Description

Technology Field

[0001] The present invention relates to a cosmetic composition for protecting the skin from ultraviolet rays. More specifically, the present invention relates to a cosmetic composition capable of effectively protecting the skin from damage caused by ultraviolet rays by protecting skin cells from dying due to ultraviolet exposure and by inhibiting the increase in the production of reactive oxygen species and matrix metalloproteinases caused by ultraviolet exposure.

[0002] This invention is based on research results conducted with support from the Ministry of SMEs and Startups and the Korea Institute for Industrial Technology Promotion's "Regional Specialized Industry Development+(R&D, S3089634)" project. Background Technology

[0004] The skin is the largest organ in the human body, performing a barrier function against the external environment, and consists of the epidermis, dermis, and subcutaneous tissue. External factors that threaten skin health include ozone, infrared rays, pollution, cigarette smoke, and ultraviolet rays, and 80% of facial skin aging is caused by exposure to ultraviolet rays. Among ultraviolet rays, ultraviolet A (UVA, 320–400 nm) and ultraviolet B (UVB, 290–320 nm) are involved in skin damage, and skin damage caused by UVB (photoaging) is known to be the biggest cause.

[0005] The body, damaged by UV rays, generates reactive oxygen species (ROS). ROS are known to reduce skin elasticity and accelerate the formation of wrinkles by promoting the activity of collagen-degrading enzymes, such as matrix metalloproteinases (MMPs), and inhibiting collagen synthesis.

[0006] Among the MMPs produced in the body, MMP-1 is a protease that specifically acts on collagen, and it is known that inhibiting the activity of MMP-1 can reduce the breakdown of collagen, thereby maintaining the elasticity of skin tissue and preventing the formation of wrinkles, and MMP-3 is known to activate pro-MMP-1 and specifically degrade type IV collagen of the basement membrane. Specific details for implementing the invention

[0021] The present invention relates to a cosmetic composition for protecting the skin from ultraviolet rays, comprising low molecular weight deer antler collagen, goji berry extract, and honeysuckle extract.

[0022] In the present invention, low-molecular-weight deer antler collagen refers to deer antler collagen that has been reduced to a low molecular weight, for example, through an enzymatic reaction.

[0023] The low-molecular-weight deer antler collagen of the present invention may be obtained by enzymatically reacting deer antler collagen with a protease to reduce its molecular weight, and as the protease, one or more combinations selected from the group consisting of commercial enzymes such as Acalase, Neutrase, Protamex, Novo-Pro, Formea, Flavourzyme, and Protana may be used. For the effect of the present invention, the low-molecular-weight deer antler collagen is preferably obtained by enzymatically reacting it with Neutrase and Flavourzyme to reduce its molecular weight, and preferably, it is obtained by enzymatically reacting the reaction product obtained after a primary enzymatic reaction with Neutrase with a secondary enzymatic reaction with Flavourzyme.

[0024] The first neutrase enzyme reaction can be induced by adding neutrase to deer antler collagen and isolating it at a constant temperature for a certain period, and the second flavozyme enzyme reaction can be induced by adding flavozyme to the deer antler collagen that has completed the first enzyme reaction and isolating it at a constant temperature. For the effect of the present invention, the low molecular weight deer antler collagen is preferably subjected to a first enzyme reaction by adding neutrase and isolating it at 45 to 55°C for 1 to 4 hours, and a second enzyme reaction by adding flavozyme to the first enzyme reaction product and isolating it at 45 to 55°C for 1 to 48 hours.

[0025] After the first and second enzyme reactions, each enzyme is preferably inactivated. Enzyme inactivation can be achieved, for example, by heating for a certain period at a temperature at which the enzyme may lose its activity, and preferably by heating at 88 to 92°C for 25 to 35 minutes.

[0026] For the effect of the present invention, the low molecular weight deer antler collagen is preferably subjected to a first enzymatic reaction by adding 0.0001 to 0.001 parts by weight of nutase to 100 parts by weight of deer antler collagen concentrated to 15 to 25 Brix, and to a second enzymatic reaction by adding flavozyme in an amount of 0.0001 to 0.001 parts by weight based on 100 parts by weight of the concentrated deer antler collagen to the first enzymatic reaction product.

[0027] Deer antler collagen can be obtained, for example, by a hot water extraction method from deer antlers. For the effect of the present invention, deer antler collagen is preferably obtained by adding water to deer antlers and heating at a temperature of 80 to 99°C for 1 to 10 hours, repeating this 1 to 10 times. More preferably, it is obtained by heating at a temperature of 90 to 99°C for 1 to 5 hours, repeating this 5 to 9 times, and even more preferably, by heating at a temperature of 95 to 99°C for 2 to 4 hours, repeating this 8 to 9 times.

[0028] The antler used as a raw material for antler collagen refers to the ossified antlers of deer and other animals. The antler of the present invention may be one or more antlers selected from the group consisting of, for example, red deer, Japanese deer, wapiti deer, and reindeer, but is not limited thereto. However, for the effect of the present invention, it is preferably the antler of a red deer.

[0029] In one embodiment, the low molecular weight deer antler collagen is prepared by the method of Korean Registered Patent No. 10-2323416 (the whole of which is incorporated herein by reference).

[0030] In the present invention, the goji berry extract and honeysuckle extract can be obtained using a conventional extraction method that uses plants as materials, such as goji berries (fruits of the goji berry tree) and honeysuckle (flower buds or flowers of the honeysuckle vine or a variety thereof of the Caprifoliaceae family). For example, extraction can be performed using various solvents known in the art, and as solvents, one or more combinations selected from the group consisting of water, alcohols having 1 to 4 carbon atoms, acetone, hexane, and ethyl acetate may be used. Preferably, the extract is obtained using water, ethanol, or a mixture thereof as a solvent. For the effect of the present invention, the goji berry extract and honeysuckle extract are hot water extracts. Furthermore, for the effect of the present invention, the goji berry extract and honeysuckle extract are obtained by adding 5 to 15 times the weight of water to each raw material (goji berry and honeysuckle) and repeating the extraction process by heating at a temperature of 90 to 100°C for 2 to 4 hours, one or more times. More preferably, it is obtained by extracting, concentrating under reduced pressure, and drying.

[0031] The cosmetic composition of the present invention may include low molecular weight deer antler collagen, goji berry extract, and honeysuckle extract in various proportions. However, for the effect of the present invention, it is preferably composed of 30 to 100 parts by weight of goji berry extract and 10 to 50 parts by weight of honeysuckle extract based on dry weight, with respect to 100 parts by weight of low molecular weight deer antler collagen. More preferably, it is composed of 40 to 90 parts by weight of goji berry extract and 20 to 45 parts by weight of honeysuckle extract based on dry weight, with respect to 100 parts by weight of low molecular weight deer antler collagen, with respect to 100 parts by weight of low molecular weight deer antler collagen, with respect to 100 parts by weight of goji berry extract and 25 to 40 parts by weight of honeysuckle extract based on dry weight, with respect to 100 parts by weight of low molecular weight deer antler collagen, with respect to 100 parts by weight of goji berry extract and 30 to 35 parts by weight of honeysuckle extract based on dry weight.

[0032] The amount of active ingredients in the cosmetic composition of the present invention, that is, the combined amount of low molecular weight deer antler collagen, goji berry extract, and honeysuckle extract, may vary. For example, the combined amount of low molecular weight deer antler collagen, goji berry extract, and honeysuckle extract in the composition may be 0.001 to 100 weight%, but is not limited thereto.

[0033] The cosmetic composition of the present invention may be the mixture itself of low molecular weight deer antler collagen, goji berry extract, and honeysuckle extract, which are active ingredients, and may further include cosmetically acceptable carriers or additives. In this case, the carriers and additives may also be pharmaceutically acceptable. Furthermore, the cosmetic composition of the present invention may be formulated into various forms, such as sun cream, moisturizing cream, toner, nourishing lotion, nourishing cream, massage cream, pack, and nourishing essence.

[0035] The present invention will be described in more detail below through examples. These examples are merely illustrative of the present invention, and therefore the scope of the present invention should not be interpreted as being limited by these examples.

[0037] [Example]

[0038] Example 1

[0039] 1-1. Low molecular weight deer antler collagen

[0040] Nokgak, the ossified antler of a red deer, was prepared. First, the nokgak was washed and dried, then cut into pieces with a diameter of about 1.0 cm. Then, 10 times the weight of the cut nokgak was added as purified water, and the process of heating at 95 to 99°C for about 3 hours was repeated 9 times to extract nokgak collagen.

[0041] The obtained deer antler collagen extract was filtered through a 5-micron filter, then filtered again through a 1-micron filter, and concentrated to about 20 Brix.

[0042] As a primary enzyme, Neutrase (Neutrase® of Novozyme) was added in an amount of about 0.0005 parts by weight per 100 parts by weight of deer antler collagen concentrate, and the enzyme reaction was carried out by isothermal treatment at about 50°C for about 2 hours, and then the enzyme was inactivated by heating at about 90°C for about 30 minutes.

[0043] As a secondary enzyme, Flavozyme (Flavorzyme® of Novozyme) was added to the primary enzyme-treated deer antler collagen concentrate in an amount of approximately 0.0005 parts by weight based on 100 parts by weight of the deer antler collagen concentrate (based on the weight before the primary enzyme treatment), and the enzyme reaction was carried out by isothermal treatment at approximately 50°C for approximately 2 hours, and then the enzyme was inactivated by heating at approximately 90°C for approximately 30 minutes.

[0044] Subsequently, low-molecular-weight deer antler collagen powder was prepared by vacuum concentration and vacuum drying.

[0046] 1-2. Goji berry extract

[0047] Water was added to the goji berries at a weight of approximately 10 times, and the extraction was repeated twice at approximately 95°C for about 3 hours. Specifically, after the first extraction, the extract was filtered to obtain the extract, then the same amount of water added during the first extraction was added to the extraction residue, and after the second extraction, the extract was filtered to obtain the extract, and the extract obtained from the first extraction and the extract obtained from the second extraction were combined.

[0048] The obtained extract was filtered, concentrated under reduced pressure, and vacuum dried to obtain goji berry extract powder.

[0050] 1-3. Honeysuckle Extract

[0051] Water was added to honeysuckle at a weight of approximately 10 times, and extraction was repeated twice at approximately 95°C for about 3 hours.

[0052] The obtained extract was filtered, concentrated under reduced pressure, and vacuum dried to obtain honeysuckle extract powder.

[0054] 1-4. Mixing

[0055] The above low molecular weight deer antler collagen powder, goji berry extract powder, and honeysuckle extract powder were mixed in a weight ratio of approximately 100:66.8:33.2.

[0057] Experimental Example 1

[0058] An experiment was conducted to see if the mixture of Example 1 could protect skin cells from dying due to ultraviolet irradiation.

[0060] 1-1. Experimental Method

[0061] HaCaT cells, a human keratinocyte cell line, were purchased from CLS-Cell Line Service (200493, Eppelheim, Germany). HaCaT cells were cultured in Dulbecco's Modified Eagle Medium (DMEM) containing 10% fetal bovine serum and 100 units / ml of penicillin / streptomycin at 37°C in a 5% CO2 incubator, and the medium was replaced with fresh medium every 2 to 3 days.

[0062] An MTT assay was performed to confirm the protective effect against skin cell death caused by ultraviolet (UVB) irradiation following treatment with the mixture of Example 1. HaCaT cells were placed in a 24-well plate at a rate of 5×10⁶4 Cells were seeded into wells. After 24 hours, the medium was removed and washed with PBS, then 100 µl of PBS was added and UVB (30 mJ / cm²) was irradiated. After washing with PBS, the medium was replaced with serum-free medium, and the mixture of Example 1 was added and cultured for 24 hours. After adding MTT solution (1 mg / ml) and incubating for 4 hours, formazan crystals were dissolved in DMSO, and the absorbance was measured at 540 nm using a microplate reader and expressed as a percentage.

[0064] 1-2. Experimental Results

[0065] Cell viability in HaCaT cell lines was measured using an MTT assay, and ascorbic acid (200 μM), which is well known for its skin protective effect against photodamage, was used as a positive control. The results are shown in Figure 1. When the mixture of Example 1 was treated at a concentration of 100 μg / ml in the non-UVB irradiation group, no cytotoxicity was observed (see item 'Green:Gold:Gold' in the graph). Subsequently, when HaCaT cells were irradiated with 30 mJ / cm² of UVB, cell viability decreased to approximately 57% (see item 'UVB' in the graph), but when treated with the mixture of Example 1, cell viability increased to approximately 85% (see item 'UVB+Green:Gold:Gold' in the graph), showing a cytoprotective effect similar to that of the positive control, ascorbic acid (200 μM) (see item 'UVB+Ascorbic acid' in the graph). Through these results, it was confirmed that the composition of the present invention strongly inhibits UVB-induced skin cell toxicity.

[0066] In addition, the results of experiments conducted by treating the mixture of Example 1 at various concentrations are shown in Figure 2. It was found that cell viability increased as the treatment concentration of the mixture of Example 1 increased, and these results demonstrate that the protective effect of the composition of the present invention against UVB on skin cells is concentration-dependent.

[0068] Experimental Example 2

[0069] An experiment was conducted to see if the mixture of Example 1 above could suppress the increase in the production of reactive oxygen species (ROS) upon UV irradiation.

[0071] 2-1. Experimental Method

[0072] HaCaT cells were pretreated with the mixture of Example 1 for 24 hours, washed with PBS, and then irradiated with UVB (30 mJ / cm²). After UVB irradiation, the cells were treated with the mixture of Example 1 for 30 minutes and then stained with 25 μM dichlorodihydro-fluorescein diacetate (DCFH-DA). The fluorescence intensity corresponding to intracellular ROS production was measured using a fluorescent spectrophotometer at an excitation wavelength of 485 nm and an emission wavelength of 530 nm.

[0074] 2-2. Experimental Results

[0075] To investigate the inhibitory activity of the mixture of Example 1 against ROS generated by ultraviolet rays within cells, HaCaT cells were irradiated with UVB, and the amount of ROS was measured. As a result, as shown in Figure 3, the UVB irradiation group (refer to the 'UVB' item in the graph) showed a rapid increase in ROS production compared to the control group (refer to the 'Control' item in the graph), whereas the mixture of Example 1 (refer to the 'Green:Gold:Gold' item in the graph) reduced the amount of ROS induced by UVB irradiation in a concentration-dependent manner at concentrations of 25, 50, and 100 μg / ml. Through these results, it was confirmed that the composition of the present invention exhibits a skin-protective effect against UVB stimulation by reducing ROS increased by UVB.

[0077] Experimental Example 3

[0078] An experiment was conducted to see if the mixture of Example 1 above could inhibit the increase in the production of matrix metalloproteinase (MMP) upon UV irradiation.

[0080] 3-1. Experimental Method

[0081] HaCaT cells were pretreated with the mixture of Example 1 for 24 hours, washed with PBS, and then irradiated with UVB (30 mJ / cm²). After UVB irradiation, the production of MMP-1 and MMP-3 in the medium was measured using a human MMP-1, MMP-3 ELISA kit.

[0083] 3-2. Experimental Results

[0084] To confirm the effect of the mixture of Example 1 on MMPs increased after UV irradiation, the production amounts of MMP-1 and MMP-3 were measured. As a result, as shown in Figures 4 and 5, the secretion of MMP-1 and MMP-3 increased in the UVB irradiation group (refer to 'UVB' in the graph) compared to the control group (refer to 'Control' in the graph), but the mixture of Example 1 (refer to 'Green:Gold:Copper' in the graph) significantly reduced the production of UVB-induced MMP-1 and MMP-3, and the inhibitory effect on the production of MMPs was greatest at a concentration of 100 μg / ml. Through these results, it was confirmed that the composition of the present invention has the effect of preventing or alleviating skin wrinkles caused by ultraviolet rays.
