[0012] The term “wild ginseng cultured root” (or “wild ginseng adventitious root”) as used herein may refer to a cultured root obtained by inducing adventitious roots from the tissues or cells of wild ginseng and proliferating them through aseptic tissue culture or bioreactor culture. The said cultured wild ginseng root may contain components derived from wild ginseng, such as saponins containing ginsenosides and polysaccharides. In this invention, the said cultured wild ginseng root may include not only raw cultured roots but also forms that have been washed, dried, freeze-dried, cut, crushed, or ground for use in a subsequent extraction process. Furthermore, the “wild ginseng extract” obtained from the said cultured wild ginseng root may refer to a ginsenoside-containing extract or fraction obtained by extracting the said cultured wild ginseng root with aqueous ethanol and undergoing one or more processes among concentration, adsorption resin purification, fractionation, and drying.
[0013] As used herein, the term “ginsenoside” may refer to saponin compounds that may be contained in cultured wild ginseng roots, and may include, for example, one or more of Rb1, Rb2, Rc, Rd, Rg1, and Re, but is not limited thereto. In this invention, “ginsenoside-containing fraction” may refer to an extract or fraction containing one or more of the above ginsenosides.
[0014] As used herein, the term “mushroom raw material” may refer to a mixed mushroom raw material comprising Sparassis crispa, Phellinus linteus, and truffles. The mushroom raw material may include polysaccharides containing beta-glucan, proteins, amino acids, and other mushroom-derived components. In this invention, the Sparassis crispa, Phellinus linteus, and truffles may be in a dried, freeze-dried, cut, crushed, or ground form, respectively, for use in subsequent hydration, enzymatic treatment, and pressurized hot water extraction. Additionally, the “mushroom extract” obtained from the mushroom raw material may refer to a beta-glucan-containing extract or fraction obtained by hydrating and enzymatically treating the mushroom raw material, followed by pressurized hot water extraction, and undergoing one or more processes among ultrafiltration, dialysis filtration, and concentration.
[0015] As used herein, the term “aqueous ethanol” may refer to a mixed solvent containing ethanol and water. Unless specifically stated otherwise, the ethanol concentration of aqueous ethanol may refer to the volume percentage of ethanol based on the volume of the total liquid mixture.
[0017] In one embodiment of the present invention, the step (a-1) may be a step of pre-hydrating the crushed cultured wild ginseng root under reduced pressure conditions, adjusting the ethanol concentration, and then performing ultrasonic treatment and primary extraction to obtain a primary extract and a residue.
[0018] In one embodiment of the present invention, the cultured wild ginseng root may be washed and dried, and then ground. The drying may be performed under conditions that secure a moisture content suitable for subsequent grinding and extraction while suppressing thermal changes of ginsenosides. Specifically, the cultured wild ginseng root may be dried at 45°C and under reduced pressure conditions of 10 kPa or less absolute pressure so that the moisture content is 8% by weight or less, and the dried cultured wild ginseng root may be ground to an average particle size of about 300 μm.
[0019] In one embodiment of the present invention, the preliminary hydration may be performed by adding purified water to the crushed cultured wild ginseng root and then repeating the reduction of pressure and return to atmospheric pressure. By repeating the reduction of pressure and return to atmospheric pressure, air inside the crushed cultured wild ginseng root is removed and the penetration of purified water is promoted, thereby allowing the extraction solvent to penetrate uniformly into the raw material during subsequent hydration ethanol extraction.
[0020] Specifically, pre-hydration can be performed by adding 500 parts by weight of purified water to 100 parts by weight of the above-mentioned crushed cultured wild ginseng root, reducing the pressure to 20 kPa at 30°C for 5 minutes, and then maintaining it at atmospheric pressure for 10 minutes, repeating this process 3 times.
[0021] In one embodiment of the present invention, ethanol may be added to the pre-hydrated cultured wild ginseng root powder to adjust the final ethanol concentration to 45 volume% to 65 volume%, specifically 55 volume%. The ethanol concentration range may be suitable for extracting ginsenosides contained in the cultured wild ginseng root.
[0022] In one embodiment of the present invention, the ultrasonic treatment may be performed by repeating the process of irradiating with ultrasound of 30 kHz to 50 kHz at 40°C to 60°C for 5 minutes and then stopping for 5 minutes six times. Specifically, the ultrasonic treatment may be performed at 50°C and 40 kHz.
[0023] The above intermittent ultrasonic treatment conditions are set to mitigate local heat accumulation caused by continuous irradiation while securing the extraction-assisting effect of ultrasonic irradiation, and may be advantageous for securing a balanced ginsenoside content and recovery rate compared to continuous ultrasonic irradiation for the same actual irradiation time, and for enhancing the growth plate cartilage cell and long bone growth-promoting effects of the final composition.
[0024] In one embodiment of the present invention, the ultrasonically treated mixture may be stirred and extracted at 50°C and 250 rpm for 3 hours, and then separated into a primary extract and a residue by centrifugation or filtration. The residue may contain unextracted cultured wild ginseng root tissue and remaining extractable components, and may be used for a subsequent secondary extraction.
[0026] In one embodiment of the present invention, the step (a-2) may be a step of secondarily extracting the residue with aqueous ethanol, mixing the obtained second extract with the first extract to prepare a crude extract of wild ginseng, and then passing it through a nonionic porous adsorption resin to obtain a ginsenoside-containing fraction.
[0027] In one embodiment of the present invention, the secondary extraction may be performed by adding 55 volume% aqueous ethanol to the residue and stirring at 50°C and 250 rpm for 2 hours. The secondary extraction may be advantageous for further recovering ginsenosides remaining in the residue after the primary extraction.
[0028] In one embodiment of the present invention, the crude wild ginseng extract obtained by mixing the first extract and the second extract may be filtered and concentrated under reduced pressure. Specifically, the crude wild ginseng extract may be sequentially filtered through a 100 μm filter mesh and a 0.45 μm microfiltration membrane, and then concentrated under reduced pressure at 40°C or lower to control the residual ethanol concentration to 1 volume% or less. Subsequently, purified water may be added to adjust the solid content concentration to about 5 weight%.
[0029] In one embodiment of the present invention, the wild ginseng concentrate may pass through a nonionic porous adsorption resin. The adsorption resin may be a styrene-divinylbenzene-based macroporous adsorption resin, and the wild ginseng concentrate may pass through at a flow rate of 1 to 2 times the resin volume per hour.
[0030] In one embodiment of the present invention, the styrene-divinylbenzene-based macroporous adsorption resin has a specific surface area of 450 m² 2 / g to 550 m 2 / g, and the average pore diameter may be 8 nm to 12 nm.
[0031] In one embodiment of the present invention, the adsorption resin may be sequentially washed and equilibrated with 95 volume% ethanol and purified water before passing the wild ginseng concentrate through it. The washing and equilibration may remove impurities that may remain on the adsorption resin and ensure that the adsorption resin is in a state suitable for the water system conditions of the wild ginseng concentrate.
[0032] In one embodiment of the present invention, the adsorption resin may be sequentially washed with purified water and 10 to 30 volume% of aqueous ethanol, and then eluted with 60 to 80 volume% of aqueous ethanol. Specifically, after washing with 3 volumes of purified water and 2 volumes of resin in 20 volume% aqueous ethanol, a ginsenoside-containing fraction may be eluted with 3 volumes of resin in 70 volume% aqueous ethanol.
[0033] In one embodiment of the present invention, purification using the nonionic porous adsorption resin may be configured to adsorb ginsenosides contained in the crude wild ginseng extract, then remove water-soluble non-target components in stages, and elute the ginsenoside-containing fraction. If the purification using the adsorption resin is not performed, the non-target components contained in the crude wild ginseng extract are not sufficiently removed, which may result in a decrease in the ginsenoside content and recovery rate, as well as a reduction in the growth plate cartilage cell and long bone growth-promoting effects of the final composition.
[0035] In one embodiment of the present invention, step (a-3) may be a step of concentrating the ginsenoside-containing fraction and freeze-drying it to obtain a wild ginseng extract containing ginsenoside.
[0036] In one embodiment of the present invention, the ginsenoside-containing fraction may be concentrated under reduced pressure at 40°C or lower to remove ethanol. Such low-temperature, reduced-pressure concentration may be advantageous for removing the solvent while suppressing thermal changes in the ginsenoside.
[0037] In one embodiment of the present invention, the concentrate may be frozen at -40°C or lower and then freeze-dried. Through the freeze-drying, a wild ginseng extract containing ginsenosides can be obtained in powder form and prepared in a state suitable for subsequent mixing with mushroom extract based on dry solid content.
[0038] The term “based on dry solid content” as used herein may mean calculating the content or weight ratio of each extract based on the weight of non-volatile solids, excluding water and volatile solvents contained in the wild ginseng extract or mushroom extract. The dry solid content may be identified by the loss on drying method, weight measurement after freeze-drying, or an equivalent method.
[0040] In one embodiment of the present invention, the step (b-1) may be a step of hydrating a mushroom raw material including Sparassis crispa, Phellinus linteus, and truffles, and sequentially enzymatically treating it with hemicellulase and neutral protease.
[0041] In one embodiment of the present invention, the cauliflower mushroom, Phellinus linteus mushroom, and truffle mushroom may be mixed after being dried and ground, respectively. Specifically, each mushroom may be dried at 50°C to a moisture content of 10% by weight or less, and then ground to an average particle size of about 500 μm.
[0042] In one embodiment of the present invention, the mushroom raw material may comprise 60% by weight of Sparassis crispa, 25% by weight of Phellinus linteus, and 15% by weight of Truffle, based on 100% by weight of the mushroom raw material. The weight ratio is adjusted so that the content and recovery rate of beta-glucan and the effects of promoting proliferation of growth plate chondrocytes, cartilage matrix formation, and longitudinal growth of long bones are balanced. If the weight ratio of the mushroom raw material deviates from the above composition, the compositional balance between the components derived from Sparassis crispa, Phellinus linteus, and Truffle may change, which may result in a decrease in the content and recovery rate of beta-glucan and a reduction in the effects of promoting proliferation of growth plate chondrocytes, cartilage matrix formation, and longitudinal growth of long bones in the final composition.
[0043] In one embodiment of the present invention, the mushroom raw material may be dried and ground, and then hydrated in stages. Specifically, purified water may be added to the mushroom raw material and hydrated in stages at 35°C for 1 hour, followed by hydration in stages at 55°C for 1 hour. The staged hydration may be advantageous for promoting the penetration of purified water into the mushroom raw material to form a state suitable for subsequent enzyme treatment.
[0044] In one embodiment of the present invention, the hydrated mushroom raw material may be first enzymatically treated with hemicellulase at a pH of 4.8 to 5.6, and then secondarily enzymatically treated with neutral protease at a pH of 6.0 to 7.0. Specifically, the first enzymatic treatment may be performed for 90 minutes at a pH of 5.2 and 50°C, and the second enzymatic treatment may be performed for 60 minutes at a pH of 6.5 and 50°C.
[0045] In one embodiment of the present invention, when the hemicellulase and neutral protease are treated simultaneously under the same pH conditions, reaction conditions suitable for each enzyme are not individually secured, so the pretreatment effect of the mushroom raw material by sequential enzyme treatment may not be sufficiently manifested, and accordingly, the content and recovery rate of beta-glucan may be reduced.
[0046] In one embodiment of the present invention, if neutral protease treatment is not performed after hemicellulase treatment, the enzymatic treatment effect on the mushroom raw material is not sufficiently secured, so the elution of components including beta-glucan during subsequent pressurized hot water extraction may be limited, and the growth plate cartilage cell and long bone growth-promoting effects of the final composition may be reduced.
[0047] In one embodiment of the present invention, if only neutral protease is treated without performing the hemicellulase treatment, the enzymatic pretreatment of the mushroom raw material is not sufficiently performed, so the content and recovery rate of beta-glucan may be reduced in the subsequent pressurized hot water extraction, and the growth plate cartilage cell and long bone growth-promoting effects of the final composition may be reduced.
[0048] In one embodiment of the present invention, the mushroom raw material after the secondary enzyme treatment is completed may be heated prior to subsequent pressurized hot water extraction to inactivate the hemicellulase and neutral protease. Specifically, the enzyme-treated mushroom raw material may be heated at 90°C for 10 minutes. The enzyme inactivation inhibits the continuation of the enzyme reaction during the subsequent process, thereby maintaining constant extraction conditions of the mushroom raw material.
[0050] In one embodiment of the present invention, the step (b-2) may be a step of obtaining an extract by pressurizing hot water extraction of the secondary enzyme-treated mushroom raw material and obtaining a membrane-containing liquid concentrated with beta-glucan by ultrafiltration of the extract.
[0051] The term “retentate” as used herein may refer to a liquid fraction that is retained on the supply side or circulation system of the membrane without passing through the ultrafiltration membrane during the ultrafiltration process, and “permeate” may refer to a liquid fraction that passes through the ultrafiltration membrane and is discharged to the permeate side of the membrane.
[0052] In one embodiment of the present invention, the pressurized hot water extraction may be performed under conditions of 105°C to 125°C and an absolute pressure of 0.15 MPa to 0.3 MPa, specifically under conditions of 115°C and an absolute pressure of 0.18 MPa. The pressurized hot water extraction may be set to promote the elution of components including beta-glucan from the tissue of the mushroom raw material relaxed by enzyme treatment. When hot water extraction is performed at atmospheric pressure, the tissue of the mushroom raw material is destroyed and the elution of components is insufficient, which may result in a decrease in the content and recovery rate of beta-glucan and the biological effect of the final composition.
[0053] In one embodiment of the present invention, the extract obtained after pressurized hot water extraction may be centrifuged and filtered to remove solids and then supplied to an ultrafiltration membrane. Specifically, the extract may be sequentially filtered through a 100 μm filter mesh and a 0.45 μm microfiltration membrane.
[0054] In one embodiment of the present invention, the fractional molecular weight of the ultrafiltration membrane may be 40 kDa to 60 kDa, and most preferably 50 kDa. The fractional molecular weight range may be set to separate a relatively low molecular weight component into a membrane permeate while maintaining a high molecular weight fraction containing beta-glucan in the membrane retaining liquid.
[0055] The term “molecular weight cut-off (MWCO)” as used herein may refer to a nominal molecular weight standard indicating the separation characteristics of an ultrafiltration membrane, and may be used to indicate the membrane permeation or inhibition characteristics of a solute having a specific molecular weight. The said molecular weight can be determined according to specifications provided by the membrane manufacturer or equivalent conventional membrane characteristic evaluation methods.
[0056] If the fractional molecular weight of the ultrafiltration membrane is less than 40 kDa, the range of components remaining together with beta-glucan widens, and the relative content of beta-glucan in the membrane retaining liquid may decrease. On the other hand, if the fractional molecular weight of the ultrafiltration membrane exceeds 60 kDa, some of the fraction containing beta-glucan may move into the membrane permeate, and the recovery rate of beta-glucan may decrease.
[0057] In one embodiment of the present invention, the ultrafiltration can be performed in a cross-flow manner. Specifically, a polyethersulfone ultrafiltration membrane can be used to perform circulating filtration at 30°C and a transmembrane pressure of 0.25 MPa, and the membrane retaining liquid can be concentrated until the volume of the membrane retaining liquid becomes one-quarter of the volume of the initial feed liquid.
[0059] In one embodiment of the present invention, step (b-3) may be a step of dialysis-filtering and concentrating the membrane holding liquid to obtain a mushroom extract containing beta-glucan.
[0060] In one embodiment of the present invention, the dialysis filtration may be performed by adding an equal volume of purified water to the membrane holding liquid and then repeating the process of ultrafiltration three times until a volume equivalent to the added purified water is removed as membrane permeate.
[0061] In one embodiment of the present invention, the membrane retained after the dialysis filtration is completed may be filtered after being adjusted to a pH of 6.0 to 7.0. Specifically, it may be filtered sequentially through a 1.0 μm filter membrane and a 0.45 μm filter membrane.
[0062] In one embodiment of the present invention, the filtrate may be concentrated under reduced pressure at 40°C or lower to control the solid content concentration, specifically, the solid content concentration may be 12% by weight. The low-temperature reduced-pressure concentration can provide a mushroom extract with a concentration suitable for subsequent mixing with wild ginseng extract based on dry solid content while suppressing thermal changes in components including beta-glucan.
[0064] In one embodiment of the present invention, step (c) may be a step of homogenizing the wild ginseng extract and the mushroom extract in a weight ratio of 1:4 to 1:6 based on dry solid content, and then aging and freeze-drying to obtain a composition for improving height growth.
[0065] Specifically, the wild ginseng extract and mushroom extract may be mixed in a weight ratio of 1:5 based on dry solid content. The weight ratio is set to ensure a balanced content of ginsenosides and beta-glucans; if the proportion of wild ginseng extract is excessively high, the content of beta-glucan may be relatively reduced, and if the proportion of wild ginseng extract is excessively low, the content of ginsenosides may be relatively reduced. Consequently, the effects of promoting the proliferation of growth plate chondrocytes, the formation of cartilage matrix, and the longitudinal growth of long bones may be reduced.
[0066] In one embodiment of the present invention, the mixing may be performed by maintaining the mushroom extract at 35°C while adding the wild ginseng extract in portions and stirring. Specifically, the wild ginseng extract may be added in five portions, and after each addition, stirring may be performed to mix at 700 rpm for a total of 60 minutes.
[0067] In one embodiment of the present invention, the mixture may be high-pressure homogenized three times at a pressure of 50 MPa, aged at 8°C for 12 hours, and then frozen and freeze-dried at -40°C or lower. The high-pressure homogenization and aging may allow the wild ginseng extract and mushroom extract to be supplied to a subsequent freeze-drying process in a uniformly mixed state.
[0069] In one embodiment of the present invention, the manufacturing method may be configured to ensure a balanced content and recovery rate of ginsenosides and beta-glucans by combining a wild ginseng extraction process comprising vacuum pre-hydration of cultured wild ginseng roots, intermittent ultrasonic extraction, and adsorption resin purification, and a mushroom extraction process comprising stepwise hydration, sequential enzymatic treatment, pressurized hot water extraction, and ultrafiltration of mushroom raw materials. When only conventional solvent extraction and atmospheric pressure hot water extraction are applied, the penetration of the extraction solvent into the raw materials, enzymatic pretreatment of the mushroom raw materials, removal of non-target components, and fractionation of active ingredients are not sufficiently achieved, which may result in a decrease in the content and recovery rate of ginsenosides and beta-glucans; consequently, the effects of the final composition on growth plate chondrocyte proliferation, cartilage matrix formation, and longitudinal growth promotion of long bones may be reduced.
[0071] In one embodiment of the present invention, the manufacturing method may further add and mix 0.5 parts by weight of zinc citrate with respect to 100 parts by weight of the total dry solid content of the wild ginseng extract and mushroom extract in step (c).
[0072] In one embodiment of the present invention, the zinc citrate may be added as a zinc source together with the wild ginseng extract and mushroom extract to promote the proliferation of growth plate chondrocytes and the production of cartilage matrix, and to complement the effect of promoting longitudinal growth of long bones. Specifically, a composition further comprising the above amount of zinc citrate can increase the production of type II collagen and agrican and enhance the effect of increasing long bone length.
[0074] The second aspect of the present invention provides a composition for improving height growth prepared by a manufacturing method according to the first aspect of the present invention. The descriptions regarding the wild ginseng extract and mushroom extract included in the composition for improving height growth according to the second aspect, the zinc citrate optionally included, the content and weight ratio of each component, and the manufacturing conditions of each extract as well as the mixing, homogenization, aging, and freeze-drying conditions may be applied in the same manner as the descriptions in the first aspect of the present invention, provided that they are not contradictory.
[0075] In one embodiment of the present invention, the composition for improving height growth can promote the proliferation of growth plate chondrocytes and increase the production of type II collagen and agrican. [0089] After washing the cultured wild ginseng roots, they were dried at 45°C and under reduced pressure conditions of an absolute pressure of 10 kPa or less until the moisture content was 8% by weight or less. The dried material was ground and sieved to obtain a ground material with an average particle size of approximately 300 μm. 500 g of purified water was added to 100 g of the ground material, and pre-hydration was performed by repeating the process three times: reducing pressure at 30°C and an absolute pressure of 20 kPa for 5 minutes, followed by standing at atmospheric pressure for 10 minutes. Subsequently, ethanol was added to adjust the final ethanol concentration to 55% by volume.
[0090] The obtained mixture was ultrasonically treated for a total of 60 minutes by repeating the process of irradiating with 40 kHz ultrasound at 50°C for 5 minutes followed by a 5-minute pause 6 times. The actual ultrasonographic irradiation time was 30 minutes. After ultrasonographic treatment, the mixture was stirred and extracted at 50°C and 250 rpm for 3 hours, and the primary extract and residue were separated by centrifugation at 5,000 rpm for 10 minutes.
[0091] (1-2) Secondary extraction and purification of adsorption resin
[0092] 800 g of 55 volume% aqueous ethanol was added to the residue, and a second extraction was performed at 50°C and 250 rpm for 2 hours. Subsequently, the second extract recovered by centrifugation was mixed with the first extract to obtain a crude wild ginseng extract. The crude wild ginseng extract was sequentially filtered through a 100 μm filter mesh and a 0.45 μm microfiltration membrane, and concentrated under reduced pressure at 40°C or below to remove ethanol so that the residual ethanol concentration was 1 volume% or less. Subsequently, purified water was added to adjust the solid content concentration to 5 weight% to obtain a wild ginseng concentrate.
[0093] A styrene-divinylbenzene-based macroporous adsorption resin was used as a nonionic porous adsorption resin. The said adsorption resin has a specific surface area of 450 m² 2 / g to 550 m 2 A resin with a g / g and an average pore diameter of 8 nm to 12 nm was used. The above adsorption resin was sequentially washed and equilibrated with 95 volume% ethanol and purified water.
[0094] After passing the above wild ginseng concentrate through an adsorption resin at a flow rate of 1 to 2 times the resin volume per hour, it was sequentially washed with 3 volumes of purified water and 2 volumes of 20 volume% aqueous ethanol. Subsequently, 3 volumes of 70 volume% aqueous ethanol were passed through at the same flow rate to elute the ginsenoside-containing fraction.
[0095] (1-3) Concentration and drying
[0096] The above ginsenoside-containing fraction was concentrated under reduced pressure at 40°C or below to remove ethanol. The concentrate was frozen at -40°C or below and then freeze-dried to obtain a wild ginseng extract containing ginsenosides.
[0097] (2) Preparation of mushroom extract
[0098] (2-1) Pretreatment, stepwise hydration, and sequential enzymatic treatment of mushroom raw materials
[0099] Sparassis crispa, Phellinus linteus, and truffles were each dried at 50°C until their moisture content was 10% by weight or less, then ground and sieved to obtain an average particle size of approximately 500 μm. 60 g of ground Sparassis crispa, 25 g of Phellinus linteus, and 15 g of truffles were mixed to prepare 100 g of ground mushroom raw material. 1,000 g of purified water was added to this, and the mixture was hydrated for 1 hour at 35°C and 200 rpm, followed by hydration for 1 hour at 55°C and the same stirring speed.
[0100] The temperature of the hydrated mushroom raw material mixture was adjusted to 50°C, and the pH was adjusted to 5.2 using an aqueous solution of citric acid or sodium citrate. Subsequently, 0.4 g of a hemicellulase preparation having an enzyme activity of 100,000 U / g and substantially no β-glucanase activity was added so that the hemicellulase activity was 400 U per 1 g of mushroom raw material, and a primary enzyme reaction was carried out at 50°C and 200 rpm for 90 minutes.
[0101] Subsequently, the pH of the reaction mixture was adjusted to 6.5, and 0.2 g of a neutral protease preparation with an enzyme activity of 100,000 U / g was added so that the neutral protease activity was 200 U per 1 g of mushroom raw material, and a second enzyme reaction was carried out at 50℃ and 200 rpm for 60 minutes.
[0102] (2-2) Pressurized hot water extraction and ultrafiltration
[0103] The secondary enzyme reaction mixture was heated at 90°C for 10 minutes to inactivate the enzyme. Subsequently, the reaction mixture was placed in a pressurized extraction vessel, and pressurized hot water extraction was performed for 90 minutes at 115°C and an absolute pressure of 0.18 MPa while stirring at 100 rpm to 200 rpm.
[0104] After extraction was completed, the pressure was gradually released, and the reaction mixture was cooled to below 50°C within 15 minutes. The cooled extract was centrifuged at 5,000 rpm for 10 minutes to recover the supernatant, and then sequentially filtered through a 100 μm filter mesh and a 0.45 μm microfiltration membrane.
[0105] The obtained filtrate was fed to a cross-flow ultrafiltration membrane made of polyethersulfone with a fractional molecular weight of 50 kDa and circulated under conditions of 30°C and a transmembrane pressure of 0.25 MPa. Ultrafiltration was performed until the volume of the membrane retained solution was reduced to one-quarter of the volume of the initially supplied filtrate. The membrane permeate was discharged, and the membrane retained solution concentrated with beta-glucan was recovered.
[0106] (2-3) Dialysis Filtration and Concentration
[0107] After adding a volume of purified water equal to the volume of the membrane holding solution at the end of concentration to the above membrane holding solution, the process of ultrafiltration was repeated three times until the membrane holding solution recovered to the volume before the addition of purified water, thereby performing dialysis filtration. Accordingly, the total amount of purified water used for dialysis filtration was made to be three times the volume of the membrane holding solution at the end of concentration.
[0108] The pH of the membrane-containing solution, after dialysis filtration was completed, was adjusted to 6.0 to 7.0, and after pre-filtering with a 1.0 μm filter membrane, it was filtered with a 0.45 μm filter membrane. Subsequently, the filtrate was concentrated under reduced pressure at 40°C or lower to adjust the solid content to 12% by weight, thereby obtaining a mushroom extract containing beta-glucan.
[0109] (3) Preparation of the composition
[0110] The above wild ginseng extract and mushroom extract were mixed in a weight ratio of 1:5 based on their respective dry solid content. Specifically, while maintaining the mushroom extract corresponding to 50 g of dry solid content at 35°C, 10 g of freeze-dried wild ginseng extract was added in five separate additions of 2 g each at 5-minute intervals, and the mixture was stirred at 700 rpm during the addition. After the addition of the wild ginseng extract was completed, the mixture was further mixed for 60 minutes at the same temperature and stirring speed. The obtained mixture was subjected to high-pressure homogenization three times at a pressure of 50 MPa, with cooling between each homogenization step to ensure the temperature of the mixture did not exceed 45°C. Subsequently, the mixture was aged at 8°C for 12 hours, frozen at -40°C or below, and then freeze-dried to prepare a composition for improving height growth.

[0137] Experimental Example 1. Content and recovery rate of ginsenosides and beta-glucan
[0138] (1) Experimental method
[0139] The ginsenoside and beta-glucan content of the final compositions prepared in Examples 1 and 2 and Comparative Examples 1 to 14 was evaluated, and the recovery rates of ginsenoside and beta-glucan were calculated in each preparation process of wild ginseng extract and mushroom extract, respectively. All analyses were repeated three times.
[0140] To measure the total ginsenoside content, 25 mL of 70 vol% methanol was added to 0.5 g of each final composition, and ultrasonic extraction was performed at 40 kHz for 30 minutes. The extract was centrifuged at 5,000 rpm for 10 minutes, and the supernatant was filtered through a 0.45 μm membrane filter and analyzed by high-performance liquid chromatography (HPLC). Using a C18 column and a purified water-acetonitrile gradient elution condition, the content of ginsenosides Rb1, Rb2, Rc, Rd, Rg1, and Re detected at 203 nm was summed to express the total ginsenoside content (mg / g) per 1 g of dry composition. The ginsenoside recovery rate was calculated by determining the total amount from the above ginsenoside content and total dry weight of the cultured wild ginseng root raw material and the wild ginseng extract, and then calculating the percentage of the total amount in the wild ginseng extract relative to the total amount in the raw material.
[0141] The total beta-glucan content was calculated by subtracting the alpha-glucan (α-glucan) content from the total glucan content measured by the enzymatic method. Glucose generated by acid hydrolysis and enzymatic reaction was quantified by a glucose oxidase / peroxidase (GOPOD) reaction, and the results were expressed as weight % based on the dry composition. The beta-glucan recovery rate was calculated by determining the total amount from the beta-glucan content and total dry weight of the mushroom raw material and mushroom extract, respectively, and then calculating the percentage of the total amount in the mushroom extract relative to the total amount in the raw material.
[0142] (2) Experimental results
[0143] [Table 2]
[0144]
[0145] Examples 1 and 2 showed a balanced content and recovery rate of ginsenoside and beta-glucan compared to the comparative examples, whereas each comparative example showed a tendency for the content or recovery rate of ginsenoside or beta-glucan to decrease.
[0147] Experimental Example 2. Effect of promoting proliferation of growth plate chondrocytes and cartilage matrix formation
[0148] (1) Experimental method
[0149] The effects of the compositions prepared in Examples 1 and 2 and Comparative Examples 1 to 14 on growth plate chondrocyte proliferation and cartilage matrix formation were evaluated. Chondrocytes isolated from the tibial growth plates of growing rats were placed in a 96-well plate at a rate of 1 × 10⁶ per well. 4 The samples were divided into groups and cultured for 24 hours at 37°C and 5% CO2. Subsequently, each composition was added to the culture medium at a concentration of 100 μg / mL and cultured for the treatment time according to each evaluation item, while a group not treated with the composition was set as the control group.
[0150] Cell viability was evaluated by CCK-8 analysis after treating with each composition at concentrations of 25 μg / mL, 50 μg / mL, 100 μg / mL, and 200 μg / mL for 24 hours. As a result, it was confirmed that no cytotoxicity was observed at 100 μg / mL, and Table 3 presents the cell viability of the 100 μg / mL treatment group. Cell proliferation rate was evaluated by quantifying double-stranded DNA content using fluorescence after treating with each composition at a concentration of 100 μg / mL for 48 hours, and the results were expressed as relative values with the control group set to 100%.
[0151] To confirm the cartilage matrix generation effect, each composition was treated at a concentration of 100 μg / mL for 72 hours, and the cell layer was recovered. The recovered cell layer was treated with a lysis buffer to extract cellular and extracellular matrix components, and the amounts of type II collagen and agrican contained in the extract were quantified using an enzyme-linked immunosorbent assay (ELISA) with respective antibodies.
[0152] In a separate identical treatment group, double-stranded DNA content was measured using the same fluorescence method to calculate cell mass, and the production of type II collagen and agrican was corrected by dividing each by the DNA content. The corrected values were expressed as relative values with the control group value set to 100%. All experiments were repeated independently three times.
[0153] (2) Experimental results
[0154] [Table 3]
[0155]
[0156] Examples 1 and 2 increased the proliferation of growth plate chondrocytes and the production of type II collagen and agrican while maintaining cell viability at a level similar to the control group, whereas Comparative Examples 1 to 14 showed lower effects of cell proliferation and cartilage matrix production than Examples 1 and 2.
[0158] Experimental Example 3. Effect of promoting axial growth in iliac bone tissue culture
[0159] (1) Experimental method
[0160] To evaluate the effect of the compositions prepared in Examples 1 and 2 and Comparative Examples 1 to 14 on promoting longitudinal growth of long bones, metatarsals were isolated from 3-day-old Sprague-Dawley rats and tissue culture of the long bones was performed. The isolated metatarsals were immersed in a culture medium containing α-MEM as the base medium, antibiotics, bovine serum albumin (BSA), and ascorbic acid, and cultured at 37°C and 5% carbon dioxide.
[0161] Each composition was added to the culture medium at a concentration of 100 μg / mL, and a group not treated with the composition was designated as the control group. Six metatarsals were used per group, and the culture medium was exchanged every two days during a six-day incubation period. Metatarsals were photographed using a stereomicroscope on the start and end dates of incubation, and the length from the proximal end to the distal end was measured using image analysis software. The longitudinal length growth rate was calculated as the percentage increase in length after incubation relative to the length before incubation.
[0162] After the culture was completed, the metatarsals were fixed with 4% paraformaldehyde and embedded in paraffin, and 5 μm thick longitudinal tissue sections were prepared. The tissue sections were stained with hematoxylin-eosin (H&E), and the total thickness of the growth plate and the thickness of the proliferation zone were measured. All measurements were performed on at least three sections per sample, and the average values were calculated.
[0163] (2) Experimental results
[0164] [Table 4]
[0165]
[0166] As Examples 1 and 2 showed higher rates of increase in long bone length, total thickness of the growth plate, and thickness of the proliferation zone than the control group and Comparative Examples 1 to 14, it was confirmed that the composition of the present invention promotes the proliferation of growth plate cartilage cells and effectively increases the longitudinal growth of the long bone.
