Abstract
The invention belongs to the field of chromatographic separation of (2R,3R)-dihydroquercetin, production of biologically active supplements for medical and pharmaceutical purposes, as well as for the food and cosmetic industry. This is a new improved method for high-efficiency extraction of (2R,3R)-dihydroquercetin, which results in a particularly pure (2R,3R)-dihydroquercetin product, characterized by a white fluffy and water-soluble consistency, which is effective in further use of the product. The solid raw material containing dihydroquercetin is dissolved in an aqueous solution of ethyl alcohol at a temperature of 20 – 29 С until the proportion of dihydroquercetin in the solution reaches 50 - 250 g / l. After that, chromatographic separation is carried out at a pressure of 100 - 500 kg / cm2 in a column with dynamic axial pressure, where the column is filled with a reversible phase sorbent, when the sorbent is elute with a pre-degassed and modified ethyl alcohol-water solution. The collection of eluate fractions is carried out by refractometric and mass spectrometric analysis methods with subsequent crystallization of the product by freezing, filtration and lyophilization at a temperature from -25 to -35 С and vacuum pressure from 0,2 to 0,4 mbar. The product obtained in this way contains not less than 99.4% (2R,3R)-dihydroquercetin and is characterized by a light-colored (white) fluffy water soluble consistency.
