Abstract
The invention discloses a method for establishing a tissue culture regeneration system for wild Gleditsia microphylla, belonging to the field of plant tissue culture technology. The method comprises the following steps: Inoculating explants derived from wild Gleditsia microphylla seeds onto a callus induction medium to induce the formation of callus. Cutting the callus from its base and inserting the cut end into a root induction medium to induce rooting. In this invention, the hypocotyls and cotyledons of sterile seedlings of wild Gleditsia microphylla are used as explants. The effects of different combinations of plant growth regulators and browning inhibitors on regeneration efficiency are investigated. The invention successfully establishes a high-efficiency regeneration system for wild Gleditsia microphylla, providing a theoretical basis and technical support for the cultivation of high-quality seedlings and genetic improvement via plant biotechnology.
