formulated solution was filled into vials, and freeze-thaw cycles were performed up to three times at -70°C to ambient temperatures. Sample vials were analyzed by visual inspection and SE-HPLC. The freeze-thaw results are shown in Table 1. A slight increase in aggregates was observed over the three freeze-thaw cycles. The results suggest that 1× PBS is acceptable for the formulation.
[0171]
[0172] Examples 4: Hang- FGFR2 - IIIb Antibody oxidation research
[0173] The anti-FGFR2-IIIb antibody has a total of 10 methionine residues; four of the methionines are in each heavy chain, and none of the methionines are located on CDR1. Additionally, there is one methionine in the light chain.
[0174] Experiments were conducted to evaluate the sensitivity of methionine to oxidation by incubating anti-FGFR2-IIIb antibodies with 0.01%, 0.1%, or 1.0% hydrogen peroxide overnight at room temperature. Subsequently, the oxidized samples were characterized by trypsin mapping LC / MS to qualitatively determine the oxidation levels of each methionine residue. The activity of the oxidized samples was determined by cell-based assays for efficacy, and binding affinity was analyzed using Viacore™ (GE Healthcare). The peptide mapping LC / MS characterizations shown in Table 2 revealed that light-chain methionine 5 and medium-chain methionine 81 were less sensitive to oxidation. Medium-chain methionine 249, 355, and 425 were more sensitive to oxidation. All oxidized samples exhibited efficacy similar to the control group by ELISA analysis.
[0175]
[0176] The binding of the anti-FGFR2-IIIb antibody to FGFR2b and protein A was measured by Viacore. Oxidation of heavy chain methionine did not interfere with the binding of the anti-FGFR2-IIIb antibody to FGFR2b (Figure 1 and Table 3). Increased oxidation interfered with binding to protein A. Considering that the activity of the anti-FGFR2-IIIb antibody was not significantly affected in highly oxidized samples, the oxidative degradation of the anti-FGFR2-IIIb antibody was not significant.
[0177]
[0178] Examples 5: Protein Regarding stability buffer The effect of pH
[0179] Formulation pH affects protein stability. The pH of the formulation can influence biochemical degradation pathways, e.g., deamidation, isomerization, and oxidation, as well as biophysical degradation, e.g., aggregation and fragmentation resulting from interactions between proteins and their environment. Buffer pH was investigated as a formulation variable that may affect product stability. The effect of pH on the stereochemical stability and structural stability of the anti-FGFR2-IIIb antibody during storage was evaluated. The anti-FGFR2-IIIb antibody was purified in two steps and formulated into nine different isotonic formulations (Formulations 1 to 9 in Table 4) with pH values ranging from 4.0 to 8.0. The protein concentration of the buffer exchange solution was adjusted to 1 mg / mL, and the solution was filled into vials for DSC analysis and evaluation of isothermal stability at 40°C and 25°C. The evaluated formulations are listed in Table 4.
[0180]
[0181] Differential Scanning Calorimetry ( DSC ) Structural stability based on analysis
[0182] DSC thermograms were generated and analyzed for formulations 1 to 9 in Table 4. Formulations 1 to 3 are marked with * in Table 5, and the results are shown in Figure 2. Three transitions were observed upon heating of the samples. After the release transition, IgG molecules formed insoluble aggregates and precipitated from the solution. Table 5 shows the release temperature (T M 1) increases with increasing pH and reaches a maximum T at approximately 69°C at pH 6 and pH 7. M It indicates that it reached 1. The buffer species did not have a significant effect on the annealing temperature.
[0183]
[0184] Isothermal stability
[0185] The effect of buffer pH on the formation of aggregates, clips (fragments), and charged variants was evaluated by monitoring sample stability under accelerated storage conditions at 40°C and 25°C.
[0186] Changes in aggregates and clips were determined by SE-HPLC. Representative SE-HPLC chromatograms of selected anti-FGFR2-IIIb antibody pH screening samples are shown in Figure 3. The main peak represents the monomer, the peak eluted before the monomer is the aggregate, and the peak eluted later is the clip. Soluble aggregates increased rapidly in the pH 4 formulation at 40°C; moderate increases in soluble aggregates with increasing pH were observed in pH 5.0 and pH 8.0 buffers. A similar effect of pH on clip formation was observed.
[0187] The effect of buffer pH on aggregates (Fig. 4) and clips (Fig. 5) at 40°C was analyzed by SE-HPLC. Formulations 1, 2, 3, 7, 8, and 9 listed in Table 5 were tested. Within the tested pH range, clip formation was most pronounced at pH 4. At neutral pH, clip formation was negligible even after storage for one month at 40°C, as shown in Fig. 5. No apparent increase in aggregates was observed in the formulations from pH 5 to pH 8 after storage for at least two months at 25°C (Fig. 6). No apparent increase in clips was observed in the formulations from pH 5 to pH 8 after storage for at least two months at 25°C (Fig. 7).
[0188] Changes in the distribution of charged variants are another common degradation pathway for antibodies and are typically associated with the pH of the formulation buffer. Charged variants of the anti-FGFR2-IIIb antibody were analyzed using the weak cation-exchange HPLC (WCX-HPLC) method. A representative chromatogram is shown in Fig. 8. Peaks appearing before the main peak represent acidic species, and peaks appearing after the main peak represent basic species. The increase in acidic species is attributed to the deamidation of asparagine in IgG molecules. Changes in the charged variant profile occurred rapidly at 40°C. Therefore, changes in the charge profile were monitored at lower temperatures, e.g., 25°C. Figs. 9 through 11 illustrate the effect of buffer pH on charged variants at 25°C as determined by weak cation-exchange high-performance liquid chromatography (WCX-HPLC). As shown in Fig. 9, acidic species increased rapidly under basic pH conditions, particularly at pH 8.0. Samples formulated at pH 5 to pH 6 remained stable even after 2 months of storage at 25°C. Figure 10 shows the effect of pH on the percentage of basic variants during storage at 25°C for 0 to 2.5 months. Figure 11 shows the effect of pH on the main peak during storage at 25°C for 0 to 2.5 months.
[0189] The results of the pH screening study indicate that the stability of the anti-FGFR2-IIIb antibody under stress storage conditions depends significantly on the pH of the formulation. At basic pH, the aggregation and formation of acidic variants were the major degradation pathways. The anti-FGFR2-IIIb antibody was determined to be most stable in the pH range of 5.0 to 6.0.
[0190] Examples 6: Protein Effect of excipients on stability
[0191] Formulation excipients, such as buffer types and extenders, were analyzed for their effect on product stability. To evaluate the effect of excipients on the stability of the anti-FGFR2-IIIb antibody, various citrate, phosphate, and histidine buffers were tested. Seven different isotonic solutions buffered at pH 6.0 tested are listed in Table 6.
[0192]
[0193] The protein concentration was adjusted to 20 mg / mL, and the solution was filled into vials. Formulations 1 to 4 in Table 6 were analyzed by DSC. Physical stability was evaluated for all formulations, and isothermal stability was evaluated for formulations 1 to 5 at 40°C, 25°C, and 5°C.
[0194] DSC Structural stability based on analysis
[0195] The unfolding temperatures of various formulations of the anti-FGFR2-IIIb antibody were determined by DSC analysis and are summarized in Table 7. Precipitation was observed in all formulations after heating to 90°C. In the formulations, the surfactant did not prevent the antibody from precipitating after denaturation. As shown in Table 7, the formulations had similar melting temperatures.
[0196]
[0197] Three thermal transitions were detected when heating various formulations of the anti-FGFR2-IIIb antibody. The anti-FGFR2-IIIb antibody began to unwind at 52.7°C in the pH 4 formulation, and the unwinding temperature (Tm) increased with increasing pH, reaching a maximum Tm1 at approximately 70°C at pH 6 and pH 7, and Tm decreased slightly at pH 8.
[0198] Physical stability
[0199] Since protein pharmaceuticals are prone to aggregation when exposed to shear stress, physical stability studies were conducted to evaluate the effect of excipients on the stability of anti-FGFR2-IIIb antibodies against multiple freeze / thaw cycles and vigorous stirring. Stability was assessed by visual observation of insoluble and soluble aggregates, A350 light scattering, and SE-HPLC. The antibody concentration in the samples was 20 mg / mL.
[0200] The sample vials were placed horizontally on an orbital shaker and subjected to stirring stress by shaking the samples at 500 RPM at room temperature for 77 hours. To evaluate the effect of polysorbate 20 on preventing aggregation formation, a head-to-head comparison was performed on arginine formulations containing 0%, 0.01%, 0.05%, and 0.10% polysorbate 20. As shown in Table 8 and Figure 12, no apparent increase in soluble aggregates was detected by SE-HPLC in any of the formulations.
[0201] A freeze / thaw study was also performed by freezing the samples at -70°C and thawing them at ambient temperature over five cycles. As shown in Figure 13, the only apparent change was observed in soluble aggregates in the histidine / NaCl formulation, which increased with increasing freeze / thaw cycles. The addition of 0.01% polysorbate 20 to the formulation improved the stability of the anti-FGFR2-IIIb antibody against shaking stress.
[0202]
[0203] Isothermal stability
[0204] The effects of buffer species and extenders on the formation of aggregates, clips, and charged variants were evaluated by monitoring the stability of the anti-FGFR2-IIIb antibody under accelerated storage conditions at 40°C, 25°C, and 5°C. Changes in aggregates and clips were monitored using SE-HPLC. The following formulations were tested: 20 mM citrate, 150 mM L-arginine, and 0.01% polysorbate 20; 20 mM phosphate, 150 mM L-arginine, and 0.01% polysorbate 20; 20 mM histidine, 150 mM L-arginine, and 0.01% polysorbate 20; 20 mM histidine, 150 mM NaCl, and 0.01% polysorbate 20; and 20 mM histidine, 150 mM L-arginine, polysorbate 20 none.
[0205] As shown in Fig. 14, aggregates formed at a higher rate in the citrate and phosphate formulations at 40°C. Minor fragmentation was observed in all formulations after 1 month of storage at 40°C. All formulations showed the same increase in aggregation after 3 months of storage at 25°C (Fig. 15).
[0206] The formulation excipients did not affect the charge profile of the anti-FGFR2-IIIb antibody during storage. As shown in Figure 16, there was an increase in acidic variants in all formulations. Figure 17 shows that there was a decrease in basic variants over time in all formulations, and Figure 18 shows the effect of excipients on the major peaks over time.
[0207] Based on antibody stability studies, L-histidine did not have a significant effect on antibody stability among the buffer species evaluated. In sodium citrate and sodium phosphate, aggregates increased after 1 month at 40°C. Therefore, L-histidine is more stable than sodium citrate or sodium phosphate; aggregates increased at a faster rate in the citrate and phosphate formulations.
[0208] Among the evaluators, NaCl produced aggregate formation induced by vigorous shaking and exposure to freezing / thawing processes. The most stable antibody profile was obtained with the L-arginine formulation.
[0209] Examples 7: pH Evaluation Study
[0210] The effect of pH on the chemical and physical stability of the formulations was tested. The following formulations were tested at pH 5.5, 6.0, and 6.5: (a) 20 mM histidine, 150 mM arginine, 0.01% polysorbate 20 and (b) 20 mM histidine, 270 mM sucrose, 0.01% polysorbate 20 (Table 9). The formulations were adjusted to 20 mg / mL and filled into glass vials.
[0211]
[0212] Effect of pH on physical stability
[0213] Stability against multiple freezing / freezing cycles and vigorous stirring was evaluated. Stability was assessed by visual observation of insoluble and soluble aggregates, A350 light scattering, and SE-HPLC.
[0214] I. mechanical stirring
[0215] Stirring stress was applied to the samples by placing the sample vials horizontally on an orbital shaker and shaking at 500 RPM at room temperature for 72 hours. No clear evidence of soluble aggregates was detected by SE-HPLC in any of the formulations. Figure 19 shows the effect of mechanical stress on aggregation in the histidine / arginine and histidine / sucrose formulations over time at pH 5.5 to 6.5 (pH 5.5, 6.0, and 6.5). Figure 19 shows the effect of mechanical stress on the clips in the histidine / arginine and histidine / sucrose formulations at pH 5.5 to 6.5. After 72 hours of mechanical stirring, there was no significant change in light scattering or concentration at A350 nm (as shown in Table 10).
[0216]
[0217] II. Freeze thawing stability
[0218] Freeze / thaw studies were performed by freezing samples at -70°C and thawing them at ambient temperature over five cycles. Figures 21 and 22 show the effects of freeze-thaw on aggregation and clipping in histidine / arginine and histidine / sucrose formulations at pH 5.5 to 6.5. In Figure 21, there was no apparent increase in soluble aggregates detected by SE-HPLC in either the histidine / arginine or histidine / sucrose formulations at pH 5.5 to 6.5. In Figure 22, no apparent evidence of clipping was detected by SE-HPLC in any of the formulations.
[0219]
[0220] After freezing and thawing, the concentration and light scattering remained unchanged.
[0221] I. Effect of pH on isothermal stability
[0222] The effect of buffer pH on the formation of aggregates, clips, and charged variants was evaluated by monitoring sample stability under accelerated storage conditions at 40°C and 25°C, and real-time stability at 5°C. The following formulations were tested: (A) 20 mM histidine, 150 mM arginine, 0.01% polysorbate 20 ("histidine / arginine formulation"), and (B) 20 mM histidine, 150 mM sucrose, 0.01% polysorbate 20 ("histidine / sucrose formulation").
[0223] Aggregation and clip formation increased with pH in the histidine / arginine and histidine / sucrose formulations after 1 month at 40°C and 3 months at 25°C. The histidine / sucrose formulation showed a slightly lower aggregation rate. After 6 months at 5°C, aggregation remained unchanged, and clip formation increased by about 1% in both the histidine / arginine and histidine / sucrose formulations.
[0224] For example, FIGS. 23A to 23C show the formation of aggregates in the histidine / arginine formulation under various pH conditions measured over (A) 1 month at 40°C, (B) 3 months at 25°C, and (C) 6 months at 5°C. FIGS. 24A to 24C show the formation of clips in the histidine / arginine formulation under various pH conditions (pH 5.5 to 6.5) measured over (A) 1 month at 40°C, (B) 3 months at 25°C, and (C) 6 months at 5°C.
[0225] FIGS. 25A to 25C show aggregate formation in the histidine / sucrose formulation under various pH conditions measured over (A) 1 month at 40°C, (B) 3 months at 25°C, and (C) 6 months at 5°C. FIGS. 26A to 26C show antibody clip formation in the histidine / sucrose formulation under various pH conditions (pH 5.5 to 6.5) measured over (A) 1 month at 40°C, (B) 3 months at 25°C, and (C) 6 months at 5°C.
[0226] The charged isoform also remained unchanged after 6 months at 5°C. For example, FIGS. 27A to 27C show acidic variants in the histidine / sucrose formulation with increased acidity after (A) 1 month at 40°C, (B) 3 months at 25°C, and (C) 6 months at 5°C.
[0227] Additionally, FIGS. 28A to 28C illustrate the effect of pH between 5.5 and 6.5 on the formation of acidic variants in a histidine / arginine formulation comprising 20 mM histidine, 150 mM arginine, and 0.01% polysorbate 20. FIG. 28A illustrates the formation of acidic variants, indicating that the % of acidic variants generally remained below 40% after 1 month at 40°C at pH 5.5 to 6.0. FIG. 28B illustrates the formation of acidic variants, indicating that the % of acidic variants generally remained below 30.0% after 3 months at 25°C at pH 5.5 to 6.0. FIG. 28C illustrates the formation of acidic variants, indicating that the % of acidic variants generally remained below 25% after 6 months at 5°C at all pH values. FIGS. 29A to 29C show the effect of pH between 5.5 and 6.5 on the formation of basic variants in a histidine / sucrose formulation containing 20 mM histidine, 270 mM sucrose, and 0.01% polysorbate 20. FIG. 29A shows the formation of basic variants, indicating that the percentage of basic variants mostly remained between 10% and 20% after 1 month at 40°C at pH between 5.5 and 6.0. FIG. 29B shows the formation of basic variants, indicating that the percentage of basic variants generally remained between 20% and 30% after 3 months at 25°C at pH between 5.5 and 6.0. Figure 29C shows the formation of basic variants, indicating that the percentage of basic variants was generally maintained at 25% to 30% after 6 months at 5°C at all pH levels.
[0228] FIGS. 30A to 30C show the effect of a pH between 5.5 and 6.5 on the formation of basic variants in a histidine / arginine formulation containing 20 mM histidine, 150 mM arginine, and 0.01% polysorbate 20. FIG. 30A shows the formation of basic variants, indicating that the percentage of basic variants remained mostly between 10% and 20% after 1 month at 40°C at a pH between 5.5 and 6.0. FIG. 30B shows the formation of basic variants, indicating that the percentage of basic variants generally remained between 15% and 25% after 3 months at 25°C at a pH between 5.5 and 6.0. Figure 30C shows the formation of basic variants, indicating that the percentage of basic variants was generally maintained at 25% to 30% after 6 months at 5°C at all pH levels.
[0229] Both the histidine / arginine and histidine / sucrose formulations maintained acceptable stability at pH 5.5 to 6.5. Overall, the histidine / sucrose formulation was more stable than histidine / arginine at pH 5.5 to 6.0.
[0230] Based on data regarding formulation stability at 25°C for 3 months and stability at 5°C for 6 months, and data from other formulation studies of the above examples, a liquid formulation was developed comprising 20 mg / ml anti-FGFR2-IIIb antibody, 20 mM L-histidine, 270 mM sucrose, and 0.01% polysorbate 20 at pH 6.0. Additionally, a second formulation was developed comprising 20 mg / ml anti-FGFR2-IIIb antibody, 20 mM L-histidine, 150 mM L-arginine, and 0.01% polysorbate 20 at pH 5.7.
