Abstract
Description
Technology Field
[0001] The present invention provides a novel triazine derivative having therapeutic effects on inflammation and fibrosis of the liver and kidneys, and a method for preparing the same. Background Technology
[0002] Chronic liver or kidney disease occurs in response to persistent, long-term liver or kidney damage caused by viral hepatitis, obesity, or alcohol abuse. While liver or kidney disease can vary, a common pathological feature is inflammation or liver fibrosis. Fibrosis of the liver or kidney, in which extracellular matrix (ECM) fills areas that are not regenerated by liver and kidney cells, leads to an excessive accumulation of inflammatory mediators due to chronic and / or severe liver and kidney damage. This disrupts the normal functioning of the liver and kidneys, eventually leading to terminal stages of liver or kidney cirrhosis or liver or kidney cancer, creating a need for treatments.
[0003] Meanwhile, PAR2 (Protease-activated receptor 2) is a family of G-protein coupled receptors (GPCRs) that is irreversibly activated through the cleavage of its N-terminus. The transmembrane receptor domain triggers cellular signaling cascades in response to inflammatory signals and other stimuli. Once the original N-terminal domain is removed, the tether ligand activates intracellular G protein or β-arrestin-mediated signaling pathways to induce a wide range of intracellular physiological responses.
[0004] These PAR2s are central mediators of diseases such as cancer, inflammation, asthma, pancreatitis, and irritable bowel syndrome. In addition, PAR2 antagonists are expected to act as therapeutic agents by blocking the inflammatory and fibrotic pathways of the liver and kidneys, and research on this is currently ongoing. [0128] [Experimental Example 3] Cell Culture
[0129] YPEN-1 endothelial cells, a rat endothelial cell line, were purchased as ATCC (CRL-2222) for the experiment. DMEM nutrient medium containing antibiotics (100 units / mL penicillin, 100 μg / mL streptomycin) and 5% FBS (fetal bovine serum) was purchased from Welgene (Daegu) and used for the experiment in a 37 ℃, 5% CO2 incubator.
[0131] [Experimental Example 4] Confirmation of Cell Survival of PAR2 Antagonists in Mouse Endothelial Cells
[0132] To confirm the toxicity of the triazine derivatives prepared according to the above synthesis example in YPEN-1 mouse endothelial cells, 1×10⁶ were placed in a 96-well culture vessel. 4 After dispensing 100 μL of cells at a cells / well concentration, the cells were treated for 24 hours with each concentration of the derivatives or the positive control (GB83) at concentrations of 0, 2, 5, 10, and 20 μM. Subsequently, 10 μL of EZ-cytox solution (Daeillab, Seoul) was added and incubated for 2 to 3 hours. The inhibitory concentration of cell viability was determined by measuring the absorbance at a wavelength of 450 nm using a microplate reader.
[0134] [Experimental Example 5] Experimental Animals
[0135] (1) LPS-induced inflammation mouse model
[0136] In vivo To confirm the anti-inflammatory activity of Cpd. 1, Cpd. 1 was orally administered to C57BL / 6J mice at a dose of 1 or 5 mg / kg once daily for 3 days. On the 3rd day, 3 hours after oral administration of the drug, the endotoxin LPS (lipopolysaccharide) was administered intraperitoneally, and the mice were euthanized 5 hours later. Organs and blood were collected and analyzed.

After the diet was finished, the mice were euthanized, and organs and blood were collected and analyzed.
[0144] [Experimental Example 6] RNA isolation, cDNA synthesis, and qRT-PCR (quantitative real-time polymerase chain reaction) analysis
[0145] Total RNA was isolated from mouse tissues and cells treated with Cpd. 1 using Ribo-Ex (GeneAll Biotechnology, Seoul, Korea), and cDNA was synthesized using the Suprime Script RT Premix with random hexamer cDNA Synthesis Kit (GeNet Bio, Daejeon, Korea). To relatively quantify the expression of inflammatory cytokine and chemokine genes induced by Cpd. 1, SensiFAST TM Real-time PCR (CFX-96, Bio-rad, Hercules, CA, USA) analysis was performed using SYBR No-ROX dye (Bioline, London, UK) and gene-specific primers. The expression levels of each gene were corrected for the relative expression levels of GAPDH (glyceraldehyde-3-phosphate dehydrogenase) and 2 - ΔΔIt was calculated using the CT method.

[0148] Mouse tissues or cells treated with Cpd. 1 were lysed with 30 μL of NP40 buffer (50 mM Tris-HCl pH 8.0, 1% NP40, Triton X-100, 150 mM NaCl, Protease inhibitor cocktail) for 20 minutes, and protein concentrations were measured using a protein assay (BCA protein assay, Pierce, Rockford, IL, USA). Each tissue or cell protein was subjected to 8 to 10% SDS-PAGE (polyacrylamide gel electrophoresis), and the proteins present on the gel were semi-dry transferred to a PVDF membrane (Bio-Rad Laboratories, Hercules, CA, USA) at 25 V for 10 minutes. The membrane was blocked with a 5% skim milk solution at room temperature for 1 hour to prevent non-specific reactions. After that, the primary antibody was reacted overnight at 4°C, and the secondary antibody conjugated with HRP was reacted for 1 hour. Next, color development was performed using the ECL reagent SuperSignal West Pico Chemiluminescent Substrate kit (Advansta, San Jose, CA, USA), and each labeled protein was identified using the Davinch-Chemi imager (Davinch-K, Seoul, Korea).
[0150] [Example 1] Confirmation of anti-inflammatory effects of triazine derivative compounds in vascular endothelial cells
[0151] vascular endothelial cells in vitroFor the study, the toxicity of the triazine derivative compounds synthesized by the above synthesis example was measured. As a result, according to Figure 3, it was confirmed that the triazine derivative compounds did not exhibit cytotoxicity even at the maximum concentration tested, which is 20 μM.
[0152] To confirm the anti-inflammatory activity of triazine derivative compounds, the samples were pretreated with 5 μM of the triazine derivative compound and a previously known PAR2 antagonist (GB83) for 1 hour, followed by treatment with 1 μg / mL of LPS for 3 hours to induce inflammation. Subsequently, the expression of the inflammation-related transcription factor NF-κB and the target pro-inflammatory genes COX-2 and p-p65 was confirmed via Western blot. As a result, according to Figure 4, it was confirmed that the LPS-induced NF-κB phosphorylation and the expression of COX-2 and p-p65 were reduced more by the triazine derivative compound than by GB8.
[0153] To confirm the inhibitory effect of triazine derivative compounds on inflammatory cytokine expression induced by PAR2 activation in vascular endothelial cells (YPEN-1 cells), inflammation was induced with a PAR2 activator after pretreatment with each compound. As a result, according to Figure 5, the expression of inflammatory cytokines was reduced in the group pretreated with triazine derivative compounds.
[0154] Using bioinformatics software to evaluate the pharmacological efficacy of triazine derivative compounds in silicoAnalysis was performed. Table 1 shows the results of predicting the binding strength of PAR2 proteins and derivative compounds using three docking simulation programs (Autodock Vina, Autodock 4, and Dock 6). It was found that most of the derivative compounds bind stably to PAR2, as most of these novel synthetic substances had binding energies similar to the control group when compared to GB83, GB88, and AZ3451, which are known as PAR2 antagonists.

[0157] [Example 2] Anti-inflammatory effect of a triazine derivative compound
[0158] LPS was administered to confirm the anti-inflammatory effect of Cpd. 1 in vivo The anti-inflammatory activity of Cpd. 1 was confirmed in an inflammation model. As a result, according to Figure 6, in the kidneys of the group administered Cpd. 1, a concentration-dependent decrease in protein expression of NF-kB phosphorylation (Figure 5A) and mRNA levels of inflammatory cytokines and chemokines (IL-1β, TNF-α, IL-6, and MCP-1) (Figure 5B) were observed.
[0159] Furthermore, changes in Cpd. 1 inflammatory proteins in vascular endothelial cells were confirmed via Western blot. As shown in Figure 7A, while LPS-induced protein expression of phosphorylated p65 in the cell nucleus and the target pro-inflammatory genes COX-2 and iNOS in the cytoplasm increased, treatment with Cpd. 1 resulted in a concentration-dependent decrease in the expression of phosphorylated p65, COX-2, and iNOS. Additionally, to determine whether Cpd. 1 treatment regulates the expression of LPS-induced NF-κB inflammatory cytokines and chemokines, Western blot and quantitative PCR techniques were used. As a result, according to Figures 7B and 7C, the expression of inflammatory proteins and cytokine and chemokine mRNAs (IL-6, TNF-α, and MCP-1) was reduced in the group treated with Cpd. 1; therefore, Cpd. It was found that 1 is effective in suppressing inflammation.
[0161] [Example 3] Inhibitory effect of triazine derivative compounds on vascular aging
[0162] To confirm the inhibition of vascular aging by Cpd. 1, adhesion molecules, which are markers of vascular aging, were examined at protein and mRNA levels in an LPS-induced inflammatory mouse model and LPS-treated vascular endothelial cells. As a result, according to Figure 8, it was confirmed that the expression of VCAM and ICAM, which was increased by LPS treatment, decreased in a concentration-dependent manner upon administration of Cpd. 1. From these results, it was found that Cpd. 1 is effective in inhibiting vascular aging.
[0164] [Example 4] Regulatory effect of triazine derivative compounds on metabolic diseases
[0165] To confirm the metabolic disease-improving effects of PAR2 inhibitors, the study was conducted in a diet-induced obesity mouse model and an obesity / diabetes db / db mouse model. Figures 9A and 9B show the changes in body weight and the results of the oral glucose tolerance test (GTT) measured during the experiment in the diet-induced obesity mouse model; Cpd. 1 was confirmed to inhibit diet-induced weight gain (Figure 9A) and to increase glucose tolerance (Figure 9B). Figure 9C shows the results of measuring blood insulin, glucose, triglycerides, and total cholesterol; it was confirmed that the levels of insulin, triglycerides, and total cholesterol decreased (Figure 9C). Furthermore, according to Figures 9D and 9E, a weight reduction effect induced by Cpd. 1 was confirmed in the obesity / diabetes db / db mouse model as well, and Cpd. It was confirmed that glucose tolerance increased (Fig. 9E), and Cpd. 1 reduced blood total cholesterol, insulin, and triglyceride levels (Fig. 9F). From the above results, it was determined that Cpd. 1 contributes to the improvement of metabolic syndrome.
[0166] In the metabolic disease diet-induced obesity mouse model and the db / db mouse model, Cpd.1 was administered at a daily dose of 2 or 6 mg / kg during the experimental period of the example, while the control drug empagliflozin was administered at a dose of 10 mg / kg and metformin at a dose of 200 mg / kg; therefore, it was determined that Cpd.1 contributed to the improvement of metabolic syndrome more effectively than the control drugs used in the experiment.
[0167] To investigate the molecular mechanism of the body weight-loss effect of Cpd. 1 in a metabolic disease (obesity / diabetes) db / db mouse model, protein and mRNA expression were examined in liver tissue. As a result, according to Figure 10, Cpd. 1 increased the expression of IRS tyrosine 632 phosphate groups and AKT phosphorylation in a concentration-dependent manner, while decreasing the expression of IRS serine 307 phosphate groups. Additionally, the expression of ACC and FAS, genes related to lipid synthesis, was reduced by the administration of Cpd. 1. Furthermore, in kidney tissue, it was confirmed that the expression of p65 phosphate groups, an inflammatory protein, in the cell nucleus was reduced by the administration of Cpd. 1. Moreover, by confirming that the mRNA expression of MCP-1, E-selectin, and VCAM, markers of vascular aging, was reduced by Cpd. 1, it was found that Cpd. 1 is effective in inhibiting inflammation and vascular aging, as well as improving metabolic syndrome.
[0169] [Example 5] Fibrosis-regulating effect of triazine derivative compounds
[0170] To confirm the fibrosis-inhibiting effect of Cpd. 1, mRNA levels of fibrosis markers (CCL5, VIM, α-SMA, TGF1β, Col3a1, and Col4a1) were examined in the liver (Fig. 11A) and kidney tissues (Fig. 11B) of a metabolic disease db / db mouse model. As a result, according to Figs. 11A and 11B, Cpd. 1 reduced the expression of these fibrosis markers in liver and kidney tissues; thus, it was found that Cpd. 1 is effective in improving liver and kidney fibrosis caused by obesity and diabetes.
[0171] In addition, mRNA levels of fibrosis markers (α-SMA, TGF1β, Col1a2, Col3a1) were examined in liver tissue (Fig. 11C) of a diet-induced obesity mouse model of metabolic disease with Cpd. 1. As a result, according to Fig. 11C, Cpd. 1 reduced the expression of these fibrosis markers in the liver tissue of the diet-induced obesity mouse model, indicating that Cpd. 1 is effective in improving liver fibrosis caused by metabolic diabetes.
[0173] [Example 6] Triazine derivative compound in silico Analysis results
[0174] Based on the docking simulation results, the pharmacophore predicting the binding between PAR2 and Cpd. 1 was analyzed. According to Figures 12A and 12B, it was confirmed that Cpd. 1 forms a total of three hydrogen bonds with the Val154 (one hydrogen bond) and Asp153 (two hydrogen bonds) residues of the PAR2 protein, and that GB83 binds to one hydrogen bond residue (Ser245) of PAR2.
[0175] Molecular Dynamics (MD) were analyzed using the complex structure of PAR2 and Cpd. 1. Through MD analysis, the Root Mean Square Deviation (RMSD), Root Mean Square Fluctuation (RMSF), and the number of hydrogen bonds were determined. As a result, according to Figures 12C, 12D, and 12E, although Cpd. 1 had a higher RMSD value than GB83, it maintained that value stably (Figure 12D), indicating that Cpd. 1 maintains a relatively stable state when complexed with PAR2. In the case of RMSF as well, the variability of GB83 and Cpd. 1 at each amino acid residue of PAR2 was generally similar (Figure 12C). This indicated that Cpd. 1 can perform a function similar to GB83 when bound to PAR2. Furthermore, Cpd. compared to GB83 As it was shown that the number of hydrogen bonds of 1 was greater over most of the running time (Fig. 12E), this was consistent with the pharmacophore analysis. From the above results, it was found that Cpd. 1 binds more strongly to PAR2 than GB83.
[0176] To determine the drug suitability of Cpd. 1, ADMET was analyzed using the ADMETlab 2.0 website (https: / / admetmesh.scbdd.com / ). As a result, according to Table 2, Cpd. 1 possesses physicochemical properties similar to GB83, passed the drug-likeness criteria of the Lipinski Rule and Pfizer Rule, and showed values equal to or slightly better than GB83 in terms of toxicity. In conclusion, in silicoWhen synthesizing the analysis, it was found that Cpd. 1 is an inhibitor that binds to and acts on PAR2, and compared to existing drugs, it has the potential to have better efficacy and safety.
