Fixed strain of rabies virus VRC-RZ2 (rabies virus), genus lyssavirus, intended for the manufacturing of drugs for specific prevention and diagnostics

Abstract

The invention relates to the field of virology and is a strain of the rabies virus of animals, isolated from an organ-tissue (puppy brain) rabies isolate and adapted for reproduction in a transplantable BSR cell culture by sequential and alternating passages on suckling mice and a BSR cell culture. The isolated VRC-RZ2 strain is propagated on the continuous BSR cell line, grown using a benchtop BelloStage™-3000 bioreactor system in BelloCellA flasks containing BioNOC II® macrocarriers, with biological activity up to 8.50 lg TCID50 / cm3. Tests for bacterial and fungal contamination have demonstrated the strain's sterility against bacterial and fungal microorganisms. The anti-rabies strain VRC-RZ2 of the animal rabies virus is deposited in the Microorganism Collection of the Research Institute for Biological Safety Problems under registration number 797 / 19 and is stored in dried ampoules at minus 80°C. The viability of the strain is maintained at a stable state by refreshing it on the continuous BSR cell line once every five years.
